Early-life exposure to electronic cigarettes: cause for concern.

Electronic nicotine delivery systems, or ENDS, are devices that heat and aerosolise a solution of propylene glycol, glycerine, nicotine, and flavourings. They have only achieved widespread use in the past 5 years or so, and therefore evidence around their potential to effect health is scarce. Importantly, they are often viewed as safer than tobacco cigarettes, meaning that at-risk populations, including pregnant women, might be more inclined to use them. No human studies, however, have assessed the potential for maternal ENDS use to effect the health of a developing baby. Experimental research

Parental smoking and childhood cancer: results from the United Kingdom Childhood Cancer Study.

There are strong a priori reasons for considering parental smoking behaviour as a risk factor for childhood cancer but case - control studies have found relative risks of mostly only just above one. To investigate this further, self-reported smoking habits in parents of 3838 children with cancer and 7629 control children included in the United Kingdom Childhood Cancer Study (UKCCS) were analysed. Separate analyses were performed for four major groups (leukaemia, lymphoma, central nervous system tumours and other solid tumours) and more detailed diagnostic subgroups by logistic regression. Intergenerational Associations of Childhood War Exposure, Adverse Childhood Experiences, and Mental Health in Offspring of Lebanese Immigrants.

Lebanese Americans, the largest Arab ethnic group in the United States, remain underrepresented in mental health research despite collective exposure to conflict, displacement, and immigration. This study examined intergenerational associations of parental childhood war exposure (CWE) during the Lebanese Civil War (1975-1990) on the mental health of second-generation Lebanese American young adults (G1) and characterized G1 clinical symptoms and service use. Using community-partnered recruitment, 92 G1 (mean age 28.1 years, SD 4.5) participants reported on their own adverse childhood experience

Preconceptional paternal antiepileptic drugs use and risk of congenital anomalies in offspring: a nationwide cohort study.

Recent studies have shown that certain pharmacological agents used by fathers before conception may increase the risk of adverse neonatal outcomes in offspring. However, little is known about the effect of paternal use of antiepileptic drugs (AEDs) on congenital anomalies in children. Based on Danish national registers, we conducted a cohort study of 733, 282 singletons born from 1997 to 2008, with follow-up throughout 2013. The children whose fathers used AEDs during the 3 months before conception were categorized as the exposed. Logistic regression model was used to examine association betwe

Birthweight and paternal involvement predict early reproduction in British women: evidence from the National Child Development Study.

There is considerable interest in the mechanisms maintaining early reproduction in the most socioeconomically disadvantaged groups in developed countries. Previous research has suggested that differential exposure to early-life factors such as low birthweight and lack of paternal involvement during childhood may be relevant. Here, we used longitudinal data on the female cohort members from the UK National Child Development Study (n = 3,014-4,482 depending upon variables analyzed) to investigate predictors of early reproduction. Our main outcome measures were having a child by age 20, and stati

Parental Alcohol Use Disrupts Offspring Mitochondrial Activity, Promoting Susceptibility to Toxicant-Induced Liver Cancer.

The early onset and incidence of liver disease and hepatocellular carcinoma have doubled in the last two decades and are primarily attributed to an unhealthy lifestyle. However, emerging studies suggest that increases in these age-related pathologies may link to heritable alterations in the control of cellular bioenergetics induced by the parental environment. Because our preclinical studies examining the fetal offspring of alcohol-exposed males and females have consistently identified epigenetic alterations in mitochondrial activity, we hypothesized that chronic parental alcohol exposure prog

Environmental and Genetic Perturbations of the Sperm Epigenome.

The sperm cell is recognized nowadays as a key epigenetic vector, transmitting not only the paternal genome but also a complex epigenetic content-including DNA methylation, chromatin code and structure, and non-coding RNAs-to the zygote. This epigenome is highly sensitive to environmental factors, with paternal exposures such as dietary intake, lifestyle choices, stress, chemical hazards, pollution, or reprotoxic treatments (among others) influencing its programming. Genetic factors also modulate the sperm epigenetic cargo, supporting that a combination of complex multifactorial cues will defi…

Relationship of parental phthalate exposure with fetal growth and placental development at birth.

BACKGROUND: Prenatal exposure to phthalates is reported to influence fetal growth and may lead to lasting adverse effects on infants and their future development; yet, the results remain inconclusive. OBJECTIVE: This study utilized a birth cohort of 73 pregnant women-newborn pairs, including biological fathers (73 triads), to investigate the relationship between parental phthalate exposure during pregnancy and birth outcomes in newborns. METHODS: Demographic, behavioral, and clinical information, along with urine samples from both parents, were collected prior to delivery. Sixteen phthalate

Congenital fibrosarcoma and history of prenatal exposure to petroleum derivatives.

Congenital fibrosarcoma (CFS) is a rare fibrous tissue malignancy that usually presents in the first few years of life. It is unique among human sarcomas in that it has an excellent prognosis. We describe a temporal clustering of a number of cases of CFS and investigate the possible associated prenatal risk factors. The Pediatric Environmental History, a questionnaire developed in our clinic that is instrumental in determining environmental risk factors for tumor-related disease, was essential in documenting the presence or absence of risk factors considered as human carcinogens. We found a hi

Preconception exposures to potential germ-cell mutagens.

