Systemic Piezo1 activation improves cerebrovascular function in Alzheimer's disease.

INTRODUCTION: Cerebral blood flow (CBF) reductions and impaired functional hyperemia (FH) are early features of Alzheimer's disease (AD) that contribute to disease progression. Here, we investigated whether systemic activation of the mechanosensitive ion channel Piezo1 affects cerebrovascular deficits in an AD mouse model. METHODS: Using two-photon in vivo imaging and laser-speckle imaging, we determined CBF, capillary stalling, and FH in the 5xFAD model of AD. RESULTS: We observed increased capillary stalling and reduced CBF and FH to whisker stimulation in 5xFAD mice compared to wild-type

The effect of tactile stimulation on spatial memory and hippocampal neuronal density in male rats with sensory deprivation during a critical period.

It is well known that sensory information driven from whiskers serves as an example of tactile perception in rodents, and plays an important role in social behavior, environmental exploration, and decision-making processes, the influence of manipulations performed during the development of whiskers, on learning has been received little attention in the literature. This study aimed to evaluate the effect of tactile stimulation (TS) on spatial memory performance and neuronal density in the hippocampus during adulthood in early sensory-deprived rats. Wistar albino male rats were divided into four

Exploring the mammalian sensory space: co-operations and trade-offs among senses.

The evolution of a particular sensory organ is often discussed with no consideration of the roles played by other senses. Here, we treat mammalian vision, olfaction and hearing as an interconnected whole, a three-dimensional sensory space, evolving in response to ecological challenges. Until now, there has been no quantitative method for estimating how much a particular animal invests in its different senses. We propose an anatomical measure based on sensory organ sizes. Dimensions of functional importance are defined and measured, and normalized in relation to animal mass. For 119 taxonomical

A system for studying facial nerve function in rats through simultaneous bilateral monitoring of eyelid and whisker movements.

The occurrence of inappropriate co-contraction (synkinesis) of facially innervated muscles in humans is a common sequela of facial nerve injury and recovery. We have developed a system for studying facial nerve function and synkinesis in restrained rats using non-contact opto-electronic techniques that enable simultaneous bilateral monitoring of eyelid and whisker movements. Whisking is monitored in high spatio-temporal resolution using laser micrometers, and eyelid movements are detected using infrared diode and phototransistor pairs that respond to the increased reflection when the eyelids c

Nonlinear integration of sensory responses in the rat barrel cortex: an intracellular study in vivo.

To study integration of converging sensory inputs on single cortical neurons, we performed intracellular recordings in vivo in the barrel cortex of the barbiturate-anesthetized rat. We deflected the principal whisker (PW) for each cell either alone or preceded (at 20, 50, and 100 msec) by the deflection of a small number of remote whiskers (RWs) far from the PW. The synaptic responses to both the PW and the RW were similar qualitatively and consisted of excitation followed by inhibition that comprised an early and a late component. The RW response was of smaller amplitude and more often subthr

Whisker plucking alters responses of rat trigeminal ganglion neurons.

Whisker plucking in developing and adult rats provides a convenient method of temporarily altering tactile input for the purposes of studying experience-dependent plasticity in the somatosensory cortex. Yet, a comprehensive examination of the effect of whisker plucking on the response properties of whisker follicle-innervating trigeminal ganglion (NVg) neurons is lacking. We used extracellular single unit recordings to examine responses of NVg neurons to controlled whisker stimuli in three groups of animals: (1) rats whose whiskers were plucked from birth for 21 days; (2) rats whose whiskers w

Mesencephalic innervation of the vibrissal follicle-sinus complex in the mouse embryo.

Peripheral projections of neurones whose cell bodies lie in the mesencephalic nucleus of the fifth cranial nerve, situated between the central grey and mesencephalic reticular formation, were studied in mouse embryos aged between day 9 and 15 and in postnatal day 1 mice. Nonspecific neural antibody staining allowed visualisation of the developing cranial nerves, in particular the descending mesencephalic tract. This facilitated successful dissection of the descending mesencephalic tract and trigeminal ganglion in the heads of fresh mouse embryos and postnatal mice. The fluorescent dye, 1,1'-di

Stage-dependent cerebrocerebellar communication during sensorimotor processing.

Cerebral cortex and cerebellum are essential for sensorimotor control, but the dynamics of their interactions remain unclear. Here, we investigated which pathways prevail during preparation and execution of spontaneous whisker movements in mice. During preparation, neuronal activity of primary motor (M1) and somatosensory (S1) cortex precede that of cerebellar crus regions, with a lead that is consistent with relaying a copy of motor commands. After movement onset, the phase of the signal inverts, indicating a dominant vector signaling from cerebellum to cerebrum. At this stage, Purkinje cell

Brainstem local microglia induce whisker map plasticity in the thalamus after peripheral nerve injury.

