Activation of the Wnt/β-catenin pathway and tissue inhibitor of metalloprotease 1 during tertiary dentinogenesis.

Tertiary dentin is deposited inside teeth after various stimuli and serves as a major defensive wall to preserve pulp cells. However, the molecular mechanisms of the activation of quiescent odontoblasts, immature pulp cells and tertiary dentin formation are still unclear. Therefore, we performed a comprehensive gene expression analysis of pulp cells after cavity preparation of 9-week-old rat molars to clarify the critical molecules in tertiary dentinogenesis. As a result, mRNA expression of various molecules was up- or down-regulated. Notably, several members of the matrix metalloprotease fami…

Structural analysis of reactionary dentin formed in response to polymicrobial invasion.

In response to microbial invasion of dentin odontoblasts secrete an altered calcified matrix termed reactionary dentin (Rd). 3D reconstruction of focused-ion-beam scanning electron microscopy (FIB-SEM) image slices revealed helical tubular structures in Rd that contrasted with regular cylindrical tubules characteristic of dentin from healthy teeth and affected so-called physiological dentin (Pd) lying exterior to Rd. This helical structure in Rd provided effective constriction of tubule lumen diameter that formed a barrier to bacterial advance towards the dental pulp. SEM of resin cast prepara…

Histologic assessment of human pulp response to capping with mineral trioxide aggregate and a novel endodontic cement.

INTRODUCTION: This study was conducted to compare human pulp response to mineral trioxide aggregate (MTA) and a novel endodontic cement (NEC) when used as pulp capping materials after a time period of 2 and 8 weeks. METHODS: Thirty-two premolar teeth that were scheduled for extraction for orthodontic reasons were exposed and capped with either MTA or NEC. Half of the specimens underwent extraction and histologic analysis after 2 weeks, and the remaining half were assessed after 8 weeks. Each slide was graded histologically according to the morphology of the dentinal bridge, thickness of the d

[The fine structure of the dental pulp].

Root pulp usually contains more collagen fibrils than coronal pulp. The fibrils consolidate to form fibrinous bundles only to a very slight extent. Involution of the odontoblasts occurs with advancing age. If such involution is absent, an irregular layer of hard substance is deposited on the canal wall. A homogeneous line of adhesive can be seen on the wall at the border with the root pulp. The tissue character of the pulp however is preserved. Perineurium is absent around the nerves in human pulp.

The application of tissue engineering to regeneration of pulp and dentin in endodontics.

Caries, pulpitis, and apical periodontitis increase health care costs and attendant loss of economic productivity. They ultimately result in … al regeneration. The pulp tissue contains stem/progenitor cells that potentially differentiate into odontoblasts in response to bone morphogenetic proteins (BMPs). There are two strategies to regenerate dentin. First, is in vivo therapy, where BMP proteins or BMP genes are directly applied to the exposed or amputated pulp. Second is ex vivo therapy and consist …

Enameloid formation in two tetraodontiform fish species with high and low fluoride contents in enameloid.

Forming teeth of parrotfish and pufferfish were viewed by transmission electron microscopy to correlate cytological features of the enameloid organ with the species' fluoride (F) content in mature enameloid. Secretory-stage inner dental epithelial cells (IDE) of parrotfish (high F) and pufferfish (low F) secreted procollagen granules into the enameloid collagen matrix. The odontoblasts of both species, less numerous than IDE cells, also contained procollagen granules at the enameloid matrix formation stage. After the full thickness of enameloid matrix collagen had been deposited, enameloid cry…

Response of human dental pulp capped with biodentine and mineral trioxide aggregate.

INTRODUCTION: Biodentine is a new bioactive cement that is similar to the widely used mineral trioxide aggregate (MTA). It has dentin-like mechanical properties, which may be considered a suitable material for clinical indications of dentin-pulp complex regeneration such as direct pulp capping. The purpose of the present study was to compare the response of the pulp-dentin complex in human teeth after direct capping with this new tricalcium silicate-based cement with that of MTA. METHODS: Pulps in 28 caries-free maxillary and mandibular permanent intact human molars scheduled for extraction f

Convergence in dental histology between the late Triassic semionotiform Sargodon tomicus (Neopterygii) and a late cretaceous (Turonian) pycnodontid (Neopterygii: Pycnodontiformes) species.

Microstructural scanning electron microscope investigation was performed on sectioned and surface-etched isolated, prehensile teeth of the L … fugally by the cells of the inner dental epithelium, probably with minor collagen contribution from odontoblasts. This is contrary to the more typical centripetal formation (beneath the basal lamina) of the acrodin layer implied for the studied pycnodontid teeth. The rare occurrences of vascular acrodin within Actinopterygii, and the demonstrated differences i …

Molecular and neurovascular mechanisms of thermal sensitivity in teeth.

The perception of dental pain and thermal stimuli is governed by the specialized structural and physiological organization of the dentin - p … late pulpal afferent fibers. Increasing experimental evidence supports an integrated model in which odontoblasts (ODs) function as mechanosensory transducers. In this concept, thermal stimuli generate dentinal fluid movement and mechanical stress that activate ion channels and enzymes in ODs, leading to the release of signaling molecules such as ATP, glutamate …

Spatial Transcriptomics-Inspired Targeted Hydrogel for Dental Pulp Therapy via Strengthen Macrophage Communication.

Tissue repair depends on how inflammatory cues drive regenerative cell-state transitions; a process poorly understood in human tissues. Human dental pulp provides an accessible, structurally well-defined injury model with robust clinical responses, facilitating in situ investigations of inflammatory signaling and endogenous regeneration. Here, we integrated high-resolution spatial transcriptomics with single-cell RNA sequencing (scRNA-seq) to chart its cellular ecosystem in health and caries. We resolved five major cell classes and four odontoblast progenitor subtypes, reconstructing their dif

The critical role of nuclear factor I-C in tooth development.

OBJECTIVES: Nuclear factor I-C (NFIC) plays a critical role in regulating epithelial-mesenchymal crosstalk during tooth development. However, it remains largely unknown about how NFIC functions in dentin and enamel formation. In the present review, we aim to summarize the most recent discoveries in the field and gain a better understanding of the roles NFIC performs during tooth formation. SUBJECTS AND METHODS: Nfic-/- mice exhibit human dentin dysplasia type I (DDI)-like phenotypes signified by enlarged pulp chambers, the presence of short-root anomaly, and failure of odontoblast differentia
