Otitis media (OM) belongs to the most common pediatric diseases and causes more medical contacts, surgical interventions, and drug prescriptions than any other infectious disease. Recent findings have identified a critical role of innate immunity in recovery from OM. The middle ear mucosa identifies invading pathogens by sensing pathogen-associated molecule patterns (PAMPs) via pattern recognition receptors such as the Toll-like receptors (TLRs). They generate immediate antimicrobial responses and cytokine release, leading to an inflammatory reaction as seen in acute or chronic OM. Cross-talk

BACKGROUND AND PURPOSE: Thromboxane A2 (TXA2) is a prostanoid produced during platelet activaton, important in enhancing platelet reactivity by activation of TP receptors. However, due to the short half-life, studying TXA2 signalling is challenging. To enhance our understanding of TP receptor-mediated platelet biology, we therefore synthesised mono and difluorinated TXA2 analogues and explored their pharmacology on heterologous and endogenously expressed TP receptor function.
EXPERIMENTAL APPROACH: Platelet functional and signalling responses were studied using aggregometry, Ca2+ mobilisation

The nasal cavity displays immune tolerance to commensal bacteria under homeostatic conditions, which is rapidly converted to a pro-inflammatory response upon infection. Yet, the factors that control this conversion are still largely unknown. Here, we provide evidence that Fc gamma receptor III (FcγRIII) stimulation breaks immune tolerance to bacteria in the human nasal cavity through activation of nasal epithelial cells, which are the first line of defense against invading microbes. While under steady-state conditions human nasal epithelial cells were completely non-responsive to Gram-negative

A cross-talk between diabetes and malaria within-host is well established. Diabetes is associated with modulation of the immune system, impairment of the healing process and to disturb the host metabolism to contribute towards propagation of parasite infection. Glucose metabolism in host is maintained by insulin and RBC has 2000 insulin receptor present on plasma membrane. These receptors are robust to relay down-stream signaling in RBCs but role of intracellular signaling in parasite growth is not been explored. The malaria parasite treated with insulin (100 ng/ml) is giving stimulation in pa

OBJECTIVE: The objective of the study was to determine whether cross-talk occurs between estrogen receptors (ERs) and nuclear factor-kappa-B (NF-kappaB), to assess the functional consequences of such an ER/NF-kappaB interaction, and to identify other unknown regulatory proteins that may participate in the NF-kappaB transcriptional complex.
STUDY DESIGN: Electromobility gel shifts, reporter gene assays, and mass spectrometry were used to identify proteins interacting with the NF-kappaB deoxyribonucleic acid (DNA) response element.
RESULTS: ER and the p65 subunit of NF-kappaB colocalized on DN

PROBLEM: Toll-like receptors (TLRs) are innate immune receptors that mediate the pattern recognition of, and response toward, pathogens and host-derived danger signals. We reported that cyclooxygenase-2 (COX-2) and microsomal prostaglandin E synthase (mPGES) mRNA were expressed in cases of endometriosis. The relationship between COX-2, mPGES-1, and TLR4 in endometriotic lesions has yet to be determined.
METHOD OF STUDY: Endometriosis samples were obtained from 37 patients with endometrial cysts. Endometrial tissues were obtained from patients undergoing surgical procedures for benign gynecolo

Neurodegenerative disorders arise from the convergence of oxidative stress, neuroinflammation, synaptic dysfunction, mitochondrial failure, and dysregulated cell death, highlighting the need for therapeutic agents capable of coordinated, multi-target modulation. Luteolin (3',4',5,7-tetrahydroxyflavone), a dietary flavonoid widely present in edible and medicinal plants, has emerged as a promising neuroactive compound with pleiotropic biological actions. Rather than focusing on isolated outcomes, this review presents an integrated summary of how luteolin orchestrates interconnected signaling net

Nesfatin-1 is a multifunctional peptide that integrates cardiovascular, metabolic, and immune regulation, particularly along the "Heart-Brain-Gut" axis. Accumulating evidence highlights its pivotal role in inter-organ communication under both physiological and pathological conditions. However, its underlying molecular mechanisms remain incompletely understood, and the prevailing orphan G protein-coupled receptor (GPCR) hypothesis is currently undergoing critical scrutiny due to accumulating data that contest its exclusivity and sufficiency in accounting for nesfatin-1's pleiotropic effects. Influenza A virus (IAV) remains a major public health threat due to its high genetic variability and ability to evade host immune responses. While antiviral drugs and vaccines remain the key measures against influenza infection, their limited efficacy and the emergence of resistant viral strains have prompted growing interest in exploring complementary therapeutic approaches. In this review, we critically evaluate current evidence on how plant-derived compounds modulate innate immune signaling networks, including toll-like receptor, retinoic acid-inducible gene I, mitogen-activated protein kin

Over the past decade, colchicine has re-emerged as a promising therapeutic candidate for atherosclerotic cardiovascular disease. Here we review evidence from large randomized controlled trials together with advances in mechanistic research that have clarified how colchicine modulates vascular inflammation and plaque stability. Canonically, colchicine disrupts microtubule dynamics and suppresses NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome activation. Beyond these actions, it limits oxidative stress, modulates cytoskeletal cross-talk, attenuates cholesterol crystal fo

