Molecular insights and integrated management of Glomerella leaf spot in apple.

Apple (Malus × domestica) is one of the world's premier fruit crops. Apple Glomerella leaf spot (GLS) is caused by Colletotrichum spp., jeopardizing global apple supply and incurring millions of dollars in annual losses. The development of resistant apple varieties has evolved from conventional selection breeding to deliberate breeding strategies that incorporate statistical analysis and experimental design, and now increasingly integrate genetic and genomic data. Innovative methods like marker-assisted selection and genetic editing have facilitated the development of cultivars resistant to GL

Mutations of PsPALM1a and PsPALM1b associated with the afila phenotype in Pea.

Semi-leafless represents an advantageous plant architecture in pea breeding due to its ability to enhance resistance to lodging and potentially to powdery mildew. The introduction of semi-leafless pea varieties is considered a seminal advancement in pea breeding over the past half-century. The afila (af) mutation leads to the replacement of lateral leaflets by highly branched tendrils; combined with the semi-dwarfing le mutation, it forms the semi-leafless cultivated variety. In this study, we identified that mutations in two tandemly-arrayed genes encoding Cys(2)His(2) zinc finger transcripti

Targeted allelic diversification of flowering genes via CRISPR-Cas enables the development of multiple early-flowering soybean lines.

CRISPR-Cas multiplex editing generated a broad range of early-flowering soybean lines, advancing flowering by up to four weeks under controlled long-day conditions and by approximately one to two weeks in a field trial. Soybean is a promising crop for European agriculture, but its cultivation area remains smaller than its potential. A major limitation to broader adoption in Europe is the restricted number of elite lines adapted to local environmental conditions, particularly the photoperiod encountered at higher latitude. At these latitudes, soybean must flower early to complete its growth cyc

Improving multi-trait genomic prediction using synthetic traits from hyperspectral data based on co-heritability.

Synthetic traits, wavelength ratios selected by co-heritability, raised multi-trait genomic predictive ability for leaf nitrogen and specific leaf area in sorghum by up to 17% over single-trait models. Genomic prediction (GP) is an essential tool in plant breeding as it can accelerate cultivar development by predicting the performance of unphenotyped lines. When using single-trait GP models, the precision of prediction is constrained by the heritability of the target trait. Multi-trait genomic prediction can be used to improve accuracy but requires identifying secondary traits with high herita…

A time-course transcriptomic analysis reveals the key responses of a resistant rice cultivar to brown planthopper infestation.

The brown planthopper (BPH) is one of the most problematic pests affecting rice (Oryza sativa L.) yields in Asia. Breeding rice varieties containing resistance genes is the most economical and effective means of controlling BPH. In this study, the key factors in resistance to BPH were investigated between the high-resistance rice variety "R26" and the susceptible variety "TN1" using RNA-sequencing. We identified 9527 differentially expressed genes (DEGs) between the rice varieties under BPH-induced stress. Weighted time-course gene co-expression network analysis (WGCNA) indicated that the incr

Genome-wide identification and evolutionary analysis of the ERF-VII gene family in the tea plant (Camellia sinensis) and functional characterization of CsRAP2.2 in response to cold stress.

The ERF-VII gene family, a critical branch of the AP2/ERF superfamily, is central to plant stress adaptation. However, its evolutionary history and function in tea plant (Camellia sinensis) remain unclear. Here, we performed integrated evolutionary, genomic, and functional analyses of ERF-VII genes across 14 plant lineages and 20 tea plant cultivars. The phylogenetic analysis revealed that ERF-VII proteins originated after vascular plant divergence, coinciding with the emergence of the N-terminal MCGGA/I motif linked to the oxygen-dependent N-degron pathway. Gymnosperms retained few conserved

In Vitro Propagation of the Blueberry 'Blue Suede™' (Vaccinium hybrid) in Semi-Solid Medium and Temporary Immersion Bioreactors.

The production of blueberries for fresh and processed consumption is increasing globally and has more than doubled in the last decade. Blueberry is grown commercially across a variety of climates in over 30 countries. The major classes of plants utilized for the planting and breeding of new cultivars are highbush, lowbush, half-high, Rabbiteye, and Southern highbush. Plants can be propagated by cuttings or in vitro micropropagation techniques. In vitro propagation offers advantages for faster generation of a large number of disease-free plants independent of season. Labor costs for in vitro pr

Wild rice locus enables microbiome re-domestication for enhanced nitrogen-use efficiency.

Domestication of crops, such as rice, has resulted in the loss of beneficial root-associated microbiota central to plant nitrogen-use efficiency. Whether these microbes can be inherited through hybridization of wild and cultivated plants and the host genes involved is unclear. Here we used two recombinant inbred line populations, derived from cultivated rice (Oryza sativa) and its wild relative Oryza rufipogon to map host quantitative trait loci associated with core bacterial taxa. Integrating microbiome profiling, quantitative trait loci mapping, population genomics and transgenic validation,

PIF4 enhances temperature-independent tolerance to TSWV in Nicotiana benthamiana.

Tomato spotted wilt virus (TSWV) significantly threatens agricultural productivity, causing significant production and yield losses. Higher temperature have been reported to further intensify the TSWV accumulation and disease severity in several crops. PHYTOCHROME-INTERACTING FACTOR 4 (PIF4) has emerged as a key regulator of plant responses to multiple biotic and abiotic stresses; however, its precise role in plant immunity against TSWV has not been defined. To investigate this, we generated PIF4 knockout mutants and PIF4-overexpression lines of Nicotiana benthamiana by gene-editing and transg

Day-neutral flowering candidate genes identified by selection mapping in an upland cotton (Gossypium hirsutum L.) RIL population.

