Exogenous Pyrroloquinoline Quinone Application Enhances Low-Phosphorus Adaptation in Rice by Modulating Redox Homeostasis Under Variable Light.

Phosphorus (P) bioavailability in soil is often limited, restricting plant growth. Pyrroloquinoline quinone (PQQ) is a natural quinone compound with potent redox-cycling capabilities. In this study, we investigated the physiological effects of exogenously applied PQQ on rice adaptation to P deficiency. A P-sensitive rice cultivar (Meixiangzhan) was treated with 0.1 μM PQQ under different light intensities (High Light, HL: 800 μmol m-2 s-1; Low Light, LL: 300 μmol m-2 s-1) and P supplies (Sufficient P, SP; Deficient P, DP). The effects of PQQ on rice growth, P uptake, photosynthesis, and the an

Facile Biological Oxidation of Dopamine to 6-Hydroxydopamine p-Quinone in a Sequential Two-Step Process: Implications for Parkinson's Disease.

6-Hydroxydopamine (6-OHDA), a hydroxyl-derivative of the endogenous neurotransmitter dopamine, can selectively induce Parkinsonian symptoms in animal models. At present, most researchers consider 6-OHDA a man-made neurotoxicant, due to the lack of strong evidence for its presence and/or formation in biological systems. The present study aims to determine whether 6-OHDA can be formed under physiologically relevant conditions. Here, we report in the Fenton reaction system (containing 15 μM Fe2+, 142 μM ascorbic acid and 80 μM EDTA in 50 mM phosphate buffer, pH 7.4), dopamine can undergo facile o

Improvement of spermogenesis impairment induced by high-fat diet in obese mice through pyrroloquinoline quinone regulation of glycolysis pathway.

This study employed an obese mouse model to investigate the effects of pyrroloquinoline quinone (PQQ) intervention on reproductive physiology and glycolytic pathways. Our findings demonstrate that PQQ markedly enhanced glycolytic activity in obese mice, promoting glucose breakdown and metabolism to improve energy supply efficiency. These metabolic improvements correlated with significant support for reproductive system functionality. Mechanistic analyses revealed PQQ's potential to augment mitochondrial respiration, ameliorate mitochondrial dysfunction, and counteract obesity-associated inflam

Multi-spectroscopic, thermodynamic and molecular simulation studies on binding of pyrroloquinoline quinone with DNA: coexistence of intercalation and groove binding modes.

The interaction between enzyme-like pyrroloquinoline quinone (PQQ) and calf-thymus DNA (CT-DNA) has been investigated by means of multi-spectroscopic (UV-Vis, fluorescence and circular dichroism), isothermal titration calorimetric (ITC), viscometry and molecular docking and metadynamics simulation techniques. Absorption spectral data suggested the formation of a PQQ/CT-DNA complex, which quenched the fluorescence of PQQ via the dynamic quenching process. The results of CD spectral studies coupled with viscosity measurements, competitive binding assays with Hoechst 33258 and ethidium bromide (E

Ruthenium-Catalyzed 1,6-Hydroalkylation of para-Quinone Methides with Ketones via the in Situ Activation of C(sp3)-H Bonds.

A simple and efficient method for the ruthenium-catalyzed 1,6-hydroalkylation of para-quinone methides (p-QMs) with ketones via the in situ activation of C(sp3)-H bonds has been disclosed. Without the need for preactivation of the substrates and oxidant, a broad range of p-QMs and ketones are well tolerated, producing the expected 1,6-hydroalkylation products with moderate to good yields. Step-by-step control experiments and DFT calculation were conducted systematically to gain insights for the plausible reaction mechanism. This finding may have potential application in the selective diarylmet

Computational Study of Aromatic Hydroxylation Catalyzed by the Iron-Dependent Hydroxylase PqqB Involved in the Biosynthesis of Redox Cofactor Pyrroloquinoline Quinone.

