APOE and cortical superficial siderosis in CAA: Meta-analysis and potential mechanisms.

OBJECTIVE: To assess potential mechanisms of cortical superficial siderosis (cSS), a central MRI biomarker in cerebral amyloid angiopathy (CAA), we performed a collaborative meta-analysis of APOE associations with cSS presence and severity. METHODS: We pooled data from published studies reporting APOE genotype and MRI assessment of cSS in 3 distinct settings: (1) stroke clinic patients with symptomatic CAA (i.e., lobar intracerebral hemorrhage, transient focal neurologic episodes) according to the Boston criteria; (2) memory clinic patients; and (3) population-based studies. We compared cSS p

Hypothesis concerning embodied calendars: A case study of number form, color spreading, and taste-color synaesthesia.

We propose a hypothesis concerning the neural basis of the mental 'calendar' we all carry around in our brains, based on observations we made on a 25year old 'projector synaesthete', EA, who displays some novel and instructive features. In addition to her grapheme-color synaesthesia, she has a circular 'calendar line', laid out vividly in front of her in the horizontal plane with December 31st passing through the middle of her chest and other months arranged in clockwise sequence ending with December on her right (July was 3 feet in front of her). Her access to episodic memories felt, subjecti

Differences in regional brain activation patterns assessed by functional magnetic resonance imaging in patients with systemic lupus erythematosus stratified by disease duration.

The mediators of tissue damage in systemic lupus erythematosus (SLE) such as antibodies, cytokines and activated immune cells have direct access to most organs in the body but must penetrate the blood-brain barrier (BBB) to gain access to brain tissue. We hypothesized that compromise of the BBB occurs episodically such that the brain will acquire tissue damage slowly and not at the same rate as other organs. On the basis of these assumptions, we wished to determine if duration of disease correlated with brain injury, as measured with functional magnetic resonance imaging (fMRI), and if this wa

The relationship of bipolar disorder lifetime duration and vascular burden to cognition in older adults.

OBJECTIVES: We describe the cognitive function of older adults presenting with bipolar disorder (BD) and mania and examine whether longer lifetime duration of BD is associated with greater cognitive dysfunction. We also examine whether there are negative, synergistic effects between lifetime duration of BD and vascular disease burden on cognition. METHODS: A total of 87 nondemented individuals with bipolar I disorder, age 60 years and older, experiencing manic, hypomanic, or mixed episodes, were assessed with the Dementia Rating Scale (DRS) and the Framingham Stroke Risk Profile (FSRP) as a m

The role of intracranial pressure variability as a predictor of intracranial hypertension and mortality in critically ill patients.

OBJECTIVE: Intracranial pressure (ICP) monitoring is a widely utilized and essential tool for tracking neurosurgical patients, but there are limitations to the use of a solely ICP-based paradigm for guiding management. It has been suggested that ICP variability (ICPV), in addition to mean ICP, may be a useful predictor of neurological outcomes, as it represents an indirect measure of intact cerebral pressure autoregulation. However, the current literature regarding the applicability of ICPV shows conflicting associations between ICPV and mortality. Thus, the authors aimed to investigate the ef

Exploring Associations Between Early Cognitive Impairment and Echocardiographic Markers in Middle-Aged Patients with Atrial Fibrillation and Cardiometabolic Comorbidities: A Pilot Study.

Objectives: Atrial fibrillation (AF), the most common sustained cardiac arrhythmia, and cardiometabolic comorbidity, have been increasingly associated with cognitive impairment and dementia. These associations, however, remain underexplored and underappreciated in middle-aged individuals with AF. This study aimed to explore the associations of early cognitive impairment with the presence of cardiometabolic comorbidities and potential associations with echocardiographic markers in middle-aged patients with and without AF. Methods: Between 2023-2024, fifty-six consecutive outpatients with a diag

A depression-like phenotype is associated with discrete defects in the primary hippocampal circuit.

Major depressive disorder is known to disturb the hippocampus, but how this impacts signal processing performed by the structure remains poorly understood. Here, we report that single housing (7-10 days) promotes a depression-like phenotype in young adult mice that is associated with a robust, yet surprisingly discreet defect in information flow across the primary hippocampal circuit. In addition to sociability disturbances and despair-like behavior, single housing eliminated preference for novelty and impaired episodic memory encoding. Additionally, the lateral habenula, an epithalamic struct

The 'active reciprocity' ecosystem: senior cohousing as a determinant of social support and cognitive vitality independent of socioeconomic status.