Whisker deafferentation in mice disrupts topographic connectivity from the brainstem to the thalamic ventral posteromedial nucleus (VPM), which represents whisker map, by recruiting "ectopic" axons carrying non-whisker information in VPM. However, mechanisms inducing this plasticity remain largely unknown. Here, we show the role of region-specific microglia in the brainstem principal trigeminal nucleus (Pr5), a whisker sensory-recipient region, in VPM whisker map plasticity. Systemic or local manipulation of microglial activity reveals that microglia in Pr5, but not in VPM, are necessary and s

Anatomically and functionally distinct thalamocortical inputs to primary and secondary mouse whisker somatosensory cortices.

Subdivisions of mouse whisker somatosensory thalamus project to cortex in a region-specific and layer-specific manner. However, a clear anatomical dissection of these pathways and their functional properties during whisker sensation is lacking. Here, we use anterograde trans-synaptic viral vectors to identify three specific thalamic subpopulations based on their connectivity with brainstem. The principal trigeminal nucleus innervates ventral posterior medial thalamus, which conveys whisker-selective tactile information to layer 4 primary somatosensory cortex that is highly sensitive to self-in

Characterisation of progressive motor deficits in whisker movements in R6/2, Q175 and Hdh knock-in mouse models of Huntington's disease.

BACKGROUND: Motor dysfunction is a major component of the Huntington's disease (HD) phenotype, both in patients and animal models. Motor function in mice is usually measured using tests that involve a novel environment, or require a degree of learning, which creates potential confounds in animals, such as anxiety and/or learning. NEW METHOD: We propose that studying whisker control provides a more naturalistic way to measure motor function in HD mice. To this end we tested three strains of HD mice; R6/2 (CAG250), zQ175 and Hdh (CAG50, 150 and 250) mice. RESULTS: We discovered a clear and pro

Neurodegeneration in the somatosensory cortex after experimental diffuse brain injury.

Disruption and consequent reorganization of central nervous system circuits following traumatic brain injury may manifest as functional deficits and behavioral morbidities. We previously reported axotomy and neuronal atrophy in the ventral basal (VB) complex of the thalamus, without gross degeneration after experimental diffuse brain injury in adult rats. Pathology in VB coincided with the development of late-onset aberrant behavioral responses to whisker stimulation, which lead to the current hypothesis that neurodegeneration after experimental diffuse brain injury includes the primary somato

Bilateral projections from rat MI whisker cortex to the neostriatum, thalamus, and claustrum: forebrain circuits for modulating whisking behavior.

In rats, whisking behavior is characterized by high-frequency synchronous movements and other stereotyped patterns of bilateral coordination that are rarely seen in the bilateral movements of the limbs. This suggests that the motor systems controlling whisker and limb movements must have qualitative or quantitative differences in their interhemispheric connections. To test this hypothesis, anterograde tracing methods were used to characterize the bilateral distribution of projections from the whisker and forepaw regions in the primary motor (MI) cortex. Unilateral tracer injections in the MI w

T cell memory in the injured facial motor nucleus: relation to functional recovery following facial nerve crush.

T cells have the ability to mount a memory response to a previously encountered antigen such that re-exposure to the antigen results in a response that is greater in magnitude and function. Following facial nerve transection, T cells have been shown to traffic to injured motor neurons in the facial motor nucleus (FMN) and may have the ability to promote neuronal survival and functional recovery. Previously, we demonstrated that early exposure to neuronal injury on one side of the brain during young adulthood elicited a T cell response that was greater in magnitude following exposure to the sam

A novel method of head fixation for the study of rodent facial function.

The rodent vibrissial system offers an excellent model for the study of both sensory and motor function. It has been widely employed to gather data pertaining to sensory and motor function involving the 5th and 7th cranial nerves and the central nervous system. Existing methods of head fixation for precise measurements of ocular and vibrissial function involve exposing the cranium and applying dental cement from which two or more threaded rods emerge. This common approach is suboptimal, requiring a relatively complicated implantation procedure, and results in a large, chronic interface between

Immunohistochemical Evaluation of Rabies Virus Dissemination to Peripheral Tissues and Organs in Naturally Infected Striped Skunks (Mephitis mephitis) from Northern Colorado, USA.

We describe the diagnostic utility of peripheral tissues from naturally infected free-ranging rabid striped skunks (Mephitis mephitis) to explore enhanced rabies surveillance testing using alternative tissues to the brainstem. Fifty-two dead or moribund adult striped skunks were collected by the Larimer County Health Department (Colorado, USA) as part of routine public health rabies surveillance during November 2018-July 2019 throughout Larimer County. Skunk carcasses were submitted to the Colorado State University Diagnostic Laboratory for rabies diagnosis using the direct fluorescent antibod

Effect of Metformin on the Functional and Electrophysiological Recovery of Crush Injury-Induced Facial Nerve Paralysis in Diabetic Rats.

The impact of metformin on the rat facial nerve following crush injury has only occasionally been documented to date. The purpose of the current investigation was to use functional and electrophysiological evaluations to investigate the effects of metformin administration on recovery following crush injury to the rat facial nerve. The rats were randomly divided into four groups: the nonDM/PBS group (n = 4), the nonDM/metformin group (n = 4), the DM/PBS group (n = 4), and the DM/metformin group (n = 4). Diabetes was generated by an intraperitoneal injection of streptozotocin. Facial nerve paral