PqqB from Methylobacterium extorquens is a unique nonheme iron-dependent hydroxylase involved in the biosynthesis of redox cofactor pyrroloquinoline quinone (PQQ). A series of recent experiments have demonstrated that PqqB catalyzes the stepwise insertions of two oxygen atoms into the tyrosine ring of the diamino acid substrate, generating the quinone moiety of PQQ; however, the reaction details have not been elucidated yet. In this paper, on the basis of the crystal structures, the enzyme-substrate complex models were constructed, and the catalytic mechanism of PqqB was explored by performing

Leflunomide induces NAD(P)H quinone dehydrogenase 1 enzyme via the aryl hydrocarbon receptor in neonatal mice.

Aryl hydrocarbon receptor (AhR) has been increasingly recognized to play a crucial role in normal physiological homeostasis. Additionally, disrupted AhR signaling leads to several pathological states in the lung and liver. AhR activation transcriptionally induces detoxifying enzymes such as cytochrome P450 (CYP) 1A and NAD(P)H quinone dehydrogenase 1 (NQO1). The toxicity profiles of the classical AhR ligands such as 3-methylcholanthrene and dioxins limit their use as a therapeutic agent in humans. Hence, there is a need to identify nontoxic AhR ligands to develop AhR as a clinically relevant d

Oxidation chemistry of norepinephrine: partitioning of the O-quinone between competing cyclization and chain breakdown pathways and their roles in melanin formation.

Aberrant oxidation of norepinephrine (1) via the transient o-quinone has been implicated as a critical pathogenetic mechanism underlying the degeneration of noradrenergic cell bodies in the locus coeruleus in Parkinson's disease, the degeneration of noradrenergic nerve terminals in Alzheimer's disease and following transient cerebral ischemia, and the onset and progression of idiopathic vitiligo. An oxidative pathway of 1 is also believed to account for the slow deposition of neuromelanin in pigmented neurons of the locus coeruleus. Remarkably, after extensive investigations spanning over seve

Drug-protein conjugates--XVIII. Detection of antibodies towards the antimalarial amodiaquine and its quinone imine metabolite in man and the rat.

A specific enzyme-linked immunosorbent assay (ELISA) was developed for the detection and characterisation of antibodies directed against amodiaquine (AQ), an anti-malarial drug associated with agranulocytosis and liver damage in man. The assay incorporated an antigen which was produced by the reaction of amodiaquine quinone imine (AQQI), a protein reactive product produced from AQ by silver oxide oxidation, and metallothionein. The protein-conjugate (AQ-MT) had a ratio of AQ to protein of 5.2:1. Specific anti-drug antibody was defined as the differential binding to AQ-MT and unconjugated MT wh

Impairment of pentose phosphate pathway mediates 6-PPD quinone-induced neurotoxicity by inhibiting NADPH-dependent antioxidant defense.

6-PPD quinone (6-PPDQ) is the derivative from N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine (6-PPD) after reaction with environmental ozone. Pentose phosphate pathway (PPP) serves as a principal source of nicotinamide adenine dinucleotide phosphate (NADPH). However, association between disruption in PPP and 6-PPDQ neurotoxicity remains largely unclear. In Caenorhabditis elegans, we first examined the role of PPP derived NADPH in controlling 6-PPDQ neurotoxicity. Exposure to 0.1-10 µg/L 6-PPDQ decreased NADPH level and inhibited expression of gspd-1 and T25B9.9 encoding PPP related enzymes

Cold plasma-induced oxidation of tannic acid enhances bactericidal activity via quinone-driven singlet oxygen generation.

This study investigates the potential of cold plasma to induce a pro-oxidant transition in tannic acid for antimicrobial applications. Cold plasma treatment of aqueous tannic acid produced plasma-activated tannic acid solution (PAT) with distinct oxidative structural modifications. Browning kinetics and spectroscopic analyses (FTIR and XPS) indicated the conversion of tannic acid into reactive quinone structures without degradation into gallic acid. This oxidative transformation triggered the generation of reactive oxygen species (ROS), including superoxide anion, hydrogen peroxide, and single

Green, Formaldehyde-Free Bio-Adhesive from Soybean Meal and Laccase-Oxidized Tannin via Quinone-Amine Crosslinking.

