Age-related Changes of GAD1 mRNA Expression in the Central Inferior Colliculus.

Encoding sounds with a high degree of temporal precision is an essential task for the inferior colliculus (IC) to perform and maintain the accurate processing of sounds and speech. However, the age-related reduction of GABAergic neurotransmission in the IC interrupts temporal precision and likely contributes to presbycusis. As presbycusis often manifests at high or low frequencies specifically, we sought to determine if the expression of mRNA for glutamic decarboxylase 1 (GAD1) is downregulated non-uniformly across the tonotopic axis or cell size range in the aging IC. Using single molecule in…

Major arc mitochondrial DNA deletions in cytochrome c oxidase-deficient human cochlear spiral ganglion cells.

CONCLUSIONS: This study suggests that cytochrome c oxidase subunit 3 (COX 3) expression is diminished in spiral ganglion cells from individuals with presbycusis. In addition to the mitochondrial DNA (mtDNA) common deletion (CD), other deletions involving the mtDNA major arc contribute to the observed deficit in COX 3 expression. OBJECTIVES: To assess COX 3 deficiency in spiral ganglion cells from individuals with presbycusis and to determine whether deletions other than the CD contribute to this deficiency. METHODS: COX 3 immunofluorescence staining of archival human temporal bone tissue sec…

Age-related hearing loss: aquaporin 4 gene expression changes in the mouse cochlea and auditory midbrain.

Presbycusis -- age-related hearing loss, is the number one communication disorder, and one of the top three chronic medical conditions of our aged population. Aquaporins, particularly aquaporin 4 (Aqp4), are membrane proteins with important roles in water and ion flux across cell membranes, including cells of the inner ear and pathways of the brain used for hearing. To more fully understand the biological bases of presbycusis, 39 CBA mice, a well-studied animal model of presbycusis, underwent non-invasive hearing testing as a function of sound frequency (auditory brainstem response -- ABR thre…

[Pathophysiology of auditory and speech perception].

Auditory perception or hearing can be defined as the interpretation of sensory evidence, produced by the ears in response to sound, in terms … environment, can give rise to considerable psychosocial and professional consequences (disability). Presbycusis is the decline in hearing sensitivity caused by the aging process at different levels of the auditory system. However, it is difficult to isolate age effects from other contributors to age-related hearing loss such as noise damage, genetic susceptib …

The ABHD10-KCMF1 Complex Mitigates Cochlear Aging by Regulating Mitochondrial Lipid Metabolism.

Presbycusis is caused by multiple factors, the mechanisms of which are not fully understood. This study investigated mitochondria-related genes in presbycusis, a condition with multifactorial and incompletely understood mechanisms, by combining Mendelian randomization analysis and functional validation. Genetically predicted higher ABHD10 expression was identified as a protective factor against presbycusis and was regulated by methylation sites such as cg15684481, whereas methylation at other sites inhibited ABHD10 expression and alleviated the condition. In contrast, ABHD10 was upregulated in…

Age-related changes in mitochondrial antioxidant enzyme Trx2 and TXNIP-Trx2-ASK1 signal pathways in the auditory cortex of a mimetic aging rat model: changes to Trx2 in the auditory cortex.

Age-associated degeneration in the central auditory system, which is defined as central presbycusis, can impair sound localization and speech perception. Research has shown that oxidative stress plays a central role in the pathological process of central presbycusis. Thioredoxin 2 (Trx2), one member of thioredoxin family, plays a key role in regulating the homeostasis of cellular reactive oxygen species and anti-apoptosis. The purpose of this study was to explore the association between Trx2 and the phenotype of central presbycusis using a mimetic aging animal model induced by long-term exposu…

Over-the-Counter Hearing Aids: A Lost Decade for Change.

BACKGROUND: Hearing aids sold directly to consumers in retail stores or through the internet, without individual prescription by audiological professionals, are termed over-the-counter (OTC) devices. This study aimed to determine whether there was any change in the electroacoustic characteristics of OTC devices compared to research carried out a decade earlier. The previous results indicated that most OTC devices were low-frequency-emphasis devices and were unsuitable for elderly people with presbycusis, who were likely to be the major consumers of these products. METHODS: Ten OTC devices wer…

GRM7 variants associated with age-related hearing loss based on auditory perception.

Age-related hearing impairment (ARHI), or presbycusis, is a common condition of the elderly that results in significant communication difficulties in daily life. Clinically, it has been defined as a progressive loss of sensitivity to sound, starting at the high frequencies, inability to understand speech, lengthening of the minimum discernable temporal gap in sounds, and a decrease in the ability to filter out background noise. The causes of presbycusis are likely a combination of environmental and genetic factors. Previous research into the genetics of presbycusis has focused solely on hearin…

[Audiological results in hearing loss in old age].

A significant loss of performance of the ear was found to be a function of age in all audiological tests. Clear cases of presbycusis were only determined in persons above 70 years of age. According to finds, a severely handicapping loss in the ability to understand speech only occurred in the 90-year-old group suffering from primary presbycusis. A disturbing impairment of hearing in younger persons is most likely due to a secondary presbycusis. Contrary to previous opinions suprathreshold tests indicate a high percentage of sensoneural hearing losses are due to cochlear lesions. The dichotic d…

Transcriptomic analysis reveals the function of m6A regulators in aged cochlea.

