The prospect of continued manned space missions warrants an in-depth understanding of how prolonged microgravity affects the human brain. Functional magnetic resonance imaging (fMRI) can pinpoint changes reflecting adaptive neuroplasticity across time. We acquired resting-state fMRI data of cosmonauts before, shortly after, and eight months after spaceflight as a follow-up to assess global connectivity changes over time. Our results show persisting connectivity decreases in posterior cingulate cortex and thalamus and persisting increases in the right angular gyrus. Connectivity in the bilatera

Long-duration spaceflight is associated with neurologic and ophthalmic clinical and imaging findings in astronauts termed spaceflight associated neuro-ocular syndrome (SANS). These microgravity-induced findings have been well documented by the National Aeronautics and Space Administration (NASA) and are clearly a potential risk for future human space exploration. The underlying pathogenesis of SANS is not well understood, although multiple hypotheses have emerged. Terrestrial analogues and potential countermeasures have also been studied to further understand and potentially mitigate SANS. In

During space exploration, the human body is subjected to altered atmospheric environments and gravity, exposure to radiation, sleep disturbance, and mental pressures; all these factors are responsible for cardiovascular diseases. Under microgravity, the physiological changes related to cardiovascular diseases are the cephalic fluid shift, dramatic reduction in central venous pressure, changes in blood rheology and endothelial function, cerebrovascular abnormalities, headaches, optic disc edema, intracranial hypertension, congestion of the jugular vein, facial swelling, and loss of taste. Gener

INTRODUCTION: During spaceflight, it is important to consider the mechanisms by which surgeries and medical procedures can be safely and efficiently conducted. Instruments used to carry out these processes need to be sterilized. Thus, we have designed and tested a three-dimensional-printed (3D-printed) portable sterilizer that implements far ultraviolet-C (Far UV-C) light radiation to disinfect bacteria and microorganisms from surgical instruments.METHODS: The sterilizer was 3D-printed with polylactic acid filament. Effectiveness was assessed through three trials at differing times of steriliz

The sensitivity of human immune system cells to gravity changes has been investigated in numerous studies. Human macrophages mediate innate and thus rapid immune defense on the one hand and activate T- and B-cell-based adaptive immune response on the other hand. In this process they finally act as immunoeffector cells, and are essential for tissue regeneration and remodeling. Recently, we demonstrated in the human Jurkat T cell line that genes are differentially regulated in cluster structures under altered gravity. In order to study an in vivo near system of immunologically relevant human cel

Long-duration exposure to weightlessness results in bone demineralization, muscle atrophy, cardiovascular deconditioning, altered sensory-motor control, and central nervous system reorganizations. Exercise countermeasures and body loading methods so far employed have failed to prevent these changes. A human mission to Mars might last 2 or 3 years and without effective countermeasures could result in dangerous levels of bone and muscle loss. Artificial gravity generated by rotation of an entire space vehicle or of an inner chamber could be used to prevent structural changes. Some of the physica…

Spaceflight presents unique gravitational, radiation, and isolation hazards for human exploration of the Moon, Mars, and beyond, yet its impact on the female reproductive system and successive generations has been largely unassessed. In the NASA Rodent Research 20 mission, we examined the impact of a 42-d spaceflight on the female reproductive axis including ovulatory capacity, implantation rate, and fecundity as well as behavioral, metabolic, and functional outcomes in F1 and F2 offspring. Females bred 5 d after return to Earth became pregnant but only exhibited a slight decline in fecundity

Microbiome-integrated Caenorhabditis elegans cultivation methods enable investigation of host-microbiome interactions in the context of space-relevant stresses using three key innovations: introduction of live bacterial communities replacing chemically defined media, implementation of auxin-inducible degradation systems to prevent progeny production, and development of complementary hardware platforms. Polyethylene bags provide gas-permeable cultivation environments for large populations with complex microbiomes supporting downstream molecular analyses, while NemaCapsules with micropillar arra

PURPOSE OF REVIEW: This review synthesizes evidence from diverse experimental platforms and proposes the Osteocyte Mechanobiology Unit (OMU), a conceptual framework integrating the mechanical and biochemical processes underlying microgravity-induced alterations in osteocytes and their surrounding microenvironment.
RECENT FINDINGS: Current evidence indicates that microgravity disrupts two interconnected OMU domains: the Mechanical Sensing Domain, through alterations in the local mechanical microenvironment and mechanosensory structures, and the Biochemical Signaling Domain, through dysregulate

This chapter has the aim to propose a theoretical framework for describing the qualitative changes in the deformation behavior of isolated human breast cancer cells under varying gravitational conditions. We describe interdisciplinary principles of statistical thermodynamics and nonlinear dynamics of eukaryotic cells. The framework aims to capture how microgravity-induced alterations in cytoskeletal organization lead to distinct mechanobiological phenotypes.

OBJECTIVE: To synthesize evidence on urinary calcium and oxalate in astronauts, comparing short- and long-duration missions and temporal patterns across mission phases.
METHODS: Studies involving astronauts that assessed microgravity as the primary exposure and collected urine specimens were included. PubMed and EMBASE were searched using Medical Subject Headings and Emtree terms. Results were synthesized narratively, with quantitative data summarized using descriptive statistics and medians with interquartile ranges where appropriate.
RESULTS: Thirty studies met the inclusion criteria, span

Crewed interplanetary return missions that are on the planning horizon will take years, more than enough time for initiation and completion of a pregnancy. Pregnancy is viewed as a sequence of processes - fertilization, blastocyst formation, implantation, gastrulation, placentation, organogenesis, gross morphogenesis, birth and neonatal development - each of which needs to be completed successfully, and each of which has a probability of success. The effects of the environment of interplanetary flight - microgravity and galactic cosmic rays (GCR) - on these probabilities are inferred from Eart

The future of space exploration will include a prolonged presence on the Moon, commercial low-orbit spaceflight, and long-range missions to Deep Space, including a prolonged Martian presence. Understanding the effects the space environment will have on astronauts' musculoskeletal system is mission critical and include both microgravity and space radiation. In spaceflight, crewmembers are exposed to a vast mixture of radiation species and energies including cosmic rays (CR) from galactic cosmic radiation (GCR), solar ejections, and neutrons. Any trip beyond the protection of the Earth's electro

Measurable changes in electrophysiology have been documented in spaceflight, creating a pathway for disease genesis and progression in astronauts. These electrophysiology changes can be measured using potential difference (PD). A probe to measure PD was developed and is used clinically on Earth; this probe relies on fluid perfusion to establish an electrical connection to make PD measurements. The changes to fluid behavior in microgravity and partial gravity (including lunar and Martian gravity) drives the need to test this probe in a spaceflight environment. Here, we test the PD probe in a no

Over the past 15 years, there has been a significant shift in biomedical research toward a major focus on stem cell research. Although stem cells and their derivatives exhibit potential in modeling and mitigating human diseases, the ongoing objective is to enhance their utilization and translational potential. Stem cells are increasingly employed in both academic and commercial settings for a variety of in vitro and in vivo applications in regenerative medicine. Notably, accessibility to stem cell research in low-Earth orbit (LEO) has expanded, driven by the unique properties of space, such as
