Characterization of Bacterial and Fungal Microbiome in Children with Hirschsprung Disease with and without a History of Enterocolitis: A Multicenter Study.

Development of potentially life-threatening enterocolitis is the most frequent complication in children with Hirschsprung disease (HSCR), even after definitive corrective surgery. Intestinal microbiota likely contribute to the etiology of enterocolitis, so the aim of this study was to compare the fecal bacterial and fungal communities of children who developed Hirschsprung-associated enterocolitis (HAEC) with HSCR patients who had never had enterocolitis. Eighteen Hirschsprung patients who had completed definitive surgery were enrolled: 9 had a history of HAEC and 9 did not. Fecal DNA was isol

Nickel quercetinase, a "promiscuous" metalloenzyme: metal incorporation and metal ligand substitution studies.

BACKGROUND: Quercetinases are metal-dependent dioxygenases of the cupin superfamily. While fungal quercetinases are copper proteins, recombinant Streptomyces quercetinase (QueD) was previously described to be capable of incorporating Ni(2+) and some other divalent metal ions. This raises the questions of which factors determine metal selection, and which metal ion is physiologically relevant. RESULTS: Metal occupancies of heterologously produced QueD proteins followed the order Ni > Co > Fe > Mn. Iron, in contrast to the other metals, does not support catalytic activity. QueD isolated from th

The case for antifungal stewardship.

PURPOSE OF REVIEW: Antimicrobial stewardship (AMS) has overwhelmingly focussed on antibiotics while antifungal agents have been largely neglected despite the few published audits of antifungal drug use demonstrating clear deficiencies in prescribing behaviour. In this review, we outline not only the elements of antifungal stewardship (AFS) in common with AMS but also features specific to antifungal drugs, combined with insights from our experience in AFS. RECENT FINDINGS: Invasive fungal diseases (IFDs) have a lower institutional incidence relative to infections caused by multiresistant bacte

Amino acid-derived 1,2-benzisothiazolinone derivatives as novel small-molecule antifungal inhibitors: identification of potential genetic targets.

We have identified four synthetic compounds (DFD-VI-15, BD-I-186, DFD-V-49, and DFD-V-66) from an amino acid-derived 1,2-benzisothiazolinone (BZT) scaffold that have reasonable MIC(50) values against a panel of fungal pathogens. These compounds have no structural similarity to existing antifungal drugs. Three of the four compounds have fungicidal activity against Candida spp., Cryptococcus neoformans, and several dermatophytes, while one is fungicidal to Aspergillus fumigatus. The kill rates of our compounds are equal to those in clinical usage. The BZT compounds remain active against azole-,

Imaging aspects and non-surgical interventional treatment in human alveolar echinococcosis.

Alveolar echinococcosis (AE) of the liver caused by the metacestode of the fox tapeworm Echinococcus multilocularis is characterized by a multivesicular structure surrounded by an extensive fibro-inflammatory host reaction. The lesions behave like a slow-growing liver cancer, without sharp limits between the parasitic tissue and the liver parenchyma. Invasion of biliary and vascular walls is another hallmark of this severe disease. Moreover, the poor vascularization of the parasitic mass often leads to necrosis in the central part of the lesion. This explains why liver abscess due to superimpo

Design, Synthesis, Antimicrobial Evaluation, DFT Analysis, Molecular Docking and Molecular Dynamics Studies of Thiadiazol-Hydrazine Derivatives as Potential Dihydrofolate Reductase Inhibitors.

A series of thiadiazol-hydrazine derivatives (3a-3d) were rationally designed, synthesized, and characterized to explore their antimicrobial potential and possible interactions with dihydrofolate reductase (DHFR). The antimicrobial activity of the synthesized compounds was evaluated against representative Gram-positive bacteria (S. aureus and S. pyogenes), Gram-negative bacteria (E. coli and P. aeruginosa), and fungal strains (C. albicans, A. niger, and A. clavatus) using the minimum inhibitory concentration (MIC) method. Among the synthesized derivatives, compound 3b exhibited the most favora

Animal-Derived Antimicrobial Peptides: Emerging Therapeutic Strategies against Multidrug-Resistant Pathogens.

