Gene Transfer of Calcium-Binding Proteins into Adult Cardiac Myocytes.

Heart failure is the leading cause of combined morbidity and mortality in the USA with 50% of cases being diastolic heart failure. Diastolic … unction in disease. This chapter provides protocols for studying wild-type parvalbumin isoforms and parvalbumins with strategically designed EF-hand motifs in adult cardiac myocytes via acute adenoviral gene transfer. These protocols have been used extensively to optimize parvalbumin function as a potential therapeutic for failing heart muscle.

Presence and distribution of three calcium binding proteins in projection neurons of the adult rat cochlear nucleus.

The presence and distribution of three cytoplasmic calcium binding proteins, calbindin, calretinin, and parvalbumin, have been investigated in the projection neurons of the cochlear nucleus complex in adult rats by using immunohistochemistry in free-floating slices. Identification of the individual cell types was carried out on the basis of their intranuclear localization, morphological characteristics, and (in the cases of pyramidal and bushy neurons) by retrograde labeling with rhodamine-dextran. The most important findings were confirmed by using confocal microscopy. The data obtained in th

Osteopontin-immunoreactive primary sensory neurons in the rat spinal and trigeminal nervous systems.

1200 micrometer(2) and 9% of those in the range 600-1200 micrometer(2) showed the immunoreactivity (ir). DRG neurons <600 micrometer(2)800 micrometer(2) showed the ir and 21% of those in the range 400-800 micrometer(2) were immunoreactive for this protein. TG neurons <400 micrometer(2) were mostly devoid of OPN-ir (2%). Virtually all (99%) Mes5 primary sensory neurons exhibited the ir. Muscle spindles in the soleus and masseter muscles contained OPN-ir spiral axon terminals. In the hard palate and incisor periodontal ligament, unencapsulated corpuscular endings exhibited the ir. Hippocampal Perineuronal Nets Are Required for the Sustained Antidepressant Effect of Ketamine.

BACKGROUND: N-methyl-D-aspartate receptor antagonists, like ketamine, produce a rapid-acting and long-lasting antidepressant effect. Although the mechanism is not completely understood, ketamine is thought to preferentially target N-methyl-D-aspartate receptors on fast-spiking parvalbumin-containing interneurons. The function of parvalbumin-containing interneurons is dependent on perineuronal nets, a specialized form of extracellular matrix that surrounds these cells. METHODS: Chondroitinase was used to enzymatically degrade perineuronal nets surrounding parvalbumin-containing interneurons in

Calcium binding protein expression in the optic tectum of Alligator during development.

The onset and distribution of the calcium binding proteins, calretinin, calbindin, and parvalbumin, were examined in the optic tectum of Alligator mississipiensis embryos between Stages 18 and 26-28. The immunoreactivity of each calcium binding protein correlated well with the results from the Western blot experiments. In terms of onset and distribution, calretinin expressison was the most widespread of the three calcium binding proteins that were examined, and was also the earliest to be visualized. Calbindin expression occurred next, whereas parvalbumin expression was the most limited and ap

Spatial distributions of chemically identified intrinsic neurons in relation to patch and matrix compartments of rat neostriatum.

The spatial distributions and dendritic branching patterns of chemically identified subpopulations of striatal intrinsic neurons, defined by immunoreactivity for choline acetyltransferase (ChAT), neuropeptide Y or parvalbumin, were studied in relation to patch and matrix compartments of rat neostriatum. ChAT-immunoreactive cells and fibers showed an uneven pattern of distribution in the striatum. ChAT immunoreactivity was higher in the dorsolateral part and lower in the ventromedial part of the striatum. This regional gradient pattern is the inverse of the overall pattern of calbindin D28k imm

Calcium binding protein immunoreactivity in nucleus rotundus in a reptile, Caiman crocodilus.

Nucleus rotundus is a prominent nucleus in the dorsal thalamus of nonmammalian amniotes. In one group of reptiles, Caiman crocodilus, previous studies have identified three parts of this neuronal aggregate. The central portion, the rotundal core, which receives visual input from the midbrain and projects to a restricted portion of the telencephalon, contains relay cells only. Previous examinations using Nissl morphology indicated that neurons of the rotundal core were not a homogeneous population of cells. The present investigation utilized another methodology to examine cell populations withi

Study of the cross-reactivity of fish allergens based on a questionnaire and blood testing.

BACKGROUND: Parvalbumin and collagen have been identified as cross-reactive allergens for fish allergies. Although doctors realize that various fish elicit allergies, the targets of food allergen labeling laws were only mackerels and salmons in Japan and mackerels in South Korea. This study aimed to reveal the causative species for fish allergy via questionnaires and blood tests. METHODS: Questionnaire research was conducted in Japan via the internet concerning allergies for fish-allergic patients or their family members. Next, IgE reactivities and cross-reactivities of 26 fish species were a

Mechanisms of sharp wave initiation and ripple generation.

Replay of neuronal activity during hippocampal sharp wave-ripples (SWRs) is essential in memory formation. To understand the mechanisms underlying the initiation of irregularly occurring SWRs and the generation of periodic ripples, we selectively manipulated different components of the CA3 network in mouse hippocampal slices. We recorded EPSCs and IPSCs to examine the buildup of neuronal activity preceding SWRs and analyzed the distribution of time intervals between subsequent SWR events. Our results suggest that SWRs are initiated through a combined refractory and stochastic mechanism. SWRs i

Subdivisions of the auditory midbrain (n. mesencephalicus lateralis, pars dorsalis) in zebra finches using calcium-binding protein immunocytochemistry.

