Migration, transformation and nitrate source in the Lihu Underground River based on dual stable isotopes of δ15N-NO3- and δ18O-NO3.

Nitrate (NO3-) pollution is a common phenomenon in karst underground rivers, which are important water sources in karst landscapes. For drinking water safety and environmental protection, it is crucial to accurately identify NO3- sources and their migration and transformation processes in the Lihu Underground River. In this study, water samples of the Lihu Underground River in Guangxi were collected in May 2014, October 2014, January 2015, and July 2015, and water chemical and dual isotopic (δ15N-NO3- and δ18O-NO3-) approaches were used to evaluate the NO3- characteristics and sources in the L

Origins of groundwater nitrate in a typical alluvial-pluvial plain of North China plain: New insights from groundwater age-dating and isotopic fingerprinting.

Identifying nitrate sources and their temporal evolution in different land use is important for the sustainable management of groundwater resources. In this study, groundwater dating (3H-3He and time series of 3H) was combined with chemical and stable isotope analyses to resolve the evolution of nitrate sources and the driving mechanism of nitrate contamination. Approximately 75% of the groundwater samples (collected in 2014 and 2018) had nitrate concentrations exceeding World Health Organization's guideline for drinking water (50 mg/L), and 44% exceeded the groundwater quality standard of Chi

Mercury and methylmercury in carapace of the marine turtle Caretta caretta, in northeastern Brazil and its potential for environmental monitoring.

The present work tests the use of carapace fragments of the marine turtle Caretta caretta as a tool for environmental biomonitoring of mercury (Hg) and to evaluate the influence of biological and ecological factors in Hg concentrations. Samples of carapace fragments were obtained during the nesting season of 2012 and 2016 and were analyzed for their total-Hg and methyl-Hg concentrations and the isotopic composition of carbon and nitrogen (δ15N and δ13C). Seventy-six females were sampled, with an average size of 87.1 to 107 cm of curved carapace length (CCL). The results showed a wide variation

Heterotrophic inorganic carbon fixation varies with temperature and resource availability in marine and soil bacterial isolates.

Inorganic carbon use in cellular metabolism is ubiquitous in all living organisms. The assimilation of inorganic carbon into biomass via heterotrophic inorganic carbon fixation (HICF) is estimated to contribute significantly to global carbon fixation, yet the controls on this process in strictly heterotrophic microorganisms remain poorly understood. To identify potential drivers of HICF, we cultivated well-characterized heterotrophic Gammaproteobacteria (Pseudomonas putida and Pseudoalteromonas carrageenovora) at two temperatures (optimal vs colder) and resource treatments (high vs low organic

Retention of deposited ammonium and nitrate and its impact on the global forest carbon sink.

The impacts of enhanced nitrogen (N) deposition on the global forest carbon (C) sink and other ecosystem services may depend on whether N is deposited in reduced (mainly as ammonium) or oxidized forms (mainly as nitrate) and the subsequent fate of each. However, the fates of the two key reactive N forms and their contributions to forest C sinks are unclear. Here, we analyze results from 13 ecosystem-scale paired 15N-labelling experiments in temperate, subtropical, and tropical forests. Results show that total ecosystem N retention is similar for ammonium and nitrate, but plants take up more la

Iron and sulfate reduction vs. methanogenesis: Contrasting terminal anaerobic organic matter degradation pathways regulate groundwater iodine enrichment.

Groundwater iodine enrichment is fundamentally controlled by contrasting natural organic matter degradation pathways, yet their molecular-scale regulatory mechanisms remain unresolved. This study integrates stable carbon isotopes, excitation-emission matrix (EEM) fluorescence spectroscopy, and Fourier-transform ion cyclotron resonance mass spectrometry (FT-ICR MS) to decipher pathway-specific drivers in the Yangtze River alluvial aquifer. Integrated geochemical evidence reveals two distinct regimes: iron and sulfate reduction-dominated systems exhibit strong δ¹³C-DIC depletion (down to -16.7‰)

Aqueous-Phase Delivery of SLIC-SABRE Hyperpolarized [15N3]Metronidazole.

