OBJECTIVE: Medication-related osteonecrosis of the jaw (MRONJ) is a pathologic process resulting in progressive destruction of the jaws. There are no established guidelines for the medical management of MRONJ. Interest in pentoxifylline and tocopherol is growing because these agents have been shown to be effective in treating osteoradionecrosis of the jaw. BACKGROUND: Alcoholic hepatitis is the most florid presentation of alcohol-related liver disease. In its severe form, defined by a Maddrey's discriminant function (DF) ≥32, the 28-day mortality rate is approximately 35%. Kindling-syndrome was modeled in male Wistar rats using repeated corazole administration (25-30 mg/kg intraperitoneally). The effects of pentoxifylline (25 and 100 mg/kg intraperitoneally) and diazepam (0.05 and 1 mg/kg intraperitoneally) were studied in 3 weeks after kindling development under conditions of acute observation on the model of penicillin-induced foci in cerebral cortex. An anticonvulsant effect of combined treatment with diazepam and pentoxifylline in the doses, ineffective if administered separately (0.05 and 25 mg/kg intraperitoneally), was demonstrated.

BACKGROUND/AIM: The aim of this study was to determine whether pentoxifylline (Pf) has an effect on sunburn cell (SBC) formation in humans.
METHODS: A novel supravital human skin model was used. Normal skin samples were placed on gauze in completed RPMI 1640 Medium and remained vital for 48 h. Three concentrations of Pf (7.5, 15.0 and 30.0 microg/ml) were tested. After 2 h, each skin sample was irradiated with 120 mJ/cm2 of UVB. Pentoxifylline and the two analogues HWA138 and HWA448, at concentrations exceeding 60 micrograms/ml, inhibited malaria antigen or lipopolysaccharide (LPS) induced TNF-alpha and IL-1 alpha secretion, but not IL-6 secretion, from human peripheral blood mononuclear cells in vitro. HWA448 had lower inhibitory activity in vitro than pentoxifylline and HWA138. A small enhancement of cytokine secretion was induced by pentoxifylline and the two analogues at low concentrations.