The midbrain nucleus mesencephalicus lateralis pars dorsalis (MLd) is thought to be the avian homologue of the central nucleus of the mammalian inferior colliculus. As such, it is a major relay in the ascending auditory pathway of all birds and in songbirds mediates the auditory feedback necessary for the learning and maintenance of song. To clarify the organization of MLd, we applied three calcium binding protein antibodies to tissue sections from the brains of adult male and female zebra finches. The staining patterns resulting from the application of parvalbumin, calbindin and calretinin an

Early activation of inhibitory neurons in the thalamic reticular nucleus during focal neocortical seizures.

The neurons of the thalamic reticular nucleus are among the main targets of corticothalamic projections and their vulnerability in pathological conditions is well established. The present experiments aimed at the description and immunocytochemical characterization of the neurons of the thalamic reticular nucleus activated in neocortical seizures. Focal seizures were induced by the topical application of isotonic, isohydric 4-aminopyridine solution to the sensorimotor neocortex of adult, anesthetized Wistar rats. The animals were perfused with fixative after 1 and 2 h of recorded seizure activi

Fish-allergic patients may be able to eat fish.

Reported fish allergy prevalence varies widely, with an estimated prevalence of 0.2% in the general population. Sensitization to fish can occur by ingestion, skin contact or inhalation. The manifestations can be IgE or non-IgE mediated. Several fish allergens have been identified, with parvalbumins being the major allergen in various species. Allergenicity varies among fish species and is affected by processing or preparation methods. Adverse reactions after eating fish are often claimed to be 'allergy' but could be a reaction to hidden food allergen, fish parasite, fish toxins or histamine in…

Auditory peripheral influences on calcium binding protein immunoreactivity in the cochlear nucleus during aging in the C57BL/6J mouse.

The C57BL/6J (C57) mouse was selected as a suitable model for early presbyacusis to determine if there were correlations between peripheral pathology (spiral ganglion loss, inner and outer hair cell loss) and calcium binding immunoreactivity in the cochlear nucleus during aging. The quantitative stereological method, the optical fractionator, was used for determining the total number of neurons and calcium binding immunopositive neurons (calbindin, parvalbumin and calretinin) during aging in the posteroventral- and dorsal cochlear nucleus (PVCN and DCN) in C57 mice. Comparing 30-month-old to 1

Selective dopamine D2 receptor deletion from Nkx6.2 expressing cells causes impaired cognitive, motivation and anxiety phenotypes in mice.

Abnormal dopamine neurotransmission is a common trait of some psychiatric diseases, like schizophrenia or bipolar disorder. Excessive dopaminergic tone in subcortical brain regions is associated with psychotic episodes, while reduced prefrontal dopaminergic activity is associated with impaired cognitive performance and reduced motivation, among other symptoms. Inhibitory interneurons expressing the calcium binding protein parvalbumin are particularly affected in both schizophrenia and bipolar disorder, as they set a fine-tuned physiological inhibitory/excitatory balance. Parvalbumin and somato

Age-Dependent Calcium-Binding Protein Expression in the Spiral Ganglion and Hearing Performance of C57BL/6J and 129/SvJ Mice.

BACKGROUND/AIMS: Calcium-binding proteins in neurons buffer intracellular free Ca2+ ions, which interact with proteins controlling enzymatic and ion channel activity. The heterogeneous distribution of calretinin, calbindin, and parvalbumin influences calcium homeostasis, and calcium-related neuronal processes play an important role in neuronal aging and degeneration. This study evaluated age-related changes in calretinin, calbindin, and parvalbumin immune reactivity in spiral ganglion cells. METHODS: A total of 16 C57BL/6J and 16 129/SvJ mice at different ages (2, 4, 7, and 12 months) were in

S100 proteins: Diagnostic and prognostic biomarkers in laboratory medicine.

S100 proteins are members of the superfamily of Ca2+-binding proteins characterized by the specific Ca2+-binding motif, the EF-hand. Proteins of this superfamily are of clinical use as important diagnostic and prognostic biomarkers in adult and pediatric Laboratory Medicine. For example, measurements of troponin are nowadays the 'gold standard' in the diagnosis of patients with acute coronary syndrome. Parvalbumins were identified as major fish allergens and blocking antibodies, induced by immunization with a hypoallergenic parvalbumin mutant, were shown to reduce allergic symptoms. Mutations …

Changes in calcium-binding protein expression in the auditory brainstem nuclei of the jaundiced Gunn rat.

Sensorineural hearing loss and auditory dysfunction are major sequelae of neonatal hyperbilirubinemia. The sites and cellular effects of bilirubin toxicity in the auditory brainstem pathway are not easily detected. Since altered intracellular calcium homeostasis may play a role in neuronal cell death, we hypothesized that the expression of calcium-binding proteins may be altered in the classic animal model of bilirubin neurotoxicity. The expression of the calcium-binding proteins, calbindin-D28k and parvalbumin, in the brainstem auditory pathway of homozygous recessive jaundiced (jj) Gunn rats

Altered distribution of inhibitory interneurons in polymicrogyria.

There is a high incidence of epilepsy in patients with polymicrogyria; however, the epileptogenic mechanisms are largely unknown. The density of parvalbumin-immunoreactive interneurons was evaluated in an experimental model of polymicrogyria, in order to assess the potential changes in the development of one population of inhibitory interneurons. Newborn hamsters received an intracerebral injection of ibotenate, and all injected animals showed abnormal cortical layers characterized by one or two microgyrus in the fronto-parietal cortex. A quantitative analysis revealed that the ratios of parva
