Alpha-, Beta-, and Gamma-synuclein Quantification in Cerebrospinal Fluid by Multiple Reaction Monitoring Reveals Increased Concentrations in Alzheimer's and Creutzfeldt-Jakob Disease but No Alteration in Synucleinopathies.

α-Synuclein (αSyn) is a major constituent of proteinaceous aggregates in neurodegenerative diseases such as Parkinson's disease (PD) and a potential biomarker candidate for diagnosis and treatment effects. However, studies about αSyn in cerebrospinal fluid (CSF) in diseases are inconsistent and mainly based on immunological assays. Quantitative information about β-synuclein (βSyn) and γ-synuclein (γSyn) in CSF is not available. Here, we present an alternative method for the simultaneous quantification of αSyn, βSyn and γSyn in CSF by multiple reaction monitoring (MRM) with a high sequence cover

Development and validation of a GEANT4 radiation transport code for CT dosimetry.

The authors have created a radiation transport code using the GEANT4 Monte Carlo toolkit to simulate pediatric patients undergoing CT examinations. The focus of this paper is to validate their simulation with real-world physical dosimetry measurements using two independent techniques. Exposure measurements were made with a standard 100-mm CT pencil ionization chamber, and absorbed doses were also measured using optically stimulated luminescent (OSL) dosimeters. Measurements were made in air with a standard 16-cm acrylic head phantom and with a standard 32-cm acrylic body phantom. Physical dose

A fast and sensitive method for measuring picomole levels of total free amino acids in very small amounts of biological tissues.

In the present study we describe a simple and fast method to measure the concentration of total free amino acids in very small amounts of biological tissues. The procedure described here is based on the reaction of free amino acids with o-phthaldialdehyde (OPA) in the presence of a reducing agent, beta-mercaptoethanol (MET), to give a complex which can be measured by fluorescence. It is a very rapid process and has the same reliability as the conventional ninhydrin method of Moore and Stein but is about 500 times more sensitive. The sensitivity of the new protocol is such to permit the determi

Increased vigabatrin entry into the brain by polysorbate 80 and sodium caprate.

The effects of a non-ionic surfactant, polysorbate 80, and the sodium salt of the saturated fatty acid, sodium caprate (C10), as potential brain absorption enhancers for vigabatrin were studied. Vigabatrin is an enzyme-activated irreversible inhibitor of gamma-aminobutyric acid (GABA) transaminase that increases brain and cerebrospinal GABA concentrations in animals and man. Before intravenous administration, a range of concentrations of the surfactants were tested using erythrocyte lysis or the red blood cell lysis test to establish the non-toxic concentration range. Vigabatrin was dissolved

Spectroscopic studies on bleomycin-iron complexes with carbon monoxide, nitric oxide, isocyanide, azide, and cyanide and comparison with iron-porphyrin complexes.

The bleomycin-iron complexes with CO, NO, C2H5NC, OH-, N-3, CN-, and CH3NH2 were characterized by electronic, ESR, 1H-NMR, and Mössbauer spectroscopies and the findings were compared with the corresponding hemoprotein complexes. The 1H-NMR and Mössbauer features for the CO and C2H5NC adducts of the bleomycin-Fe(II) complex are consistent with an S = 0 ferrous assignment. The OH-, CH3NH2, and N-3 adducts of the bleomycin-Fe(III) complex show the ESR, 1H-NMR, and Mössbauer spectra typical of a low-spin Fe(III). The unique Mössbauer parameters of the bleomycin-Fe(II)-NO complex demonstrate mixing

An integrated experimental-deterministic photon transport framework for radiation dosimetry: Application to naturally occurring radioactive building materials.

Conventional radiological assessment of building materials predominantly relies on empirical hazard indices that provide screening information but do not explicitly describe photon transport within finite material geometries. This study presents an integrated experimental and deterministic modelling framework for evaluating 17 naturally occurring radioactive building materials collected in Katsina State, Nigeria. Activity concentrations of 226Ra, 232Th and 40K were measured using NaI(Tl) gamma-ray spectrometry and used as source-term inputs to a deterministic photon-transport model based on th

THP-1-derived polarized macrophages differ by drug transporter transcriptomics, P-glycoprotein activity and rifampicin uptake kinetics.

Inferior efficacy of antibiotics (e.g. rifampicin) in M2 macrophage-dominant tuberculosis granuloma might be related to poor drug uptake into these cells, potentially mediated by altered expression and activity of drug transporters such as P-glycoprotein (P-gp, encoded by ABCB1). Consequently, THP-1 cells were differentiated (200 nM phorbol 12-myristate 13-acetate; 72 h) and polarized to M1 (50 ng/mL lipopolysaccharide, 20 ng/mL interferon-gamma; 48 h) or M2 (20 ng/mL interleukin-4, interleukin-13; 48 h) macrophages. Comparative Interactome Profiling of Nonstructural Protein 3 Across SARS-CoV-2 Variants Emerged During the COVID-19 Pandemic.

