Plant immunity to pathogen infections is a dynamic response that involves multiple organelles and defence signalling systems such as hypersensitive response (HR) and systemic acquired resistance (SAR). The latter requires the function of Pathogenesis-related (PR) proteins, a common plant protein family with diverse roles in plant innate immunity. Our previous proteomics study showed that a PR gene (ITC1587_Bchr9_P26466_MUSBA) was differentially regulated during a compatible banana-M. incognita interaction, substantiating the isolation of this gene in the current study. Here, we successfully is…

Chitosan, a major fungal cell wall component, activates plant immunity, yet the underlying perception mechanism remains unknown. In this study, we identified OsFID1, an apoplast-localized DUF26 domain protein, as a chitosan-binding receptor essential for chitosan-triggered immunity. OsFID1 forms a receptor complex with OsFID2, a plasma membrane-localized DUF26 receptor-like kinase, to mediate chitosan sensing and immune activation, requiring OsFID2 phosphorylation for function. The OsFID1-OsFID2 complex confers broad-spectrum resistance to fungal pathogens. OsFID1 homologs are conserved in lan…

Identified over twenty years ago and distantly related to animal caspases are a group of cysteine proteases known as metacaspases. Throughout the years, much like caspase roles in metazoans, metacaspases have been shown to be involved in regulating cellular death in non-metazoan organisms. Yet, continued research on metacaspases describes these proteins as intricate and multifunctional, displaying striking diversity on distinct biological functions. In this review, we intend to describe the recent advances in our understanding of the divergence of metacaspase functionality in plants and fungi.

Gall midges constitute an important group of plant-parasitic insects. The Hessian fly (HF; Mayetiola destructor), the most investigated gall midge, was the first insect hypothesized to have a gene-for-gene interaction with its host plant, wheat (Triticum spp.). Recent investigations support that hypothesis. The minute larval mandibles appear to act in a manner that is analogous to nematode stylets and the haustoria of filamentous plant pathogens. Putative effector proteins are encoded by hundreds of genes and expressed in the HF larval salivary gland. Cultivar-specific resistance (R) genes med

A 27-aa peptide coding sequence NLP27 was moderately overexpressed in potato, conferring enhancing plant resistance to Phytophthora infestans and Streptomyces scabies infection without yield penalty through plant immunity promotion. The conserved short peptides derived from pathogen effectors can stimulate plant innate immunity response against pathogens through exogenous application. However, it remains to be explored whether the ectopic expression of these peptides in plants can trigger plant defense responses and affect plant growth. Here, we modified the nucleotide sequence of conserved 27…

Dysbiosis, the disruption of plant-associated microbial communities, is increasingly recognized as a driver of impaired plant health. Notably, plant dysbiosis parallels disruptions of the human gut microbiome, where imbalances similarly contribute to host dysfunction and disease. Here, we discuss the current understanding of plant dysbiosis by integrating host-microbiome interactions and emerging analytical methods. Dysbiosis can extend beyond local compartments, affecting systemic processes such as immunity and nutrient acquisition. Advances in sequencing and quantitative metrics now enable i

Accumulating evidence shows that proper degradation of proteins that affect defense responses in a positive or negative manner is critical in plant immunity. However, the role of plant degradation systems such as the 26S proteasome in plant immunity is not well understood. Loss-of-function mutations in EDR2 (ENHANCED DISEASE RESISTANCE 2) lead to increased resistance to the adapted biotrophic powdery mildew pathogen Golovinomyces cichoracearum. To study the molecular interactions between powdery mildew pathogen and Arabidopsis, we performed a screen for suppressors of edr2 and found that mutat…

Plant wall-associated kinases (WAKs) and WAK-like kinases (WAKLs) are closely related subfamilies of receptor-like kinases (RLKs) that bridge the plant cell wall and cytoplasm and mediate essential signaling functions. Beyond their developmental roles, accumulating evidence highlights their important contributions to plant immunity. Initially identified in Arabidopsis thaliana, these proteins have emerged as important regulators of plant responses to biotic stresses. This review focuses on the disease-resistance functions of this protein family and their associated signaling pathways across di…