The global escalation of antimicrobial resistance (AMR) has outpaced the conventional antibiotic discovery pipeline, with bacterial AMR associated with an estimated 4.71 million deaths in 2021 and forecast to contribute to rising mortality through 2050. The World Health Organization's 2024 Bacterial Priority Pathogens List continues to identify carbapenem-resistant Gram-negative organisms and rifampicin-resistant Mycobacterium tuberculosis as the most urgent targets for new therapeutics. Antimicrobial peptides (AMPs), ancient and evolutionarily conserved effectors of innate immunity, are one o

Establishing Quantifiable Guidelines for Antimicrobial α/β-Peptide Design: A Partial Least-Squares Approach to Improve Antimicrobial Activity and Reduce Mammalian Cell Toxicity.

Antimicrobial peptides (AMPs) are promising candidates to combat pathogens that are resistant to conventional antimicrobial drugs because they operate through mechanisms that involve membrane disruption. However, the use of AMPs in clinical settings has been limited, at least in part, by their susceptibility to proteolytic degradation and their lack of selectivity toward pathogenic microbes vs mammalian cells. We recently reported on the design of α- and β-peptide oligomers structurally templated upon the naturally occurring α-helical AMP aurein 1.2. These α/β-peptide oligomers are more proteo

A Novel Tricyclic Polyketide, Vanitaracin A, Specifically Inhibits the Entry of Hepatitis B and D Viruses by Targeting Sodium Taurocholate Cotransporting Polypeptide.

UNLABELLED: Anti-hepatitis B virus (HBV) drugs are currently limited to nucleos(t)ide analogs (NAs) and interferons. A challenge of drug development is the identification of small molecules that suppress HBV infection from new chemical sources. Here, from a fungus-derived secondary metabolite library, we identify a structurally novel tricyclic polyketide, named vanitaracin A, which specifically inhibits HBV infection. Vanitaracin A inhibited the viral entry process with a submicromolar 50% inhibitory concentration (IC50) (IC50 = 0.61 ± 0.23 μM), without evident cytotoxicity (50% cytotoxic conc

Enhancing chemical and aroma profiles of Fu brick tea via Bacillus licheniformis-mediated enhanced fermentation: evidence from targeted chemistry, volatilomics, and metagenomics.

Fu brick tea, a typical post fermented tea, derives its quality characteristics largely from microbial metabolism during the golden flower formation stage. To explore the application potential of functional microorganisms in Fu brick tea fermentation, this study systematically evaluated the effects of enhanced fermentation with Bacillus licheniformis across three substrates, including dark tea, golden flower loose tea, and Fu brick tea, with a particular focus on chemical composition, volatile flavor compounds, and sensory quality. The results demonstrated that B. licheniformis could stably co

Characterization of the atmospheric microbiome in a semi-rural area of Central Europe using flow cytometry.

Characterizing bioaerosols is important for understanding their potential impacts on the environment and public health. In this study, we developed a novel flow cytometry-based approach to determine the low nucleic acid (LNA), high nucleic acid (HNA), dead, and intact bioaerosol populations in samples collected with a wet cyclone at Payerne, Switzerland, during spring and summer 2024. We found that the average bioaerosol number concentration reached (2.47 ± 3.35)×104 m-3. The HNA and intact populations were the most abundant populations, representing the largest fraction of total bioaerosols w

Effect of the degree of blockiness on the glycosylation and emulsification of citrus pectin-whey protein isolate conjugates.

BACKGROUND: Maillard reaction-derived pectin-protein conjugates are common functional food emulsifiers. As a key structural parameter of citrus pectin (CP), the blockiness degree (DB) independently regulates the Maillard reaction between CP and whey protein isolate (WPI) and the emulsifying properties of their conjugates, which remains poorly understood. This study aimed to clarify such regulatory mechanisms to support the targeted development of high-performance pectin-protein emulsifiers. METHOD: CP samples with similar methoxylation degrees (34.2-36.7%) and varied DB (27.4-88.3%) were prep

Species-specific patterns in fine-root traits and their coordination in the multidimensional root economics space at the Mongolian forest-steppe ecotone.

In the bioclimatically sensitive forest-steppe ecotone of northern Mongolia, belowground strategies can be elucidated using an integrated approach that considers root functional traits and symbiotic associations. We elucidated how three dominant tree species, namely Larix sibirica, Pinus sylvestris, and Betula platyphylla, coordinate their morphological, chemical, and symbiotic (ectomycorrhizal [EM]) traits within a multidimensional root economics space (RES) to adapt to this harsh ecotone. Although the RES framework generally associates thicker roots with greater reliance on fungi, EM coloniz
