Thalassemia is one of the most common hereditary disorders in the Mediterranean region and studies have shown that the prevalence of beta-thalassemia trait is high in the southern part of Turkey. Gaziantep is a city located near this region and, therefore, the authors investigated the prevalence and hematological characteristics of the beta-thalassemia traits in primary school students in Gaziantep. Sixty primary schools were selected from a list of all primary schools using a systematic sampling method. Data were collected by a face-to-face questionnaire. Osmotic fragility testing (OFT) using…

A Chinese family with concurrent hereditary spherocytosis (HS) and haemoglobin (Hb) Q-Thailand is described. The Hb Q-Thailand mutation was found on the remaining alpha1 globin gene on a chromosome 16 containing the (-alpha 4.2) deletion. Active haemolysis in members of this family is segregated with the HS phenotype, and the Hb Q-Thailand in the heterozygous state does not seem to show any modulating effect on HS.

Aged HS erythrocytes with a defined primary defect in band 3 protein or ankyrin were incubated with amphiphiles (detergents) at sublytic concentrations (37 C, 60 min) or glucose-starved (37 C, 24 h). In line with previous studies, the release of AChE (exovesicles) from HS erythrocytes during glucose-starvation was significantly higher (11%) compared to that from control erythrocytes (1%). Control and HS cells responded, however, similarly to amphiphile-treatment (non-starving conditions). Amphiphiles induced similar types of shape alterations and a similar amount of AChE release (14-15%). Furt

The chemical reactions underlying the chemo-mechanical leak formation in human erythrocytes upon treatment with the carboxyl-modifying reagent 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) and subsequent minimal shearing, described in the preceding paper (Thelen, P. and Deuticke, B. (1988) Biochim. Biophys. Acta 944, 285-296), are here characterized in more detail. The capacity to form leaks under minimal shearing results from the transformation of certain membrane carboxyl groups into an activated state, i.e., the O-acylisourea derivative of the original COO- group. This activated state

1. The calcium content of human erythrocytes, after removal of the buffy coat and washing free from plasma with isotonic sodium chloride, has been determined by atomic absorption spectrophotometry. The mean value found for normal subjects was 0.634 mug/ml. of packed erythrocytes (0.0158 mug-atom/ml.). The corresponding values for magnesium and zinc were 79.7 and 20.1 mug/ml., respectively.2. The calcium is considered to be mostly and perhaps exclusively located in the erythrocyte membrane, since, after osmotic haemolysis, the same amount was found in the ghost cells as was present in the eryth

This study aimed to determine the erythrocytic lipid peroxidation and haemoglobin oxidation as contributory factors causing anaemia in cattle (Friesian x Egyptian native breed) infected with Babesia bigemina. Blood was collected from 32 cows infected with B. bigemina along with 18 healthy cows as controls for determination of erythrocytic malondialdehyde (MDA), blood methaemoglobin (MetHb), plasma free haemoglobin (PHb), corpuscular osmotic fragility (COF), red blood cell count (RBC), total haemoglobin (Hb) and packed cell volume (PCV). Percentage of parasitaemia varied from 14% to 36%. MDA, M…

Two experiments were conducted to reevaluate the dietary vitamin E requirement of fingerling channel catfish. Purified diets containing 1% cod liver oil, 4% stripped lard and adequate selenium supplemented with graded levels of DL-alpha-tocopheryl acetate were used in both experiments. The results indicate that the previously reported requirement value, which was based on growth and pathological changes, was underestimated. The minimum dietary vitamin E requirement in the form of DL-alpha-tocopheryl acetate was determined to be 50 mg/kg of diet based on 16-week liver microsomal ascorbic acid-s

The most noticeable biochemical abnormality of red cells of patients with autoimmune haemolytic anaemia with warm autoantibodies is the increased osmotic fragility of the red cells. This appeared to be most pronounced in patients with incomplete IgG warm autoantibodies and to be significantly correlated with the 51Cr T½ of the patients' red cells. It appeared to be a disturbance of the cells which slowly increased with time. It only occurs in vivo and the reaction of the cell with the autoantibodies is necessary. Possibly, it is caused by damage done to the sensitized red cells by the RES. It …

Ora-pro-nobis is a bioactive food plant widely distributed across Latin America and the Caribbean, with its biological activities largely at … hemolysis in a dose-dependent manner and mitigated protein oxidation. They also reduced erythrocyte osmotic fragility by lowering H50 (0.401-0.424%) and osmotic hemolysis (45.2-74.3%). Overall, FAE concentrated the highest load of bioactive compounds and emerged as the most promising fraction for nutraceutical development.

Hereditary spherocytosis (HS) is a congenital disorder of the red blood cell membrane and is characterized by hemolytic anemia, variable jaundice, and splenomegaly. In neonates, the diagnosis of HS can be difficult in the absence of family history. Herein, we describe clinical and molecular genetic findings in a Korean neonate with HS. A one-month-old girl presented with severe anemia and jaundice. Spherocytes were frequently observed on peripheral blood smear, but the erythrocyte osmotic fragility test result was normal. Targeted next-generation sequencing (NGS) revealed the patient was heter…

Hemolytic anemia caused by oxidative drugs is thought to result from the oxidation of intracellular and membrane sulfhydryl groups of the er … )C] NEM binding) necessary to cause this effect was lower than that necessary to produce changes in osmotic fragility or cation permeability, or to cause Heinz body formation. In the absence of glucose, these sulfhydryl groups became disulfides, but could be returned to the reduced state by restoring glycolysis or by adding dithiothreitol. Phenylhydrazine hemolytic …

Two male castrated Whippet littermates were presented at 1 year of age for pallor, tachycardia, systolic heart murmur, dark yellow to orange feces, intermittent lethargy, pigmenturia, and muscle shivering or cramping after exercise. Persistent macrocytic hypochromic anemia with marked reticulocytosis and metarubricytosis was found when CBC results were compared with reference values for Whippets. Increased serum creatine kinase activity and hyperkalemia also were sometimes present over the 4-year period of evaluation. Progressively increasing serum concentrations of N-terminal prohormone brain

We describe two families with the 'cryohydrocytosis' form of stomatocytosis. Both show a mild stomatocytic anaemia with Hb levels of 12-16 g … ew mutation of a dominant condition. Cold storage of the red cells led to a very marked increase in osmotic fragility and macrospherocytosis, explaining why a diagnosis of 'hereditary spherocytosis' can easily be reached in these pedigrees.

AIMS: STAT3 is a key modulator of activation and differentiation of macrophages. But it is still unknown if deficiency of STAT3 activates macrophages to destroy erythrocytes by phagocytosis. We generated STAT3 conditional knockout mice by crossing floxed STAT3 mice with Tie2 promoter-driven Cre-recombinase transgenic mice and clarified that Stat3 plays a critical role in the formation and activation of macrophages.
MAIN METHODS: Blood cell count, reticulocyte count, serum lactate dehydrogenase, erythropoietin, iron and ferritin concentration, and life span of the erythrocytes in Tie2 promoter
