Rewiring oncogenic signalling to precision ablation of metastatic cancer.

Despite recent advances, long-term survival in metastatic carcinomas such as ovarian cancer remains limited by off-tumour toxicities of targeted therapies and low response rates to immunotherapy. Synthetic proteins have been engineered for selective recognition of oncogenic signalling states, but how they can be used to treat metastatic disease in vivo remains unclear. Addressing cancers driven by ErbB-family receptor tyrosine kinases such as EGFR and HER2, we used engineered proteins to restrict replication of a clinically approved viral backbone to kill cells with aberrant ErbB signalling. Identification of FXYD5 as an oncogene in pediatric papillary thyroid cancer.

Papillary thyroid cancer is one of the most common malignancies in pediatrics, with increasing prevalence. We analyzed the single-cell transcriptomic landscape from 11 pediatric patients. The compositions and functions in the tumor microenvironment showed remarkable differences. Endo_tip was primarily from tumor, receiving angiogenesis-associated signals from epithelia. Abundant immune cells infiltrated into tumor. SPP1+ M2-macrophage and tumor cells interacted with T cells via immunosuppressive ligand-receptor pairs. Trajectory inference identified genes in evolutionary branchpoints that may

Oncogene inactivation-induced senescence facilitates tumor relapse.

Oncogene-directed therapies can induce profound tumor regression in oncogene-addicted cancers, but their long-term benefit is often limited by resistance and relapse. Here we show that oncogene inactivation rapidly induces senescence and a pro-inflammatory senescence-associated secretory phenotype (SASP). In vivo, oncogene inactivation-induced senescence (OIIS) predisposes tumors to relapse, accompanied by polyploidy, chromosomal instability, acquisition of alternative oncogenic pathways including mouse double minute 2 homolog (Mdm2) upregulation, and tumor microenvironmental remodeling toward

Repression of Septin9 inhibits the oncogenic phenotype of bladder cancer.

PURPOSE: Septin 9 (SEPT9) is a member of the GTPase family and has a vital role in metastasis due to its role in cytoskeletal structure, actin remodeling process, and cell migration. Although SEPT9 has been implicated in the pathogenesis of several cancers, its role in bladder cancer remains unclear. MATERIALS AND METHODS: SEPT9 expression in bladder cancer tissues was analyzed using at both transcriptional and translational levels and also validated in clinical specimens using Gene Expression Omnibus (GEO) datasets. Additionally, the SEPT9 promoter methylation level was determined by bioinfo

Oncogenic infections: targets highly amenable to cancer prevention.

In the 60 years since Epstein-Barr virus (EBV) was identified as the first human oncogenic virus, the International Agency for Research on Cancer (IARC) has classified 13 infectious agents as carcinogenic. Globally, approximately 2.3 million cancer cases (12% of all cancers) are attributable to infection each year. Main causes are Helicobacter pylori (850 000), human papillomavirus (HPV; 730 000), hepatitis B virus (HBV; 380 000), EBV (240 000), and hepatitis C virus (160 000). Infection-attributable cancer burden varies greatly, exceeding 25% of all cancers in parts of Asia and in sub-Saharan

SOX4 as a Key Oncogene Driving Tumor Invasion in Retinoblastoma.

PURPOSE: This study aims to explore the role of SRY-related HMG box transcription factor 4 (SOX4) in promoting invasion in retinoblastoma (RB) and to elucidate the underlying oncogenic pathways. METHODS: SOX4 expression in human retina, intraocular RB, and extraocular RB samples was evaluated using bulk RNA sequencing (RNA-seq) and single-cell RNA sequencing (scRNA-seq), with further evaluation in RB cell lines (Y79, WERI-RB1). Small interfering RNA-mediated knockdown was performed in RB cell lines, followed by functional assays including CCK-8, EdU, colony formation, and transwell assays. An

COPZ1: an example of non-oncogene addiction in human tumors.

Non-oncogene addiction (NOA) indicates that tumor cell growth/survival requires the activity of genes/pathways not oncogenic per sé, and dispensable for normal cells. NOA genes provide a wide repertoire of novel therapeutic exploitable tumor vulnerabilities. A large body of evidence demonstrates the dependency of several tumors such as breast, prostate, ovary, thyroid, glioblastoma and LUAD, on the activity of COPZ1, a component of the heptameric COPI complex. Thus, COPZ1 is emerging as a potential novel therapeutic target for tumors of different origin. In different tumor models COPZ1 inhibit

BCAT1 is a NOTCH1 target and sustains the oncogenic function of NOTCH1.