Cholera toxin (CT) is a bacterial exotoxin secreted by Vibrio cholerae. CT is comprised of one toxic A subunit (CTA) and five nontoxic B subunits (CTB). CTB is required for internalization of CT through binding with a membrane glycolipid ganglioside GM1. Previously, we clarified that CTB could induce production of proinflammatory cytokine interleukin-1β (IL-1β) from murine resident peritoneal macrophages (RPMs) in synergy with lipopolysaccharide (LPS) through a ganglioside GM1-dependent manner. Then, CTB was incorporated into endoplasmic reticulum (ER) depending on GM1 and activate ER stress s

Recurrent cystic echinococcosis (RCE) occurs by the spillage of live protoscoleces during unsuccessful surgery or spontaneous rupturing of fertile hydatid cyst. Currently, RCE presents a significant concern within surgical community. Managing RCE cases is challenging due to limitations in diagnostic follow-up, therapeutic pitfalls, emerging drug resistance, and innate immunological responses between host-parasite cross-talk. Recent advances suggest that parasite-derived microRNAs hold promise as biomarkers for early detection and monitoring of RCE. Additionally, combining benzimidazoles (paras

How cancer-associated chromatin abnormalities shape tumor-immune interaction remains incompletely understood. Recent studies have linked DNA hypomethylation and de-repression of retrotransposons to anti-tumor immunity through the induction of interferon response. Here, we report that inactivation of the histone H3K36 methyltransferase NSD1, which is frequently found in squamous cell carcinomas (SCCs) and induces DNA hypomethylation, unexpectedly results in diminished tumor immune infiltration. In syngeneic and genetically engineered mouse models of head and neck SCCs, NSD1-deficient tumors exh

The coexistence of osteogenesis imperfecta and inflammatory arthritis has been very rarely described. Nevertheless, systemic inflammation has been found in osteogenesis imperfecta. The COL1A1 mutations may affect collagen synthesis as well as post-translational modifications, extracellular matrix interactions, and receptor-mediated signaling. Major collagen binding ligands forming the interactome, such as cytokines, cell adhesion molecules, matrix metalloproteinases, proteoglycans, and other molecules, are autoimmunity targets involved in rheumatoid arthritis pathogenesis. Mast cells (MCs) are important effector cells in asthma and pulmonary inflammation, and their proliferation and maturation is maintained by stem cell factor (SCF) via its receptor, c-Kit. Cysteinyl leukotrienes (cys-LTs) are potent inflammatory mediators that signal through CysLT1 R and CysLT2 R located on the MC surface, and they enhance MC inflammatory responses. However, it is not known if SCF and cys-LTs cross-talk and influence MC hyperplasia and activation in inflammation. Here, we report the concerted effort of the growth factor SCF and the inflammatory mediator LTD4 in MC activation. S

Here, we have studied vascular endothelial growth factor receptor-3 (VEGFR-3) expression in mycosis fungoides (MF), the most common type of cutaneous T-cell lymphoma (CTCL). Immunohistochemistry revealed that in two-thirds of 34 patients, VEGFR-3 was expressed in situ by both tumor and stromal cells irrespective of the disease stage. The natural VEGFR-3 ligand, VEGF-C, partially protected malignant T-cell lines from growth inhibition by the histone deacetylase inhibitor, suberoylanilide hydroxamic acid (SAHA). Whereas the malignant T cells did not produce VEGF-C in vitro, its expression was in

Altered protein homeostasis underlies degenerative diseases triggered by misfolded proteins, including spinal and bulbar muscular atrophy (SBMA), a neuromuscular disorder caused by a CAG/glutamine expansion in the androgen receptor. Here we show that the unfolded protein response (UPR), an ER protein quality control pathway, is induced in skeletal muscle from SBMA patients, AR113Q knock-in male mice, and surgically denervated wild-type mice. To probe the consequence of UPR induction, we deleted CHOP (C/EBP homologous protein), a transcription factor induced following ER stress. CHOP deficiency

Parathormone (PTH) and vitamin D are two critical hormonal regulators of calcium homeostasis. An important cross-talk exists between the PTH and vitamin D hormonal systems. PTH enhances vitamin D hydroxylation on carbon 1 in kidney cells thereby allowing the systemic release of 1-25-dihydroxy-vitamin D, which represents the fully active hormone. Conversely, parathyroid gland represents a direct target for vitamin D. Parathyroid cells express the vitamin D receptor and the 1-α-hydroxylase enzyme, which allows the local formation of 1-25-dihydroxy-vitamin D. Because of its potential implication

Lymphangioleiomyomatosis (LAM) is a potentially fatal lung disease characterized by nodules of proliferative smooth muscle-like cells. The exact nature of these LAM cells and their proliferative stimuli are poorly characterized. Herein we report the novel findings that the lymphangiogenic vascular endothelial growth factors (VEGF) C and D induce LAM cell proliferation through activation of their cognate receptor VEGF-R3 and activation of the signaling intermediates Akt/mTOR/S6. Furthermore, we identify expression of the proteoglycan NG2, a marker of immature smooth muscle cells, as a character

Cell-cell interactions and cross-talk between signaling pathways specify Caenorhabditis elegans vulval precursor cells (VPCs) to adopt a spatial pattern: a central "1 degrees " VPC, in which epidermal growth factor receptor (EGFR)-mitogen-activated protein kinase (MAPK) activity is high and LIN-12/Notch activity is low, flanked by two "2 degrees " VPCs, in which LIN-12/Notch activity is high and EGFR-MAPK activity is low. Here, we identify a microRNA gene, mir-61, as a direct transcriptional target of LIN-12 and show that expression of mir-61 promotes the 2 degrees fate. We also identify vav-1
