Determining the effects of LLD and MCI on brain decline according to machine learning and a structural covariance network analysis.

BACKGROUND: Late-life depression (LLD) and mild cognitive impairment (MCI) are risk factors for Alzheimer disease (AD). However, the interactive effect between LLD and MCI in the progression to AD remains unknown. The purpose of this research is to clarify whether this interaction exists and determined the characteristics of the structural change patterns in LLD and MCI. METHOD: To address this question, a total 225 participants (91 with intact cognitive function (IC), 34 with MCI, 35 with LLD-IC, 47 with LLD-MCI and 18 with AD) were recruited for the current study and their T1 scanning were

Identification of Recurrence-associated Gene Signatures and Machine Learning-based Prediction in IDH-Wildtype Histological Glioblastoma.

Glioblastoma (GBM) is a highly aggressive brain tumor with frequent recurrence, yet the molecular mechanisms driving recurrence remain poorly understood. Identifying recurrence-associated genes may improve prognosis and treatment strategies. We applied weighted gene co-expression network analysis (WGCNA) to transcriptomic data from IDH-wildtype histological GBM in the CGGA-693 (n = 190) and CGGA-325 (n = 111) cohorts to identify recurrence-associated genes. These genes were validated using RT-qPCR and single-cell RNA sequencing (scRNA-seq) datasets (GSE174554, GSE131928). Their associations wi

An Integrated Machine Learning and Genomic Framework for Precise Detection of Gastric Cancer.

This study presents a novel integrative approach for the analysis of high-dimensional gene expression data, leveraging the complementary strengths of unsupervised clustering and supervised classification. Using K-means clustering, the data set is stratified into three distinct clusters, revealing intrinsic biological patterns and relationships. The resulting cluster assignments are subsequently used as pseudolabels to train machine learning models, including support vector machines, random forest, and a stacking ensemble classifier. To validate and enhance the robustness of clustering, complem

OocystMeter, a machine-learning algorithm to count and measure Plasmodium oocysts, reveals clustering patterns in the Anopheles midgut.

We present OocystMeter, a machine learning-based software developed to automate the segmentation of malaria oocysts from images of mosquito midguts stained with mercurochrome. Existing bioimage analysis tools, including machine learning-based ones, often struggle with the unique staining patterns, complex midgut backgrounds, and variable morphology of oocysts, making the determination of oocyst size and numbers cumbersome. To overcome these challenges, we curated a high-quality dataset comprised of 11,178 Plasmodium falciparum oocysts in Anopheles gambiae midguts annotated by expert parasitolo

Integration of deep neural network modeling and LC-MS-based pseudo-targeted metabolomics to discriminate easily confused ginseng species.

Metabolomics covers a wide range of applications in life sciences, biomedicine, and phytology. Data acquisition (to achieve high coverage and efficiency) and analysis (to pursue good classification) are two key segments involved in metabolomics workflows. Various chemometric approaches utilizing either pattern recognition or machine learning have been employed to separate different groups. However, insufficient feature extraction, inappropriate feature selection, overfitting, or underfitting lead to an insufficient capacity to discriminate plants that are often easily confused. Using two ginse

Machine learning-based integration of tumor deposit molecular signatures improves prognostic stratification in colon adenocarcinoma.

BACKGROUND: Colon adenocarcinoma (COAD) remains a leading cause of cancer-related mortality worldwide. Although tumor deposits (TDs) are established prognostic indicators, their molecular characteristics and potential for improving risk stratification remain unexplored. METHODS: We performed an integrative analysis of transcriptomic and clinical data from TCGA and GEO databases to identify TD-associated molecular signatures. A hybrid ML framework combining random survival forest and stepwise Cox regression was developed to construct a risk stratification model. Model performance was validated

Temporal interference stimulation of peripheral nerves induces functionally diverse limb movements revealed by automated pose estimation and unsupervised behavioral analysis.

Peripheral nerve stimulation can help restore limb movement after paralysis and enable advanced rehabilitation technologies; however, current extraneural interfaces are typically limited by low fascicle selectivity and laborious functional evaluation. This study has developed an extraneural peripheral nerve interface with high fascicle selectivity, and an AI-facilitated video analysis pipeline for assessing limb movement during neuromodulation. This was achieved by deploying temporal interference stimulation (TIS) in a high-density nerve cuff electrode and by using machine learning algorithms

Logic programming to predict cell fate patterns and retrodict genotypes in organogenesis.

Caenorhabditis elegans vulval development is a paradigm system for understanding cell differentiation in the process of organogenesis. Through temporal and spatial controls, the fate pattern of six cells is determined by the competition of the LET-23 and the Notch signalling pathways. Modelling cell fate determination in vulval development using state-based models, coupled with formal analysis techniques, has been established as a powerful approach in predicting the outcome of combinations of mutations. However, computing the outcomes of complex and highly concurrent models can become prohibit

Professional chess expertise modulates whole brain functional connectivity pattern homogeneity and couplings.

Previous studies have revealed changed functional connectivity patterns between brain areas in chess players using resting-state functional magnetic resonance imaging (rs-fMRI). However, how to exactly characterize the voxel-wise whole brain functional connectivity pattern changes in chess players remains unclear. It could provide more convincing evidence for establishing the relationship between long-term chess practice and brain function changes. In this study, we employed newly developed whole brain functional connectivity pattern homogeneity (FcHo) method to identify the voxel-wise changes

Exhaled breath analysis with the use of an electronic nose to predict response to immune checkpoint inhibitors in patients with metastatic melanoma: melaNose trial.

INTRODUCTION: Immune checkpoint inhibitors (ICIs) have significantly improved the overall survival for patients with different solid tumors. However, there is an urgent need for predictive biomarkers to identify patients with metastatic melanoma who do not benefit from treatment with ICIs, to prevent unnecessary immune related adverse events (irAEs). Electronic noses (eNoses) showed promising results in the detection of cancer as well as the prediction of response outcome in patients with cancer. In this feasibility study, we aimed to investigate whether the breath pattern measured using eNose

Machine learning in diagnostic stewardship: A technical approach in improving diagnostic accuracy, optimizing antibiotic use in false positive CSF cultures.

Diagnostic stewardship emphasises ordering the right tests, at the right time for the patient and also promotes the judicious use of rapid and accurate molecular diagnostic tools to enable the initiation of proper antibiotic therapy, while avoiding excessive use of broad-spectrum antibiotics hence antimicrobial stewardship. Proper interpretation of the test is crucial to avoid over-diagnosis and excessive healthcare costs. This study aimed to access the CSF diagnostic accuracy by evaluating contamination rate through machine learning models, including ROC analysis and principal component analy