Variants of flowering time genes identified from selective sweep selection of cotton breeding lines. Day-neutrality is essential for modern upland cotton (Gossypium hirsutum L.) cultivation at temperate latitudes, yet the molecular basis of the transition from photoperiod sensitive to day-neutral achieved by selective breeding remains incompletely understood. We conducted whole-genome resequencing of the day-neutral cultivar DP61, the reniform nematode-resistant line MT2468 Ren1, and 40 recombinant inbred lines (RILs) derived from their cross, all selected for day-neutral flowering. Since MT24

Rice cold tolerance: Physiological responses, molecular regulatory networks, and improvement strategies.

Low-temperature stress is a critical abiotic constraint that severely impairs rice yield and quality, posing a substantial threat to global food security-especially as rice, a staple food for more than half of the world's population, is inherently susceptible to cold stress. This review systematically summarizes the typical phenotypic and physiological impairments induced by cold stress across key rice growth stages, and elaborates on the multi-layered molecular regulatory networks underpinning rice cold tolerance, which integrate the entire process of cold signal perception, intracellular tra

Unravelling the treasure trove of drought-responsive genes in wild-type peanut through transcriptomics and physiological analyses of root.

Peanut is one of the most valuable legumes, grown mainly in arid and semi-arid regions, where its production may be hindered by the lack of water. Therefore, breeding drought tolerant varieties is of great importance for peanut breeding programs around the world. Unlike cultivated peanuts, wild peanuts have greater genetic diversity and are an important source of alleles conferring tolerance/resistance to abiotic and biotic stresses. To decipher the transcriptome changes under drought stress, transcriptomics of roots of highly tolerant Arachis duranensis (ADU) and moderately susceptible A. ste

Are genetically modified plants useful and safe?

So far, plants have been genetically modified essentially to achieve resistance to herbicides, or to pathogens (mainly insects, or viruses), but resistance to abiotic stresses (such as cold, heat, drought, or salt) is also being studied. Genetically modified (GM) plants with improved nutritional qualities have more recently been developed, such as plants containing higher proportions of unsaturated fatty acids (omega-3 and omega-6) in their oil (to prevent cardio-vascular diseases), or containing beta-carotene as in the golden rice (to prevent vitamin A deficiency). Possible risks for human he

Fine mapping of the dominant female sterility gene and novel model of hybrid seed production in cabbage (Brassica oleracea L. var. capitata).

Hybrid breeding based on male sterility requires the removal of male parents, which is time- and labor-intensive; however, the use of female sterile male parent can solve this problem. In the offspring of distant hybridization between Brassica oleracea and Brassica napus, we obtained a mutant, 5GH12-279, which not only fails to generate gynoecium (thereby causing female sterility) but also has serrated leaves that could be used as a phenotypic marker in seedling screening. Genetic analysis revealed that this trait was controlled by a single dominant gene. Further analysis revealed that Bo2g005

Unlocking wheat yield potential through evaluation of high-performing hybrid lines in semiarid conditions of northern Pakistan.

Wheat (Triticum aestivum L.) yield in Pakistan has remained stagnant over the past two decades mainly due to the narrow genetic diversity of cultivated varieties. To overcome this limitation and explore new genetic resources for improved productivity, twelve Chinese wheat hybrid lines were evaluated in comparison with two local cultivars at the Agronomy Research Farm, the University of Agriculture Peshawar, Pakistan over two seasons (2021-2022, 2022-2023). Results showed that early emergence occurred in the hybrid lines 20 BH 37 and 20 BH 53 (12.5 days). Wheat lines 20 BH 53 and 20 BH 56 produ

Estimating on-farm genotypic performance and variability using ranking data.

Our scalable two-step method estimates genotypic performance and genetic parameters from ranking data, producing reliable results comparable to quantitative analyses, enabling the integration of ranking data into breeding pipelines. Plant breeding research has chiefly relied on on-station experiments to evaluate varietal performance. Nevertheless, these trials often fail to represent on-farm growing conditions and farmers' preferences, potentially leading to poorly defined breeding targets. Recent work has demonstrated the potential of using on-farm verification trials combined with ranking da…

Accelerating iron biofortification in millets: progress, challenges, and future prospects.

Integrating conventional breeding, omics, and CRISPR-based genome editing can overcome genetic and antinutrient constraints, enabling efficient iron biofortification of millets for sustainable and nutrition-secure food systems. Iron (Fe) deficiency remains one of the most widespread forms of micronutrient malnutrition. Biofortification of staple crops has emerged as a particularly sustainable and scalable strategy to combat this issue. Millets are nutrient-dense staple cereals with exceptional nutritional quality and climate resilience. However, genetic variations and the presence of antinutri

Assessment of Pollen Viability and Germination Through Histochemical, In Vivo and In Vitro Methods.

Pollen viability is a key parameter for understanding the functioning of plant reproductive systems and for supporting strategies aimed at genetic resource conservation and plant breeding. Accurate assessments of this attribute are essential for identifying genotypes with a higher reproductive rate, monitoring prezygotic barriers, testing compatibility between individuals, and planning controlled crosses, especially in cases of floral asynchrony or spatial isolation. Accordingly, this chapter compiles the main methods used for this purpose, providing detailed and up-to-date protocols. It cover…

Effects of granular feed on reproductive metabolism of breeding pigeons, intestinal development and microbiota of squab pigeons-A double-edged sword.

Pigeons like to eat raw grains, but meat pigeon enterprises often use compound feeds instead of raw grains to feed breeding pigeons to increase economic efficiency, which may change the pigeon's dietary behavior, and consequently lead to health and welfare problems. The purpose of this study was to investigate the effect of granular feeds on the health of high-yielding breeding pigeons and squabs in lactation. The results showed that, compared with raw grain group, the provision of granular feed resulted in lower total feed intake without affecting the weight of lactating breeding pigeons. Mea