Metronidazole is an antiprotozoal antibiotic drug holding promise for tumor microenvironment analysis in hyperpolarized MRI due to its hypoxia-sensitive metabolism. Previous studies have explored the 15N hyperpolarization of metronidazole using dissolution dynamic nuclear polarization (d-DNP) and signal amplification by reversible exchange (SABRE). Here, we demonstrate that by using spin-lock-induced crossing (SLIC), 15N polarization levels up to 10-11% are achieved for 30 mM [15N3]metronidazole, improving polarization levels by a factor of 1.4 ± 0.1 compared to polarization levels achieved in

1H, 15N and 13C backbone resonance assignments of flap endonuclease from Plasmodium falciparum.

Flap endonuclease (FEN) enzymes are a group of metallonucleases that have essential roles in DNA repair and the maintenance of genomic resilience. They catalyse hydrolytic cleavage of a phosphodiester bond to remove 5'-flaps present on double-stranded DNA molecules formed during DNA replication. FEN locates a target scissile bond through the structural recognition of a pronounced bend in the double-stranded DNA substrate along with the presence of both a 5'-flap and a 1-nucleotide 3'-flap. FEN enzymes share a common structural architecture and are functionally conserved across all living organ

¹ ³C labeling unveils soil and microbial changes in continuously cropped Pogostemon cablin.

Continuous cropping (CC) disrupts agriculture by causing soil degradation, yield decline, and microbial imbalances due to nutrient loss and pathogen buildup. However, the mechanisms underlying these impacts on Pogostemon cablin remain unclear. A greenhouse pot experiment was conducted during the rapid growth stage when CC-induced stress is most pronounced to address this gap. The ¹ ³C stable isotope labeling was employed to trace microbial shifts and decomposition of organic matter within the plant-soil-microbe interface. The results indicated that CC significantly alleviated plant growth and

Elimination of 15N-thymidine after oral administration in human infants.

BACKGROUND: We have developed a new clinical research approach for the quantification of cellular proliferation in human infants to address unanswered questions about tissue renewal and regeneration. The approach consists of oral 15N-thymidine administration to label cells in S-phase, followed by Multi-isotope Imaging Mass Spectrometry for detection of the incorporated label in cell nuclei. To establish the approach, we performed an observational study to examine uptake and elimination of 15N-thymidine. We compared at-home label administration with in-hospital administration in infants with te

Body size is more important than diet in determining stable-isotope estimates of trophic position in crocodilians.

The trophic position of a top predator, synonymous with food-chain length, is one of the most fundamental attributes of ecosystems. Stable isotope ratios of nitrogen (δ15N) have been used to estimate trophic position of organisms due to the predictable enrichment of 15N in consumer tissues relative to their diet. Previous studies in crocodilians have found upward ontogenetic shifts in their 'trophic position'. However, such increases are not expected from what is known about crocodilian diets because ontogenetic shifts in diet relate to taxonomic categories of prey rather than shifts to prey f

Diets, stress, and disease in the Etruscan society: Isotope analysis and infantile skeletal palaeopathology from Pontecagnano (Campania, southern Italy, 730-580 BCE).

Susceptibility to morbidity and mortality is increased in early life, yet proactive measures, such as breastfeeding and weaning practices, can be taken through specific investments from parents and wider society. The extent to which such biosocialcultural investment was achieved within 1st millennium BCE Etruscan society, of whom little written sources are available, is unkown. This research investigates life histories in non-adults and adults from Pontecagnano (southern Italy, 730-580 BCE) in order to track cross-sectional and longitudinal breastfeeding and weaning patterns and to characteriz

Compound-specific hydrogen isotope analysis of heteroatom-bearing compounds via gas chromatography-chromium-based high-temperature conversion (Cr/HTC)-isotope ratio mass spectrometry.