SARS-CoV-2 virus and its variants remain a global health threat, due to their capacity for rapid evolution. Variants throughout the COVID-19 pandemic exhibited variations in virulence, impacting vaccine protection and disease severity. Investigating nonstructural protein variants is critical to understanding viral evolution and manipulation of host protein interactions. We focus on nonstructural protein 3 (nsp3), with multiple domains with different activities, including viral polyprotein cleavage, host deubiquitylation, de-ISGylation, and double-membrane vesicle formation. Nanospray Laser-Induced Plasma Ionization Mass Spectrometry for Analysis of Trace Short-Chain Chlorinated Paraffins in Single Cells.

Short-chain chlorinated paraffins (SCCPs) are a group of persistent organic pollutants that pose potential toxicity and health risks to biosystems and ecosystems. Analysis of trace SCCPs in single cells gives deeper insights into their cytotoxic, carcinogenic, and mutagenic investigations. Here a nanospray laser-induced plasma ionization mass spectrometry (nLIPI-MS) method was developed for accurate and sensitive analysis of trace SCCPs in single cells. An inner-wall molecularly imprinted polymer nanopipette was prepared, which allowed the exhaustive extraction of trace SCCPs from a single cel

Persistent metabolic toxicities following developmental exposure to hexafluoropropylene oxide trimer acid (HFPO-TA): Roles of peroxisome proliferator activated receptor gamma.

BACKGROUND: Hexafluoropropylene oxide trimer acid (HFPO-TA), a perfluorooctanoic acid (PFOA) substitute, exhibited strong affinity and capability to activate peroxisome proliferator activated receptor gamma (PPARγ), a lipid metabolism regulator, suggesting potential to induce metabolic toxicities. METHODS: Fertile chicken eggs were exposed to 0, 0.5, 1 or 2 mg/kg (egg weight) HFPO-TA and incubated until hatch. Serum from 0- and 3- month-old chickens were subjected to liquid chromatography ultra-high resolution mass spectrometry for HFPO-TA concentration, while liver, pancreas and adipose tiss

Cytomegalovirus Infection Triggers the Secretion of the PPARγ Agonists 15-Hydroxyeicosatetraenoic Acid (15-HETE) and 13-Hydroxyoctadecadienoic Acid (13-HODE) in Human Cytotrophoblasts and Placental Cultures.

INTRODUCTION: Congenital infection by human cytomegalovirus (HCMV) is a leading cause of congenital abnormalities of the central nervous system. Placenta infection by HCMV allows for viral spread to fetus and may result in intrauterine growth restriction, preeclampsia-like symptoms, or miscarriages. We previously reported that HCMV activates peroxisome proliferator-activated receptor gamma (PPARγ) for its own replication in cytotrophoblasts. Here, we investigated the molecular bases of PPARγ activation in infected cytotrophoblasts. RESULTS: We show that onboarded cPLA2 carried by HCMV particl

Congenital Bile Acid Synthesis Defect Type 2 Mimicking Biliary Atresia: A Case Report of Delta-4-3-Oxosteroid 5-Beta-Reductase Deficiency in a Two-Month-Old Infant.

Neonatal cholestasis presents a complex diagnostic challenge, frequently necessitating the differentiation of numerous etiologies, including the anatomically correctable biliary atresia (BA), from various metabolic and genetic disorders. The timely and accurate identification of inborn errors of bile acid metabolism is particularly critical, as these conditions can lead to progressive liver damage if left untreated. Congenital bile acid synthesis defects (CBASDs) are rare autosomal recessive inborn errors of metabolism that can manifest as neonatal cholestasis. Their clinical and biochemical p

Long-term behaviour of 137Cs in archived honey samples from a lowland mosaic landscape and a forest-influenced locality in southern Slovakia (1957-2001).

Honey and honeybee products are widely recognised as effective bioindicators of environmental contamination due to their ability to integrate pollutants over large foraging areas. In this study, archived honey samples were used to reconstruct the long-term dynamics of radiocaesium (¹³⁷Cs) in southern Slovakia. Archived honey samples originating from two contrasting environments in southern Slovakia were analysed retrospectively using high-resolution gamma spectrometry. The dataset comprised honey samples from a lowland locality (1958-2001, n = 35) and honey samples originating from a nearby fo

Effects of Gamma-Tocotrienol on Partial-Body Irradiation-Induced Intestinal Injury in a Nonhuman Primate Model.

Exposure to high doses of radiation, accidental or therapeutic, often results in gastrointestinal (GI) injury. To date, there are no therapies available to mitigate GI injury after radiation exposure. Gamma-tocotrienol (GT3) is a promising radioprotector under investigation in nonhuman primates (NHP). We have shown that GT3 has radioprotective function in intestinal epithelial and crypt cells in NHPs exposed to 12 Gy total-body irradiation (TBI). Here, we determined GT3 potential in accelerating the GI recovery in partial-body irradiated (PBI) NHPs using X-rays, sparing 5% bone marrow. Sixteen

Proteomic changes in female rat hippocampus following exposure to a terrified sound stress.

Stress plays a profound role in the onset of affective disorders, including an elevation in risk factors for depression and anxiety. Women are twice as vulnerable to stress as men because of greater sensitivity to a substance produced during times of anxiety. To better define the abnormal proteins implicated in cognitive deficits and other stress-induced dysfunction, female rats were exposed to terrified sound stress, and two-dimensional electrophoresis (2-DE) and matrix-assisted laser desorption ionization-time-of-flight mass spectrometry (MALDI-TOF MS) were utilized to determine the differen