High levels of branched-chain amino acid (BCAA) transaminase 1 (BCAT1) have been associated with tumor aggressiveness and drug resistance in several cancer types. Nevertheless, the mechanistic role of BCAT1 in T-cell acute lymphoblastic leukemia (T-ALL) remains uncertain. We provide evidence that Bcat1 was over-expressed following NOTCH1-induced transformation of leukemic progenitors and that NOTCH1 directly controlled BCAT1 expression by binding to a BCAT1 promoter. Further, using a NOTCH1 gain-of-function retroviral model of T-ALL, mouse cells genetically deficient for Bcat1 showed defects i

Oncogenic fusions converge on shared mechanisms in initiating astroblastoma.

Chromosomal rearrangements and gene fusions are the initial events in the development of many cancers. Astroblastoma (ABM), a brain cancer of unknown cellular origin and challenging to treat, is associated with diverse in-frame gene fusions, including MN1-BEND2 and MN1-CXXC5 (refs. 1,2). However, it remains unclear whether these gene fusions contribute to tumorigenesis. Here we show in mice that these two ABM-associated fusions converge on similar molecular activities and initiate malignancy specifically in ventral telencephalon neural progenitors. BEND2 and CXXC5 recognize similar DNA motifs,

Etiopathogenesis and Current Management of Oncogenic Pruritus - A Narrative Review.

Oncogenic pruritus or malignancy associated pruritus is an emerging cause of systemic pruritus in patients with malignancies. It is a debilitating condition and worsens the patient's quality of life, often interfering with their palliative care. Oncogenic pruritus can arise de-novo in such patients due to release of pruritogens and other inflammatory mediators from tumour cells into blood stream, or it may present as a component of paraneoplastic syndrome. A detailed history and clinical examination are necessary to rule out other causes of systemic pruritus. Objective evaluation of itch is re

Senataxin prevents replicative stress induced by the Myc oncogene.

Replicative stress (RS) is emerging as a promising therapeutic target in oncology, yet full exploitation of its potential requires a detailed understanding of the mechanisms and genes involved. Here, we investigated the RNA helicase Senataxin (SETX), an enzyme that resolves RNA-DNA hybrids and R-loops, to address its role in preventing RS by oncogenic Myc. Upon Myc activation, silencing of SETX led to selective engagement of the DNA damage response (DDR) and robust cytotoxicity. Pharmacological dissection of the upstream kinases regulating the DDR uncovered a protective role of the ATR pathway

USP36 plays an oncogenic role in colorectal cancer cells.

Cancer stem cells (CSCs) have emerged as crucial contributors to tumor relapse and chemoresistance, making them promising targets for treating cancers like colorectal cancer (CRC). However, the mechanisms governing CSC maintenance in CRC remain poorly characterized. In this study, we investigated the potential role of ubiquitin-specific protease 36 (USP36) in CRC. Our bioinformatic analysis revealed a significant upregulation of USP36 expression in CRC, and high USP36 levels were associated with poor prognosis in CRC patients. Furthermore, we observed an increase in USP36 expression in CRC cel

Non-oncogene dependencies: Novel opportunities for cancer therapy.

Targeting oncogene addictions have changed the history of subsets of malignancies and continues to represent an excellent therapeutic opportunity. Nonetheless, alternative strategies are required to treat malignancies driven by undruggable oncogenes or loss of tumor suppressor genes and to overcome drug resistance also occurring in cancers addicted to actionable drivers. The discovery of non-oncogene addiction (NOA) uncovered novel therapeutically exploitable "Achilles' heels". NOA refers to genes/pathways not oncogenic per sé but essential for the tumor cell growth/survival while dispensable …

Mechanisms of Resistance to Oncogenic KRAS Inhibition in Pancreatic Cancer.

