Effects of in utero alcohol exposure on B cell development in neonatal spleen and bone marrow.

The effects of in utero alcohol exposure on neonatal lymphopoiesis were examined in a murine model of fetal alcohol syndrome. At birth, both immature and mature B cells were decreased in the spleens of neonatal animals and these subpopulations of B cells did not recover to normal levels until 3-4 weeks of life. Pre-B cells and total B cells were decreased as well in the bone marrow of ethanol-exposed animals. By 3-4 weeks of life, the number of B cells in the bone marrow recovered to normal levels, but the pre-B cells remained below normal levels through 5 weeks of age. Furthermore, a recently

Brain perfusion SPECT and MRI in foetal alcohol syndrome.

Six boys and five girls with a mean age of 8.6 (range 3 to 13) years with foetal alcohol syndrome (FAS) were studied by MRI and single photon emission computed tomography (SPECT) to find specific areas of vulnerability. Morphological anomalies shown in six of 11 patients by MRI were situated both cortically and subcortically: cortical atrophy (N = 2), dilated ventricle (N = 1), corpus callosum hypoplasia (N = 1), cerebellar atrophy (N = 2), one of the latter with Arnold-Chiari malformation (N = 1). Delayed myelination of the white matter was seen in two patients. Volumetric studies of the hipp

Ethanol-induced neural crest apoptosis is coincident with their endogenous death, but is mechanistically distinct.

The ability of both acute and chronic ethanol exposures to elicit cell death within specific embryonic and adult tissues is believed to partly underlie ethanol's pathogenicity; however, the mechanism underlying this cell death is unknown. This study partially characterized the mechanism of ethanol-induced neural crest cell death in a chick embryo model of fetal alcohol syndrome. In situ DNA end-labeling demonstrated this cell death was apoptotic and occurred at embryonic ethanol levels as low as 42 mM. Regardless of the initial exposure time, this apoptosis always appeared at a distinct develo

Blackcurrant seed oil, zinc, and fetal alcohol syndrome.

The effects of dietary supplementation by blackcurrant seed oil (BSO) and zinc sulphate on PGI2 gastric release and on serum zinc levels were investigated in 50 pregnant Sprague-Dawley rats which received watery alcohol solutions from 5% to 30% (v/v) as well as in their newborns. Blood zinc was assayed by atomic absorption spectroscopy; PGI2 was evaluated by platelet aggregation PGI2-dependent test. Dietary supplementation by BSO increased gastric release of PGI2 both in the mothers and their newborns; blood zinc levels were significantly (p < 0.01) lower in both vs control. The content of PGI

Effect of surrogate fostering on splenic lymphocytes in fetal ethanol exposed rats.

This study evaluated the effects of surrogate fostering as a procedure to control for postnatal effects of ethanol on the maternal female that may indirectly affect the offspring. Effects of fostering on the development of splenic lymphocytes, as well as possible differential effects of fostering on female and male offspring were examined. Litters from prenatal ethanol exposed (E), pair-fed (PF), and ad libitum-fed control (C) conditions were fostered at birth to surrogate untreated dams who had given birth within the same 12-hr period, or were reared by their biological mothers. At 15 and 60

Magnetic resonance imaging of brain anomalies in fetal alcohol syndrome.

OBJECTIVE: Postmortem studies of fetuses, infants, and young children with fetal alcohol syndrome (FAS) have demonstrated a variety of severe central nervous system (CNS) anomalies. We undertook this magnetic resonance study (1) to assess the spectrum of CNS anomalies that occur in a clinical sample of typical patients with FAS who are medically stable; and (2) to examine the relationship between CNS and facial anomalies. METHODOLOGY: Magnetic resonance imaging was performed on a series of 10 patients (4 children, 3 adolescents, and 3 adults) who met criteria for FAS. We systematically evalua

Morphometric analysis of the postnatal mouse optic nerve following prenatal exposure to alcohol.

Pregnant female mice were divided on day 12 post coitum into a control and an experimental group. The experimental group was given a single intraperitoneal dose of 0.015 ml/g body weight of 25% solution of alcohol in distilled water while the control group was exposed to a similar weight related dose of normal saline. The optic nerves were isolated from the offspring of both control and experimental groups at wk 2, 3 and 5 (i.e. during the juvenile period of postnatal development) and analysed by light and electron microscopy. Although in both groups the optic nerve grew in size rapidly during

A case definition and photographic screening tool for the facial phenotype of fetal alcohol syndrome.

OBJECTIVES: The purpose of this study was to demonstrate that a quantitative, multivariate case definition of the fetal alcohol syndrome (FAS) facial phenotype could be derived from photographs of individuals with FAS and to demonstrate how this case definition and photographic approach could be used to develop efficient, accurate, and precise screening tools, diagnostic aids, and possibly surveillance tools. STUDY DESIGN: Frontal facial photographs of 42 subjects (from birth to 27 years of age) with FAS were matched to 84 subjects without FAS. The study population was randomly divided in hal

Multifaceted alterations of the thalamo-cortico-thalamic loop in adult rats prenatally exposed to ethanol.