The traditional high-temperature conversion (HTC) approach toward compound-specific stable isotope analysis (CSIA) of hydrogen for heteroatom-bearing (i.e., N, Cl, S) compounds has been afflicted by fractionation bias due to formation of byproducts HCN, HCl, and H2S. This study presents a chromium-based high-temperature conversion (Cr/HTC) approach for organic compounds containing nitrogen, chlorine, and sulfur. Following peak separation along a gas chromatographic (GC) column, the use of thermally stable ceramic Cr/HTC reactors at 1100-1500 °C and chemical sequestration of N, Cl, and S by chr

Soil fertilization leads to a decline in between-samples variability of microbial community δ13C profiles in a grassland fertilization experiment.

Gas chromatography combustion isotope ratio mass spectrometry (GC-C-IRMS) was used to measure the (13)C/(12)C ratios of PLFAs at natural abundance levels from a temperate grassland nitrogen (N) and phosphorus (P) factorial fertilization experiment in northern Greece. In each plot two rhizosphere samples were derived centred around individual Agrostis capillaris and Prunella vulgaris plants. It was hypothesized that the isotopic signal of microbes that preferentially feed on recalcitrant litter such as fungi would be modified by fertilization more strongly than that of opportunistic microbes us

Systemic Inflammation, Tumor Isotopic Signatures, and Prognosis in Oral Squamous Cell Carcinoma: Exploratory Integration of Blood- and Tissue-Derived Biomarkers-An Exploratory Retrospective Secondary Analysis.

Background/Objectives: Oral squamous cell carcinoma (OSCC) remains clinically heterogeneous, and prognosis is not always fully explained by conventional clinicopathological parameters. Systemic inflammation and tumor metabolic alterations may provide complementary information on tumor biology. This study aimed to assess associations between preoperative inflammatory markers, isotope ratio mass spectrometry (IRMS)-derived tumor signatures, clinicopathological features, and survival outcomes in OSCC. Methods: This exploratory retrospective secondary analysis included 50 consecutive patients with

Mechanistic deductions from isotope effects in multireactant enzyme mechanisms.

In the enzymatic mechanism with two or more substrates, comparison of the isotope effects on the maximum velocity and on the apparent V/K values when each substrate concentration is varied allows one to deduce the kinetic mechanism and obtain quantitative information on the relative rates at which substrates dissociate from the enzyme, as opposed to undergoing reaction to give products. Theory is also presented for using the effects of other reactants on the apparent isotope effects determined by the equilibrium perturbation method to determine the same information. With liver alochol dehydrog

Accurate quantification of protein concentrations and state populations by 2D 1H-N Q-BEST NMR.

We introduce 2D Q-BEST NMR as a quantitative approach for determining protein concentrations and relative state populations from a set of three 1H-15N correlation spectra. The BEST methodology ensures short overall experimental times and highest sensitivity. The method was validated using ubiquitin as a model system, where a precision of approximately 15-20% was achieved at the level of individual cross-peaks. This precision can be further improved by averaging corrected intensities of well-resolved resonances corresponding to the same protein state. The approach was subsequently applied to th

Diet composition and trophic ecology of two Antarctic storm-petrel species.

Availability and quality of food shape the distribution and movements of animal populations. In sympatric species, sharing limited resources, coexistence is typically achieved through niche segregation. However, information on dietary niches is lacking particularly for small and elusive seabirds, which often forage in remote oceanic areas. In this study, we aimed to characterize the trophic ecology of two highly pelagic seabird species, Wilson's storm-petrel Oceanites oceanicus and Black-bellied storm-petrel Fregetta tropica, breeding sympatrically on King George Island, maritime Antarctica, u

[Utilization of N15-labelled urea in the laying hen. 5. N15-labelling of the content and tissue of separate parts of the gastrointestinal tract].

In the series of experiments with labelled urea three colostomized laying hybrids were butchered after a six-day application of 1% urea with 96.06 atom-% 15N excess (15N') in the ration and another 2 days with a supplement of 1% unlabelled urea. Out of the individual samples from crop, gizzard, small intestine, caecum and rectum, the content of the small intestine and the caecum showed the highest labelling with greater than 1 atom-% 15N'. The TCA soluble fraction of the content of the gizzard was more highly and that of the intestines less labelled than the total nitrogen. The tissue of the g