Radiation and other agents can cause germ-cell mutations in animal systems. No human germ-cell mutagen has been identified, but this does not mean that human germ-cells are not vulnerable to mutagenesis. There has been particular concern about the possible health effects on offspring following parental preconception exposure to ionizing radiation-both occupational and therapeutic. A strong association with preconception radiation exposure in the fathers of the cases was found in a case-control study of young people with leukaemia living near the Sellafield nuclear plant in the UK. Subsequent s

Parental smoking during pregnancy and cardiovascular structures and function in childhood: the Generation R Study.

BACKGROUND: Foetal smoke exposure might lead to foetal developmental adaptations that permanently affect the cardiovascular system. We assessed the associations of both maternal and paternal smoking during pregnancy with childhood cardiovascular structures and function. METHOD: In a prospective cohort study among 5565 children, we examined whether maternal and paternal smoking during pregnancy are associated with blood pressure, carotid-femoral pulse wave velocity and left cardiac structures and function in 6-year-old children. RESULTS: As compared with children from non-smoking mothers, chi

Parental exposure to triphenyltin inhibits growth and disrupts thyroid function in zebrafish larvae.

Triphenyltin (TPT) is widely used and commonly found in a water environment, so its effects on aquatic systems are of great concern. This study aimed to reveal the effects of chronic parental exposure of TPT on thyroid disruption and growth inhibition in zebrafish. Adult zebrafish (F0 generation) were exposed to environmentally relevant concentrations (1, 10, and 100 ng/L) of TPT for 60 days, and the larvae (F1 generation) were tested without TPT treatment. Results demonstrated that parental exposure to TPT disrupts thyroid function in zebrafish offspring: serum thyroxine (T4) significantly de

Cyclosporine A exposure during pregnancy in mice: effects on reproductive performance in mothers and offspring.

BACKGROUND: Pregnancies after organ transplantation and under immunosuppressive treatment are associated with slightly elevated risks for obstetric and post-natal complications but can usually be managed well. However, little is known about the effects of intrauterine exposure (IUE) to immunosuppressants in the growing and adult offspring. One major issue is the potentially negative effects of immunosuppressive medication on reproduction. This study investigates the effect of exposure during pregnancy to the most commonly used immunosuppressant in organ transplantation, cyclosporine A (CsA), o

Recurrent pregnancy loss: unexplained or misunderstood?

PURPOSE: Recurrent pregnancy loss (RPL) is a multifactorial condition with varying definitions across professional societies and is often misunderstood. This review summarizes recent insights into genetic, paternal, anatomic, metabolic, immunologic, and infectious contributors that may explain otherwise unexplained RPL. RECENT FINDINGS: In most RPL cases, a cause can be identified when standard evaluation is combined with genetic testing of products of conception (POC). When no clear etiology emerges, additional factors should be considered, including the couple's metabolic health, chronic en

Maternal-fetal HLA sharing and preeclampsia: variation in effects by seminal fluid exposure in a case-control study of nulliparous women in Iowa.

Whereas histocompatibility is critical for transplantation, HLA histoincompatibility is associated with successful pregnancy. Literature on HLA sharing and preeclampsia has been inconsistent; most studies focused on maternal-paternal rather than maternal-fetal sharing. This study examines whether maternal-fetal histocompatibility is associated with preeclampsia, and whether effects vary by semen exposure history. This case-control study of nulliparous women was designed to examine associations among HLA sharing, semen exposure, and preeclampsia. 258 preeclampsia cases and 182 normotensive cont

Intergenerational influences of paternal combat-related trauma on offspring behavioral and brain function.

Post-traumatic stress (PTS) is a debilitating mental health condition that is highly prevalent in Veteran populations owing to their increased exposure to combat-related trauma. PTS is associated with numerous comorbid conditions including major depressive disorder, anxiety, and chronic pain. Although the causal mechanisms are currently unknown, paternal trauma has been linked to an increased risk for pathology in their offspring. Therefore, using a preclinical model of combat trauma, we examined the relationship between paternal PTS and offspring socio-emotional functioning, pain perception,

West Nile virus infection induces susceptibility of in vitro outgrown murine blastocysts to specific lysis by paternally directed allo-immune and virus-immune cytotoxic T cells.

Day 3 post-coitum BALB/c and (BALB/c x CBA/H)F1 blastocysts were isolated and hatched in replicate wells. Some were treated with interferon-gamma (IFN-gamma). Whilst others were infected with West Nile Virus (WNV) at 100 plaque-forming units per cell, for 18 h. Controls were mock-treated. Gamma-irradiated (2000 rads) CBA/H, (paternal) WNV-specific and allo(CBA/H)-specific cytotoxic T (Tc) cells were then added to replicates of infected, mock-infected or IFN-gamma-treated cultures for 20 h. [3H]Thymidine was then added for a further 8 h. [3H]Thymidine incorporation was inhibited by 40-50% in WN

Association of Maternal and Paternal Depression in the Postnatal Period With Offspring Depression at Age 18 Years.

IMPORTANCE: Paternal depression during the postnatal period has been associated with adverse child outcomes. Family environment has been reported as a pathway for risk transmission from fathers to children. The influence of paternal depression during the postnatal period on offspring depression remains to be clarified. OBJECTIVE: To investigate the association between paternal depression in the postnatal period and offspring depression and explore potential mediating and moderating factors that influence any association between paternal and offspring depression.