Nanoparticles are recently employed as a new strategy to directly kill pathogens (e.g., bacteria and fungus) and acted as nanofertilizers. However, the influences of this foliar deposition of nanoparticles on plant physiology particularly plant immunity are poorly understood. The uptake and physiological effects of Fe3O4 nanoparticles (Fe3O4NPs), and plant resistance response against Tobacco mosaic virus (TMV) after foliar spraying were studied. Specifically, Fe3O4NPs entered leaf cells and were transported and accumulated throughout the whole Nicotiana benthamiana plant, and increased plant d…

Plant peptide hormones orchestrate growth, development and immunity by engaging membrane-resident receptor kinases. GmSubPEP, a defence-eliciting peptide derived from a subtilase precursor, was initially isolated from soybean leaf extracts. Here we uncover an unexpected expansion of GmSubPEP variants with highly diverse sequences, yet each capable of triggering robust, tissue-specific pathogen resistance. Integrated transcriptomic and phosphoproteomic profiling shows that individual GmSubPEPs differentially activate overlapping and unique immune pathways, enabling fine-tuned defence modulation

Both above- and below-ground parts of plants are constantly challenged with microbes and interact closely with them. Many plant-growth-promoting rhizobacteria, mostly interacting with the plant's root system, enhance the immunity of plants in a process described as induced systemic resistance (ISR). Here, we characterized local induced resistance (IR) triggered by the model PGPR Pseudomonas simiae WCS417r (WCS417) in Arabidopsis thaliana. Hydroponic application of WCS417 to Arabidopsis roots resulted in propagation of WCS417 in/on leaves and the establishment of local IR. WCS417-triggered loca

Salicylic acid (SA) is a ubiquitous plant hormone with a long history in human civilization1,2. Because of the central role of SA in orchestrating plant pathogen defence, understanding SA biosynthesis is fundamental to plant immunity research and crop improvement. Isochorismate-derived SA biosynthesis has been well defined in Arabidopsis. However, increasing evidence suggests a crucial function for phenylalanine-derived SA biosynthesis in many other plant species1. Here we reveal the phenylalanine-derived SA biosynthetic pathway in rice by identifying three dedicated enzymes - peroxisomal benz…

Iron is critical during host-microorganism interactions1-4. Restriction of available iron by the host during infection is an important defence strategy, described as nutritional immunity5. However, this poses a conundrum for externally facing, absorptive tissues such as the gut epithelium or the plant root epidermis that generate environments that favour iron bioavailability. For example, plant roots acquire iron mostly from the soil and, when iron deficient, increase iron availability through mechanisms that include rhizosphere acidification and secretion of iron chelators6-9. Yet, the elevat

The homeostasis of resistance (R) proteins in plants must be tightly regulated to ensure precise activation of plant immune responses upon pathogen infection, while avoiding autoimmunity and growth defects when plants are uninfected. It is known that CPR1, an F-box protein in the SCF E3 complex, functions as a negative regulator of plant immunity through targeting the resistance (R) proteins SNC1 and RPS2 for degradation. However, whether these R proteins are also targeted by other E3 ligases is unclear. Here, we isolated Arabidopsis MUSE16, which encodes a RING-type E3 ligase, from a forward …

Pseudomonas syringae employs a type III secretion system to inject 20-30 different type III effector (T3SE) proteins into plant host cells. A major role of T3SEs is to suppress plant immune responses and promote bacterial infection. The YopJ/HopZ acetyltransferases are a superfamily of T3SEs found in both plant and animal pathogenic bacteria. In P. syringae, this superfamily includes the evolutionarily diverse HopZ1, HopZ2 and HopZ3 alleles. To investigate the roles of the HopZ family in immunomodulation, we generated dexamethasone-inducible T3SE transgenic lines of Arabidopsis for HopZ family
