Regioselective phosphorylation of polyhydroxyglycosides remains a long-standing challenge in carbohydrate chemistry. Herein, we report a mild and efficient Fe(acac)3-catalyzed strategy for the selective phosphorylation of free vicinal diols in glycosides. The protocol exhibits a broad substrate scope and excellent functional group tolerance, delivering site-selectively phosphorylated glycosides in moderate to good yields. This work offers a streamlined catalytic approach to phosphorylated carbohydrate derivatives, providing a valuable tool for glycan functionalization in chemical biology and d…

The molecular chaperone Hsp90 is a major protein folding factor in the cytosol of eukaryotic cells. Its conformational cycle is regulated by various co-chaperones and post-translational modifications (PTMs) such as phosphorylation. Most of the phosphorylation sites are conserved between the 2 isoforms of human Hsp90, Hsp90α and Hsp90β. The analysis of the function of these sites has revealed general functional principles of Hsp90. To what extent isoform-specific phosphorylation regulates Hsp90 function is less well understood. Here, we explore the effect of the phosphorylation of a residue (th…

Gibberellin (GA) promotes plant growth primarily by triggering degradation of DELLA transcription regulators, yet how DELLA activity is fine-tuned dynamically by phosphorylation independently of proteolysis remains poorly understood. Here, we show that the CDK8 kinase module of the Mediator complex attenuates activity of the Arabidopsis DELLA protein REPRESSOR OF ga1-3 (RGA) by modulating coactivator recruitment. Using TurboID-based proximity labeling, biochemical and genetic analyses, we identify CDK8 as an in planta kinase that phosphorylates RGA at Ser170 within its disordered PolyS/T regio…

Gasdermin D (GSDMD), the most important executor of pyroptosis, is activated through caspase cleavage to release the N-terminal fragment (GD-NT), which oligomerizes and forms pores in the plasma membrane to induce pyroptosis. We previously reported that AMP-activated protein kinase (AMPK) phosphorylates GD-NT, thereby inhibiting its cytolytic activity. Here, we further investigated the role of phosphorylation in GD-NT-mediated pyroptosis and found that the specific AMPK agonist A-769662 protected tumor cells from GD-NT-mediated pyroptosis and that metformin significantly reduced interleukin 1β…

Understanding the mechanisms underlying chemoresistance is critical for improving cancer therapies. SIN1 plays a pivotal role in maintaining mTORC2 integrity and activation, which regulates key cellular processes. In this study, we demonstrate that elevated SIN1 expression in pancreatic ductal adenocarcinoma (PDAC) correlates with poor patient survival outcomes. Conversely, SIN1 deletion reduces tumor growth and enhances PDAC sensitivity to chemotherapy. We identify USP33 as a bona fide deubiquitanase of SIN1, essential for its stabilization in PDAC. This stabilization promotes chemoresistance

Cholangiocarcinoma (CCA) is a highly heterogeneous group of malignant tumors with different molecular etiologies and clinical manifestations. Post-translational modifications (PTM) such as ubiquitination and phosphorylation are widely involved in the progression of CCA. Our aim was to elucidate the effect of the deubiquitinating enzyme OTU domain-containing protein 3 (OTUD3) on the molecular pathogenesis of CAA. OTUD3 and ARID3A exhibit direct binding interactions and cytosolic substructural co-localization in CCA cells.OTUD3 specifically removes the ARID3A proteins K240 and K329 OTUD3 specifi…

Deoxynucleotides (dNTPs) in postmitotic tissues rely on deoxynucleoside salvage pathways in order to repair and replicate nuclear and mitoch … to display cytosolic thymidylate kinase 2 activity. Together, the results indicate that a two-step phosphorylation of thymidine to TDP occurs because the proximity of TK2 and CMPK2 in the mitochondria prevents TMP from diffusing from the two enzymes.

Aggregation of alpha-synuclein (αsyn) plays an integral role in Parkinson's disease (PD) and Dementia with Lewy bodies (DLB). 14-3-3θ is a highly expressed brain protein with chaperone-like activity that regulates αsyn folding. 14-3-3θ overexpression reduces αsyn aggregation, transmission between cells, and neuronal loss, while 14-3-3 inhibition promotes αsyn pathology. We previously observed increased 14-3-3θ phosphorylation at serine 232 in human PD and DLB brains. Here we examine 14-3-3θ phosphorylation's effects on αsyn aggregation and toxicity. Using a paracrine αsyn model, we found that …

BACKGROUND: Phosphorylation is a crucial post-translational modification mechanism that enhances proteomic diversity, and its malfunction has been confirmed to be associated with complex traits, especially brain disorders. One of the factors contributing to this malfunction is the missense mutations given that they may alter the peptides flanking the phosphorylated residues. However, the specific effects of these missense mutations on phosphorylation remain unclear. METHODS: To ascertain these, a deep learning phosphorylation prediction model (DeepMEP), which is the first to be developed on a …

Alpha-synuclein is a 140 aa neuronal protein pathologically and genetically linked to Parkinson's disease (PD). In PD, the major protein modification of alpha-synuclein is phosphorylation at serine 129. Alpha-synuclein phosphorylation also occurs at low levels in normal brains and cultured CNS neurons, but its regulation and role are only beginning to be investigated. The enteric nervous system is now recognized as a second brain in its own right, and therefore, we set out to examine the regulation of alpha-synuclein phosphorylation in enteric neurons. To this end, primary cultures of rat ente…

The pleiotropic cytokine macrophage migration inhibitory factor (MIF) elevates sarcoplasmic reticulum (SR) Ca2+ content and enhances Ca2+ transient in cardiac muscle. Our imaging and immunoblot data indicated that the MIF-evoked effect is caused mainly by the phosphorylation of the SR Ca2+-pump regulator phospholamban (PLN). Gene expression data suggested that the cluster of differentiation 74 (CD74) and the C-X-C motif chemokine receptor 7 (CXCR7) form a major MIF receptor complex in cardiomyocytes, but CXCR7 activation alone seemed sufficient to exert the MIF-evoked effect. Our pharmacologic…

INTRODUCTION: Most neurodegenerative diseases, including Alzheimer's disease (AD) and multiple sclerosis (MS), feature abnormal tau phosphorylation (p-tau) in the brain. Prior immunostaining studies have shown p-tau accumulation in the AD retina, suggesting it may mirror brain tau pathology.
METHODS: We used mass spectrometry to quantify p-tau peptides in matched retinal and hippocampal samples from non-demented controls (NC, n = 8), AD (n = 12), and MS (n = 4). We compared p-tau levels across diagnoses and analysed correlations between retinal p-tau variants, hippocampal p-tau, and neuropath…

Nuclear factor-κB (NF-κB) is a transcription factor that regulates the expression of various genes involved in inflammatory diseases and immune responses. Recently, a novel transcriptional regulatory mechanism of NF-κB involving the phosphorylation of serine 536 (534 in mice; S534) of its p65 subunit was reported; however, further research is required to elucidate the physiological role of S534 phosphorylation. Therefore, we generated S534A knock-in (KI) mice, in which the S534 of p65 was substituted with alanine. Similar to the wild-type (WT) mice, S534A KI mice developed normally. After stim…
