[Circadian regulation of sleep-wake cycles and food anticipation].

The circadian clock is crucial for efficient physiological function and drives the temporal regulation of the sleep-wake state, metabolism, and behavior. The timing of food intake and the accompanying behavior are both controlled by the internal clock, which is located in the suprachiasmatic nucleus (SCN) of the anterior hypothalamus. The SCN is considered as the master clock because the circadian rhythms for most physiological and behavioral processes are terminated after SCN ablation. The molecular framework of circadian oscillations can be best studied in the SCN. A "core" set of circadian

Abnormal sleep/wake dynamics in orexin knockout mice.

STUDY OBJECTIVES: Narcolepsy with cataplexy is caused by a loss of orexin (hypocretin) signaling, but the physiologic mechanisms that result in poor maintenance of wakefulness and fragmented sleep remain unknown. Conventional scoring of sleep cannot reveal much about the process of transitioning between states or the variations within states. We developed an EEG spectral analysis technique to determine whether the state instability in a mouse model of narcolepsy reflects abnormal sleep or wake states, faster movements between states, or abnormal transitions between states. DESIGN: We analyzed…

Sleep-wake regulation and hypocretin-melatonin interaction in zebrafish.

In mammals, hypocretin/orexin (HCRT) neuropeptides are important sleep-wake regulators and HCRT deficiency causes narcolepsy. In addition to fragmented wakefulness, narcoleptic mammals also display sleep fragmentation, a less understood phenotype recapitulated in the zebrafish HCRT receptor mutant (hcrtr-/-). We therefore used zebrafish to study the potential mediators of HCRT-mediated sleep consolidation. Similar to mammals, zebrafish HCRT neurons express vesicular glutamate transporters indicating conservation of the excitatory phenotype. Visualization of the entire HCRT circuit in zebrafish…

Sleep-wake schedules in preschool children who snore.

PURPOSE: The aim of the study was to compare sleep-wake schedules between snoring and nonsnoring preschool age children. MATERIAL AND METHODS: Daytime and nighttime sleep duration, daytime and nighttime symptoms were assessed in 34 children at preschool age who snore (5.38+/-1.21 years) and in 66 age- and sex-matched nonsnorers (5.67+/-1.12 years). The snoring group consisted of children with obstructive sleep apnea (OSA) scores <+3.5 and >-1, the nonsnoring group with OSA score <-1. RESULTS: Children who snore differ from the nonsnorers in daytime sleep duration (51.62+/-28.9 minutes vs. 10

The effect of clomipramine on wake/sleep and orexinergic expression in rats.

We have previously found that neonatal treatment with clomipramine (CLI) induced a decrease in brain orexins during the juvenile period and that these changes were reversed at adulthood. This study investigated the effect of CLI on the orexinergic component and sleep/wake states. Two groups of adult male rats were conducted for 48-h polysomnographic recording. One group of rats was treated with CLI (20 mg/kg every 12 h), and a second group was treated with equivolume of saline (SAL) simultaneously after the first 24 h of polysomnographic recording. Rats were killed 2 h after the third dose of

Sleep-wake disturbances in sporadic Creutzfeldt-Jakob disease.

BACKGROUND: The prevalence and characteristics of sleep-wake disturbances in sporadic Creutzfeldt-Jakob disease (sCJD) are poorly understood. METHODS: Seven consecutive patients with definite sCJD underwent a systematic assessment of sleep-wake disturbances, including clinical history, video-polysomnography, and actigraphy. Extent and distribution of neurodegeneration was estimated by brain autopsy in six patients. Western blot analyses enabling classification and quantification of the protease-resistant isoform of the prion protein, PrPSc, in thalamus and occipital cortex was available in fo

Dopaminergic-adrenergic interactions in the wake promoting mechanism of modafinil.

Adrenergic signaling regulates the timing of sleep states and sleep state-dependent changes in muscle tone. Recent studies indicate a possible role for noradrenergic transmission in the wake-promoting action of modafinil, a widely used agent for the treatment of excessive sleepiness. We now report that noradrenergic projections from the locus coeruleus to the forebrain are not necessary for the wake-promoting action of modafinil. The efficacy of modafinil was maintained after treatment of C57BL/6 mice with N-(2-chloroethyl)-N-ethyl 2-bromobenzylamine (DSP-4), which eliminates all noradrenaline

Sleep and gastroesophageal reflux disease: a wake-up call.

The medical community has mostly ignored the relationship between sleep and gastroesophageal reflux (GER), although recent studies have begun to explore the connection and suggest possible treatments. These studies have found that the relationship is reciprocal; reflux influences sleep and sleep influences reflux. It has been found that nighttime GER can result in a number of complications, and more generally, lower the quality of life for its sufferers, establishing sleep-related GER as a more intolerable form of GER. Though some lifestyle modifications may help reduce the risk of nighttime r

[Aspects of medical expert assessment of sleep-waking disorders].

Knowledge gained in sleep medicine over the past 10 years has not yet been incorporated into laws for the severely handicapped and pension law. The impact of increased accident incidence of patients with hypersomnia has not yet been recognized in the guidelines for medical diseases in automobile traffic in governmental driving regulations. In preparation for implementation of sleep-wake disorders into medico-legal jurisdiction they must be introduced to the guidelines as a separate medical entity according to the International Classification of Sleep Disorders. Due to different prognoses sleep

The effects of regularizing sleep-wake schedules on daytime sleepiness.

The present study evaluated the differential effects of two manipulations of sleep-wake schedules on daily subjective ratings of daytime sleepiness of college undergraduate students. Two experimental conditions were compared: a sleep only group and a regularity group. Subjects in both conditions were given a lower limit for total sleep time (7.5 hours). Subjects in the regularity group received an additional instruction to keep a regular sleep schedule. The study was longitudinal and prospective. Following a baseline period (12 days), the experimental conditions were introduced. The experiment

[Sleep and wake-cycle as circadian rhythm (author's transl)].

Sleep-wake cycles are closely tied with the circadian period of other bodily functions. Optimal synchronization is essential for troublefree alterations of sleep and wake periods. The neurophysiologic control areas are in various of the brain are far apart from each other for which reason a great variety of possible causes for sleep difficulties do exist. It has been shown recently that spezific mono-aminergic and cholinergic systems in the brain stem do control sleep. A case study will show that it is possible to improve sleeping difficulties with the serotonin precursor L-Troyptophan.

Circadian photoreceptor CRYPTOCHROME promotes wakefulness under short winter-like days via a GABAergic circuitry.

A cardinal symptom of seasonal affective disorder (SAD, also known as winter depression) is hypersomnolence, while the cause of this 'winter … e degraded by light, thus rendering CRY to be functional within this GABAergic circuitry to enhance wakefulness only under short-day length. Taken together, we demonstrate a mechanism that specifically regulates wakefulness under short winter-like days, which may provide insights regarding the winter sleepiness in SAD.

GABA neurons in the sublaterodorsal tegmental nucleus suppress wakefulness in healthy and narcoleptic mice.

The sleep-wake cycle is generated by competing neural circuits that control the oscillation between wakefulness, rapid eye movement (REM) sleep, and non-REM (NREM) sleep. While the sublaterodorsal tegmental nucleus (SLD) is recognized for its role in REM sleep generation, the functional contribution of its GABAergic neurons (SLDGABA) to sleep-wake regulation remains poorly understood. Here, we found that SLDGABA neurons function as a suppressor of wakefulness in both healthy (i.e., orexin+/+) and narcoleptic (i.e., orexin-/-) mice. In healthy mice, optogenetic silencing of SLDGABA neurons rapi…
