Anatomical localization of beta 1 integrin-like immunoreactivity in rat brain.

beta 1 Integrin-like immunoreactivity was localized in rat brain using a polyclonal antibody raised in rabbit against rat liver beta 1 integrin. One-dimensional immunoblotting of whole rat brain membranes indicated that this antiserum recognized a single molecular species at 116,000 M(r), indicative of rat beta 1 integrins. Specific staining of beta 1 integrin-like immunoreactivity was found in the vascular structures of the brain, including microvessels, the ventricular ependymal cells, and pia mater. The pineal gland stained densely, and diffuse staining was present throughout the gray matte

A simple efficient method of sequential G-banding and fluorescence in situ hybridization.

Several methods of chromosome identification to be used in combination with G-banding by the trypsin technique with Giemsa staining (G-banding) and fluorescence in situ hybridization (FISH) have been developed in the past few years. Unfortunately, these methods are impractical to use in the clinical laboratory and can provide inconsistent results. We report a simple, efficient, and reliable method of G-banding and FISH for use in clinical cytogenetics. G-banded chromosomes are codenatured with a direct- or indirect-labeled DNA prove (by using either chromosome painting or a chromosome centrome

SPIONs-labeled hUCMSCs for in vitro safety analysis and in vivo tracking in scarred monkey uteri.

Cesarean section has been increasingly used, stem cells have a potential to improve the repairing of scarred uteri, but its mechanism is still unclear. Cell labeling and tracking are becoming increasingly important in stem cell research. In this study, we evaluated the safety and efficacy of labeling human Umbilical Cord Mesenchymal Stem Cells (hUCMSCs) with Superparamagnetic Iron Oxide Nanoparticles (SPIONs), both in vitro and in vivo, with a particular focus on their application as Magnetic Resonance Imaging (MRI) contrast agents injected into the uterus. Monkeys were used as animal models b

Mammary Paget Disease With Melanocytic Proliferation Mimicking Malignant Melanoma in situ: A Case Report.

Several cases of pigmented mammary Paget's disease (PMPD) mimicking cutaneous malignant melanoma have been reported. In these cases, the tumor cells are colonized by melanocytes, particularly with the presence of a population of melanocytes staining for HMB-45 and S100. Here, we report a case of mammary Paget disease (MPD) which was misdiagnosed as melanoma in situ due to the interpretation of the staining of melanocytic markers S-100, Melan-A, and HMB-45. The tumor cells strongly expressed CK7 and GATA3, and a dual-labeling showed negative PHH3 labeling for the melanocytes. Pathologists need

Helicobacter pylori infection-induced H3Ser10 phosphorylation in stepwise gastric carcinogenesis and its clinical implications.

BACKGROUND: Our previous works have demonstrated that Helicobacter pylori (Hp) infection can alter histone H3 serine 10 phosphorylation status in gastric epithelial cells. However, whether Helicobacter pylori-induced histone H3 serine 10 phosphorylation participates in gastric carcinogenesis is unknown. We investigate the expression of histone H3 serine 10 phosphorylation in various stages of gastric disease and explore its clinical implication. MATERIALS AND METHODS: Stomach biopsy samples from 129 patients were collected and stained with histone H3 serine 10 phosphorylation, Ki67, and Helic

Curcumin labeling of neuronal fibrillar tau inclusions in human brain samples.

The study aimed to characterize curcumin (CCM) (fluorescent yellow curry pigment) labeling of neuronal fibrillar tau inclusions (FTIs) in representative cases of 3 main tauopathies: Alzheimer disease (AD), progressive supranuclear palsy, and Pick disease. After identification of FTIs in hematoxylin and eosin-stained brain sections, sequential labeling and signal colocalization image analysis were used to compare CCM with thioflavine S (ThS), monoclonal antibody AT8 immunofluorescence, and Gallyas silver staining by visualizing the same FTIs. Curcumin preference for specific tau isoforms was te

An affinity reagent-conjugated biotin ligase for amplified cell surface labelling in vitro and in vivo.