OBJECTIVE: Presbycusis is a prevalent health issue among the elderly. Previous studies have shown mechanisms related to this condition, but the underlying mechanisms of presbycusis remain elusive. N6-methyladenosine (m6A) modification in regulating gene expression and cellular functions has been implicated in the development of various diseases. Nevertheless, the potential role of m6A regulators in presbycusis is still unclear. In this study, we aim to determine the expression of m6A regulators in the cochleae of young and old mice, and to investigate their potential role in aging. METHODS: W…

Diffusion tensor imaging and MR morphometry of the central auditory pathway and auditory cortex in aging.

Age-related hearing loss (presbycusis) is caused mainly by the hypofunction of the inner ear, but recent findings point also toward a central component of presbycusis. We used MR morphometry and diffusion tensor imaging (DTI) with a 3T MR system with the aim to study the state of the central auditory system in a group of elderly subjects (>65years) with mild presbycusis, in a group of elderly subjects with expressed presbycusis and in young controls. Cortical reconstruction, volumetric segmentation and auditory pathway tractography were performed. Three parameters were evaluated by morphometry…

Functional changes in the human auditory cortex in ageing.

Hearing loss, presbycusis, is one of the most common sensory declines in the ageing population. Presbycusis is characterised by a deterioration in the processing of temporal sound features as well as a decline in speech perception, thus indicating a possible central component. With the aim to explore the central component of presbycusis, we studied the function of the auditory cortex by functional MRI in two groups of elderly subjects (>65 years) and compared the results with young subjects (<lt;30 years). The elderly group with expressed presbycusis (EP) differed from the elderly group with m…

The effect of overexpression of PGC-1α on the mtDNA4834 common deletion in a rat cochlear marginal cell senescence model.

Aging is a natural process usually defined as a progressive loss of function with an accumulation of senescent cells. The clinical manifestations of this process include age-related hearing loss (AHL)/presbycusis. Several investigations indicated the association between a mitochondrial common deletion (CD) (mtDNA 4977-bp deletion in humans, corresponding to 4834-bp deletion in rats) and presbycusis. Previous researches have shown that peroxisome proliferator-activated receptor-gamma coactivator-1α (PGC-1α) is a key regulator of mitochondrial biogenesis and energy metabolism. However, the expre…

Pathophysiological changes of the cochlea during ageing--a review.

Research of the pathophysiology of the age-related human presbycusis was largely initiated by those investigations in which the cochlea's damages were experimentally induced by various acoustic exposures. It became apparent that the outer hair cells are more vulnerable to different noise influences than the inner hair cells. On the other hand, the role of the altered ultrastructural elements of various types, cannot be ruled out. The evolving of the age-related presbycusis was to a great extent established using large-scale model experiments, including mainly rodents. It seems that the age-bou…

Altered cortical spectrotemporal processing with age-related hearing loss.

Presbycusis (age-related hearing loss) is a prevalent disability associated with aging that impairs spectrotemporal processing, but the mechanisms of such changes remain unclear. The goal of this study was to quantify cortical responses to frequency-modulated (FM) sweeps in a mouse model of presbycusis. Previous studies showed that cortical neurons in young mice are selective for the rate of frequency change in FM sweeps. Here single-unit data on cortical selectivity and response variability to FM sweeps of either direction and different rates (0.08-20 kHz/ms) were compared across young (1-3 m…

Morphological correlates of aging in the chinchilla cochlea.

The inner ears from 80 chinchillas ranging in age from premature to 19.2 years were examined as plastic-embedded flat preparations to determine the morphological changes associated with aging. Three of the four forms of human presbycusis defined by Schuknecht were found in the chinchillas. All animals had losses of sensory cells or sensory presbycusis. Inner (IHCs) and outer hair cells (OHCs) degenerated at a rate of about 0.29% and 1.0% per year, respectively. Age-related degeneration of inner (IPs) and outer pillars (OPs) occurred at a much slower rate. In four animals (5%) the dendritic pro…

Age-related cochlear degeneration in senescence-accelerated mouse.

Age-related hair cell loss and strial atrophy were investigated in an accelerated senescence-prone strain, SAMP1 mice, and an accelerated se … in SAMP1 than in SAMR1. Hearing impairment in SAM may be due to a combination of sensory and strial presbycusis as well as to neural presbycusis, as reported previously. The morphological changes in the cochlea observed in SAMP1 and SAMR1 make these strains suitable for the study of the mechanisms of presbycusis.

[Image analysis of arterial vessels of the internal auditory meatus during presenile and aged with hypertension and atherosclerosis].

The image analysis of the arterial vessels of the internal auditory meatus in patients with atherosclerosis and hypertension during presenility and presbycusis was done. The inner and outer diameters of the blood vessels in these patients reduced more markedly than those in the controls. There was a negative correlation between the vessel diameter and correlation age. The lumen of the vessels reduced more in patients with atherosclerosis and hypertension than those patients without these conditions. The tunica adventitia showed fibroid thickness. It suggests that the progress of the vessel cha…