To develop a fully green and non-toxic wood adhesive with improved water resistance and bonding performance for soybean meal (Glycine max (L.) Merr.)-based adhesives, oxidized tannin (OTN) was obtained by the laccase treatment of waxberry tannin (TN), a natural polyphenolic polymer, and then blended with soybean meal (SM) to prepare an oxidized tannin-soybean meal adhesive (OTS). Laccase-mediated oxidation converted the tannin polymer into quinone-rich oxidized polymeric structures, which reacted with amino groups in soybean meal proteins through Michael addition and Schiff base reactions to f

Moderately Flexible Copper-Triazolate Framework with Quinone-Based Linkers: Balancing Effective CO2 Capture and Low-Energy Regeneration.

The development of advanced physisorption-based adsorbents is essential for carbon dioxide (CO2) capture technologies. While metal-organic frameworks (MOFs) offer a promising alternative to traditional carbon capture methods, they often face a trade-off between high adsorption affinity and energy-intensive regeneration. In this work, we report an anionic copper-triazolate framework, NCKU-56 (NCKU = National Cheng Kung University), incorporating quinone-based organic linkers. The 1D channels exhibit moderate structural flexibility and reversible lattice adjustments that are specifically correla

BF3·OEt2 Promoted 1,6-Conjugate Addition of para-Quinone Methides: A Regioselective Approach to Diarylmethyl-Substituted Carbo- and Heterocycles.

In this work, we report a base-free and transition metal-free 1,6-conjugate addition strategy for synthesizing unsymmetrical diaryl-substituted methanes possessing carbo- and heterocycles such as 1,3-indandione, aminouracils, and barbituric acid derivatives from para-quinone methides (p-QMs) using BF3·OEt2 as promoter. The high functional group tolerance, good to excellent yields, and mild reaction conditions of this protocol underscore its significant potential for the synthesis of a broad spectrum of titled products. Furthermore, gram-scale synthesis and versatile subsequent conversions high

Pyrroloquinoline quinone targets senescent osteoclasts and reduces spinal pain and endplate degeneration in a lumbar spine instability mouse model.

BACKGROUND: Pyrroloquinoline Quinone (PQQ) is a redox cofactor with potent antioxidant and anti-inflammatory properties. This study investigated the therapeutic potential of PQQ in alleviating spinal pain and degeneration in a lumbar spine instability (LSI) mouse model. LSI is commonly associated with spinal pain and structural degeneration, with senescent osteoclasts (SnOCs) involved in spinal hypersensitivity and degeneration. METHODS: LSI was induced in 8-week-old C57/BL6 mice by surgically resecting the L3–L5 spinous processes and associated ligaments. The mice were treated with PQQ, and b

Studies on 1,4-Quinone Derivatives Exhibiting Anti-Leukemic Activity along with Anti-Colorectal and Anti-Breast Cancer Effects.

Colorectal cancer (CRC), breast cancer, and chronic myeloid leukemia (CML) are life-threatening malignancies worldwide. Although potent therapeutic and screening strategies have been developed so far, these cancer types are still major public health problems. Therefore, the exploration of more potent and selective new agents is urgently required for the treatment of these cancers. Quinones represent one of the most important structures in anticancer drug discovery. We have previously identified a series of quinone-based compounds (ABQ-1-17) as anti-CML agents. In the current work, ABQ-3 was ta…

The Therapeutic Potential of Imidazole or Quinone-Based Compounds as Radiosensitisers in Combination with Radiotherapy for the Treatment of Head and Neck Squamous Cell Carcinoma.

The addition of platinum chemotherapy to primary radiotherapy (chemoradiation) improves survival outcomes for patients with head and neck squamous cell carcinoma (HNSCC), but it carries a high incidence of acute and long-term treatment-related complications, resulting in a poor quality of life. In addition, patients with significant co-morbidities, or older patients, cannot tolerate or do not benefit from concurrent chemoradiation. These patients are often treated with radiotherapy alone resulting in poor locoregional control and worse survival outcomes. Thus, there is an urgent need to assess