Targeting cells for ablation, gene therapy or drug delivery is an ongoing challenge in development of therapeutics. With the goal of efficiently labeling cell surfaces based on target recognition, we linked the well-established engineered ultraID biotin ligase to cell-surface affinity reagents to enable catalytic installation of tags on the surface of cells. Biotin ligase conjugated to nanobodies against cell surface proteins exhibited an order of magnitude increase in cell labeling compared to nanobody alone. Biotin labeling of cells remained efficient at low nanomolar concentrations of the c

Investigation of microbial contamination of commercial Romanowsky stain variant solutions used in veterinary clinics.

BACKGROUND: Diff-Quik (DQ) staining is widely used in clinical practice for its ease of use and rapid results. However, its potential contamination remains a concern. HYPOTHESIS/OBJECTIVES: This study aimed to investigate DQ solution contamination in veterinary hospitals and identify factors affecting it through a questionnaire on stain usage practices. MATERIALS AND METHODS: A survey was conducted to assess the use of DQ staining in local animal clinics. Samples of DQ solution II collected from each hospital participating in the survey were cultured and analysed microscopically for contamin

A refined Golgi-Cox method for the staining of embryonic neurons in the mouse brain.

The Golgi-Cox stain remains a valuable technique used to investigate the morphology of individual neurons. Despite this, Golgi-Cox staining protocols are predominantly designed to impregnate adult neurons. Protocols optimised for the staining of immature embryonic mouse neurons have been previously developed but have limitations, including being time-consuming and being reliant on the use of expensive commercial kits. Here, we present a simple and inexpensive method for Golgi-Cox staining of embryonic neurons in the mouse brain. We identified that a 60 minute, 4 % paraformaldehyde (PFA) brain

Comparison of special stains (Giemsa stain and Modified Toluidine Blue stain) with immunohistochemistry as gold standard for the detection of H. pylori in gastric biopsies.

BACKGROUND AND STUDY AIMS: Helicobacter pylori (H. pylori) has been extensively implicated in the etiology and pathogenesis of gastric ulcers and carcinomas, which has necessitated its efficient and cost-effective identification in gastric biopsy samples. We have sought to compare various staining methods, namely, Hematoxylin and Eosin (H&E), Giemsa, and Modified Toluidine Blue (MTB), with immunohistochemistry (IHC; gold standard) in terms of efficacy, staining costs, and duration of performing the stains in settings with limited resources. PATIENTS AND METHODS: Gastric biopsy specimens of 50

Characteristics of magnetic labeling on liver tumors with anti-alpha-fetoprotein-mediated Fe3O4 magnetic nanoparticles.

For preoperative and intraoperative detection of tumor distribution, numerous multimodal contrast agents, such as magnetic nanoparticles (MNPs) with several examination indicators, are currently in development. However, complex materials, configuration, and cost are required for multimodal contrast agents, accompanied by a high possibility of toxicity and low popularity in clinics. Nevertheless, the magnetic labeling of MNPs using bioprobes should be feasible not only in preoperative magnetic resonance imaging (MRI), but also in intraoperative examination based on other magnetic properties. Hierarchical evaluation of histology and p16-labeling can improve the risk assessment on cervical intraepithelial neoplasia progression.

OBJECTIVE: High-grade cervical lesions (HSIL) are associated with the presence of high-risk HPV types, tissue expression of p16, and increased chance of malignant progression, requiring surgical intervention. To improve risk evaluation, we assessed the discriminatory power of the histological findings associated with p16 immunohistochemistry (IHC) staining to classify the low-grade cervical lesion (LSIL) and HSIL. METHODS: We collected cervical biopsies from colposcopy-visible lesions and non-affected tissue (adjacent to the lesions) of 62 Brazilian women and labeled them with anti-p16 antibo

Labeling and imaging of neurons with a genetically encoded autonomous bioluminescence system.

The bacterial bioluminescence system, consisting of a luciferase and enzymes for intracellular luciferin synthesis, enables autonomous bioluminescence imaging of mammalian cells. Its continuous light emission without the need for exogenous substrate administration makes this system particularly well-suited for the long-term imaging of living cells. An additional feature is its dependence on metabolic energy, allowing the monitoring of cellular energy status and vitality. However, the system’s relatively low brightness and the requirement of six different genes complicate cellular labeling and