The thalamo-cortico-thalamic loop was investigated in adult rats exposed to ethanol during the last week of fetal life. Animals underwent either cortical or thalamic injections of lectin-conjugated horseradish peroxidase. Results demonstrate that prenatal exposure to ethanol causes permanent changes in the thalamocortical circuits. Alterations of thalamo-cortical and cortico-thalamic projections are concentrated at the level of axon terminal fields. The most severe thalamic damage is observed in the anterior intralaminar and midline nuclei; crossed cortico-thalamic projections also appear to b

Use of international classification of diseases coding to identify fetal alcohol syndrome--Indian Health Service facilities, 1981-1992.

Fetal alcohol syndrome (FAS) is one of the leading causes of preventable birth defects and developmental disabilities in the United States (1). Since 1979, surveillance systems for estimating and tracking FAS have categorized cases using International Classification of Diseases, Ninth Revision, Clinical Modification (ICD-9-CM), code 760.71 (2). This code comprises noxious influences affecting the fetus or newborn through placenta or breast milk, specifically alcohol, and includes FAS. Because the code is not specific for FAS and may reflect maternal alcohol consumption during pregnancy or othe

Mental development of 2-year-old children exposed to alcohol in utero.

In a prospective follow-up study, 60 children exposed to alcohol in utero were assessed by a psychologist (Bayley Mental scale) and a speech therapist (Reynell Verbal Comprehension scale) at a mean age of 27 months. Many mothers had been able to reduce their alcohol consumption during pregnancy, so the children could be divided into those exposed to heavy drinking during the first trimester only (group 1, n = 20), those exposed during the first and second trimesters (group 2, n = 20), and those exposed throughout pregnancy (group 3, n = 20). Forty-eight nonexposed children were examined to set

Ethanol inhibits C6 cell growth: fetal alcohol syndrome model.

Maternal consumption of ethanol produces a pattern of malformations, including nervous system abnormalities, in the developing fetus, a state called Fetal Alcohol Syndrome. We report the dose-dependent inhibition by ethanol of the growth of a glioma derived cell line, C6 cells; the effects occur at ethanol concentrations commonly encountered in the blood during human intoxication. The effects occur with different morphological subtypes of the cell line and do not occur when the cells are exposed to iso-osmolar concentrations of other chemicals. The results demonstrate that C6 cells are a model

Diminished proliferative response of con A-blast cells to interleukin 2 in adult rats exposed to ethanol in utero.

The fetal alcohol syndrome is associated with altered immunity. We attempted to delineate the mechanism of the decline in cell-mediated immunity observed by others by using rats which were exposed to alcohol in utero and tested for immune integrity 3 months after birth. We found that concanavalin A-stimulated T-lymphoblast (Con A T-blast) cells that were obtained from ethanol-exposed rats had significantly diminished proliferative responses to both crude and recombinant interleukin 2 compared to those obtained from normal and nutritional controls. The blunted response of Con A T-blast cells to

Pathological changes in peripheral nerves in experimental fetal alcohol syndrome.

The peripheral nerve pathology is described in experimental fetal alcohol syndrome. The marked retraction and shrinkage of axons with vacuolar spaces in the periaxonal area are the striking features. The direct toxicity of ethanol or its metabolites on the nerve fibers is considered to be the cause for pathological changes. Additional biochemical factors, pyridoxal phosphate dependent enzyme-depletion in the genesis of nerve damage, is proposed.

Development of the central nervous system functions in rat pups prenatally exposed to alcohol (study on the behavioural teratology of ethanol in CFY rat pups).

As a model of fetal alcohol syndrome (FAS) the rate of the maturation of the functions of the central nervous system was studied in the offspring of pregnant CFY rats receiving (from the 7th-15th day of gestation) either oral ethanol treatment or liquid diet containing ethanol. Both types of exposure caused numerous behavioural impairments, besides high perinatal mortality also the opening of the eyes and ears, and the appearance of postural reflexes were delayed. The newborn rats could be characterized by hyperactivity and weak motor coordination. The learning capacity, the avoidance conditio

Ethanol-induced growth inhibition: the role of cyclic AMP-dependent protein kinase.

Growth retardation is the single most common deficit observed in infants exposed to ethanol in utero, and the molecular mechanisms responsible for this growth inhibition are a focus of ongoing research. Several lines of research have suggested that ethanol-induced changes in the adenylate cyclase-protein kinase cascade may be involved in this process. Using an embryonic chick model, it was demonstrated that ethanol exposure during early development caused increases in cellular prostaglandin E2 and cyclic AMP levels that were inversely correlated with brain weight. Paradoxically, basal adenylat

Synaptogenesis in the hippocampal dentate gyrus: effects of in utero ethanol exposure.

In animal models of Fetal Alcohol Syndrome, ethanol causes a number of changes in brain development, with many of these changes being very transient. This is especially true for the process of synaptogenesis in different brain areas. Our quantitative electron microscopic study of synaptogenesis in the molecular layer of the rat dentate gyrus supports the above statement, by demonstrating that ethanol has no effect on the appearance of synapses in the dentate gyrus during early postnatal life (10-30 days old). However, prenatal ethanol exposure does appear to affect the process of synapse turno