KRAS inhibitors demonstrate clinical efficacy in pancreatic ductal adenocarcinoma (PDAC); however, resistance is common. Among patients with KRASG12C-mutant PDAC treated with adagrasib or sotorasib, mutations in PIK3CA and KRAS, and amplifications of KRASG12C, MYC, MET, EGFR, and CDK6 emerged at acquired resistance. In PDAC cell lines and organoid models treated with the KRASG12D inhibitor MRTX1133, epithelial-to-mesenchymal transition and PI3K-AKT-mTOR signaling associate with resistance to therapy. MRTX1133 treatment of the KrasLSL-G12D/+; Trp53LSL-R172H/+; p48-Cre (KPC) mouse model yielded

The JAK3Q988P mutation reveals oncogenic potential and resistance to ruxolitinib.

T-cell acute lymphoblastic leukemia (T-ALL) arises from the malignant transformation of T-cell progenitors at various differentiation stages. Given that patients who relapse have a dismal prognosis, there is an urgent need to identify the molecular alterations that are present in such patients and promote leukemogenesis to implement personalized therapies with higher efficacy and fewer adverse effects. In the present manuscript, we identified the JAK3Q988P mutation in a T-ALL patient who did not achieve a durable response after the conventional treatment and whose tumor cells at relapse presen

BCL6, a key oncogene, in the placenta, pre-eclampsia and endometriosis.

BACKGROUND: The key oncogene B-cell lymphoma 6 (BCL6) drives malignant progression by promoting proliferation, overriding DNA damage checkpoints and blocking cell terminal differentiation. However, its functions in the placenta and the endometrium remain to be defined. OBJECTIVE AND RATIONALE: Recent studies provide evidence that BCL6 may play various roles in the human placenta and the endometrium. Deregulated BCL6 might be related to the pathogenesis of pre-eclampsia (PE) as well as endometriosis. In this narrative review, we aimed to summarize the current knowledge regarding the pathophysi

Oncogenic Wnt3a is a promising sensitive biomarker for monitoring hepatocarcinogenesis.

BACKGROUND: The effective treatment for hepatocellular carcinoma (HCC) depends on early diagnosis. Previously, the abnormal expression of Wnt3a as the key signaling molecule in the Wnt/β-catenin pathway was found in HCC cells and could be released into the circulation. In this study, we used rat model of hepatocarcinogenesis to dynamically investigate the alteration of oncogenic Wnt3a and to explore its early monitor value for HCC. METHODS: Sprague-Dawley rats (SD) were fed with diet 2-fluorenylacetamide (2-FAA, 0.05%) for inducing hepatocarcinogenesis, and grouped based on liver morphologica

Distribution of RET proto-oncogene variants in children with appendicitis.

BACKGROUND: In addition to patient-related systemic factors directing the immune response, the pathomechanisms of appendicitis (AP) might also include insufficient drainage leading to inflammation caused by decreased peristalsis. Genetic predisposition accounts for 30%-50% of AP. M. Hirschsprung (HSCR), also characterized by disturbed peristalsis, is associated with variants in the RET proto-oncogene. We thus hypothesized that RET variants contribute to the etiology of AP. METHODS: DNA from paraffin-embedded appendices and clinical data of 264 children were analyzed for the RET c.135A>G varia

The oncogenic roles of NTRK fusions and methods of molecular diagnosis.

The NTRK gene family is composed of NTRK1, NTRK2, and NTRK3, which encode three tropomyosin-receptor kinases, belonging to a class of tyrosine kinase receptors. These proteins are known to play roles in cell proliferation, differentiation, apoptosis, and survival. Fusions involving the NTRK genes are long known as drivers in many tumors. Although they occur in less than 5% of all malignancies, their occurrence in a great diversity of tumors has been documented. Several rare tumors including infantile fibrosarcoma, secretory breast carcinoma, and mammary analogue secretory carcinoma are accompa

Antioxidants Associated With Oncogenic Human Papillomavirus Infection in Women.

BACKGROUND: Human papillomavirus (HPV) infection is a major cause of cervical cancer. Studies showed the onset of HPV carcinogenesis may be induced by oxidative stress affecting the host immune system. The association between antioxidants and oncogenic HPV remains unclear. In this study, we aim to identify antioxidants associated with vaginal HPV infection in women. METHODS: The associations between the 15 antioxidants and vaginal HPV infection status (no, low-risk [LR], and high-risk [HR] HPV) were evaluated using 11 070 women who participated in the 2003-2016 National Health and Nutrition E
