HMG-CoA Reductase inhibitors: an updated review of patents of novel compounds and formulations (2011-2015).

Statins are remarkably safe and efficient medications that are the mainstay of hypercholesterolemia treatment and have proven to be an invaluable tool to lower the risk of acute cardiovascular events. These compounds are inhibitors of 3-hydroxy-methylglutaryl CoA reductase (HMG-R), the rate-limiting enzyme in cholesterol biosynthesis. In spite of their success, they present undesirable side effects and are now loosing patent protection, which provides a great opportunity for the development of new and improved statins. Areas covered: This review summarizes the new patents for HMG-R inhibitors

Malaria chemotherapy: recent advances in drug development.

Malaria is a disease caused by parasitic protozoa of the genus Plasmodium. Despite significant advances in understanding the disease and the parasite biology, malaria still remains one of the leading causes of morbidity and mortality, particularly in malaria-endemic regions of the world. The main factor hampering malaria control is the high degree of resistance developed by Plasmodium species against several classes of drugs. Artemisinin-based Combination Therapy (ACT) is the most rapidly acting antimalarial treatment effective against multi-drug resistant strains, and is, at present, the only

Older and new formulations of cladribine. Pharmacology and clinical efficacy in hematological malignancies.

The purine nucleoside analog (PNA)--cladribine (2-CdA, 2-chlorodeoxyadenosine) is a cytotoxic agent of high efficacy in lymphoid and myeloid malignancies. This drug was approved by the FDA for treatment of hairy cell leukemia and in some European countries for treatment of refractory/relapsed chronic lymphocytic leukemia. 2-CdA is usually administered as continuous or intermittent intravenous infusion. Recently however, new formulations of this agent has been developed for subcutaneous and oral administration. In contrast to other PNA, 2-CdA is equally cytotoxic to both proliferating and quies

Emerging therapies for polycystic kidney disease.

Polycystic kidney diseases are the most common, monogenetic, inherited diseases in humans. Numerous human genes or gene loci are associated with a renal cystic phenotype. Currently, there are no treatments available to slow the development of renal cystic pathology; however, animal studies have identified several potential approaches to intervene in the disease process. The most advanced therapy is the use of vasopressin V(2) receptor antagonists, which reduce renal cAMP, a known promoter of renal cystic enlargement. Other therapies under study include the use of c-myc antisense oligonucleotid

Coarctation and other obstructive aortic arch anomalies: their relationship to the ductus arteriosus.

Forty-three specimens with an obstructive aortic arch anomaly, obtained from infants that had died under the age of one year, were studied to establish the relationship between the arch anomalies and the ductus arteriosus. All specimens were studied morphologically. Twenty of the 43 were also studied histologically. The morphological part of the study included measurements of all the segments of the aortic arch and establishment of the localisation of any associated coarctation. The specimens were divided according to the appearance of the isthmus. The isthmus was not present in 4, was atretic

A Comprehensive Review on Repurposing the Nanocarriers for the Treatment of Parkinson's Disease: An Updated Patent and Clinical Trials.

Parkinson's Disease (PD) is a progressive neurodegenerative disorder marked by the deterioration of dopamine-producing neurons, resulting in motor impairments like tremors and rigidity. While the precise cause remains elusive, genetic and environmental factors are implicated. Mitochondrial dysfunction, oxidative stress, and protein misfolding contribute to the disease's pathology. Current therapeutics primarily aim at symptom alleviation, employing dopamine replacement and deep brain stimulation. However, the quest for disease-modifying treatments persists. Ongoing clinical trials explore nove

A current review on P2X7 receptor antagonist patents in the treatment of neuroinflammatory disorders: a patent review on antagonists.

Chronic inflammation is defined by an activated microglial state linked to all neurological disorders, including Alzheimer's disease, Parkinson's disease, and amyotrophic lateral sclerosis (a motor neuron disease that affects the brain and spinal cord). P2X7 receptors (P2X7R) are ATP-activated ion-gated channels present on microglial surfaces. Prolonged ATP release under pathological settings results in sustained P2X7R activation, which leads to inflammasome development and cytokine release. P2X7R and its enabling roles have recently been linked to neurodegenerative diseases, making it a poten