INTRODUCTION: This study aims to explore the relationship between residential context and health-related outcomes. It seeks to analyze the differences in sociodemographic profiles, health status, psychosocial well-being, and cognitive performance between individuals living in self-managed senior collaborative housing (senior cohousing) and those in conventional single-family housing. METHODS: Using a cross-sectional comparative design, 171 older people (83 in cohousing, 88 in single-family homes) were assessed using a multidimensional battery that included sociodemographic data, objective and

Efficacy of theta binaural beat therapy on pain, cognition and anxiety in adults: A systematic review and meta-analysis of randomized controlled trials.

BACKGROUND: Binaural beat stimulation is a non-pharmacological neuromodulatory approach with potential to influence pain, emotions and cognition. Binaural beats are auditory illusion generated when two tones of slightly different frequencies are presented simultaneously to each ear. While prior reviews have pooled heterogeneous frequency bands or outcomes, the specific therapeutic relevance of theta-frequency (4-8 Hz) binaural beats remains unclear. OBJECTIVE: To systematically evaluate and quantitatively synthesize evidence on the efficacy of theta binaural beat therapy on pain, anxiety and

Repeating Task-Irrelevant Contexts Does Not Bias Task Choice and Performance in Voluntary Task Switching.

Goal-directed behaviour requires both efficient task execution as well as adequate task choice. Task choice may reflect deliberate evaluation of costs and benefits, but it may also be biased by bottom-up memory retrieval. Specifically, re-encountering a previously experienced context could retrieve a prior task choice, thereby influencing subsequent decisions. In the present study, using the voluntary task-switching (VTS) paradigm, we examined whether repeating a context would retrieve a task by manipulating whether a task-irrelevant context stimulus repeated or changed across consecutive tria

Task-dependent changes in effective connectivity from limbic to cognitive networks during motor imagery of freezing of gait in Parkinson's disease.

Freezing of gait (FOG) is a debilitating symptom of Parkinson's disease (PD) that has been related to abnormal functional connectivity across motor, cognitive, and limbic networks. However, it remains unclear how directional (i.e., effective) connectivity between various networks is altered by an actual FOG-related motor imagery task. Twenty-four individuals with PD (11 freezers and 13 non-freezers) and 15 healthy controls underwent functional MRI. Participants performed a motor imagery task while observing two videos: (1) a person with PD engaged in typical walking, and (2) a person with PD e

Interactions Between Blood Nutritional Biomarkers and Apolipoprotein E ε4 in the Progression of Mild Cognitive Impairment in Alzheimer's Disease.

BACKGROUND/OBJECTIVES: Mild cognitive impairment (MCI), the prodromal stage of Alzheimer's disease, may be influenced by nutritional status and genetic susceptibility. This systematic review synthesised evidence on how nutritional biomarkers interact with genetic variants, particularly APOE ε4, to influence cognitive outcomes in individuals with MCI. METHODS: Following PRISMA 2020 guidelines, seven studies were included (three longitudinal, two randomised controlled trials, and two cross-sectional) involving adults aged ≥55 years with MCI. Nutritional exposures comprised plasma or serum conce

Cerebellar Cognitive Affective Syndrome in Spinocerebellar Ataxia Type 6.

BACKGROUND: Spinocerebellar ataxia type 6 (SCA6) has traditionally been classified as a "pure" cerebellar syndrome; however, converging evidence points to non-motor involvement. Characterization of its cognitive-affective profile remains limited, constraining the understanding of its full clinical spectrum. OBJECTIVES: To investigate non-motor manifestations in SCA6 and contribute to a more comprehensive characterization of its cognitive-affective profile through standardized neuropsychological assessment. METHODS: A monocentric, cross-sectional case-control study included nine patients with

Staged Bilateral Magnetic Resonance-Guided Focused Ultrasound Thalamotomy for Essential Tremor: Prospective Single-Centre Cohort and Systematic Review With Meta-Analysis.

BACKGROUND: Staged bilateral Magnetic Resonance-guided Focused Ultrasound (MRgFUS) thalamotomy is an incisionless option for medication-refractory essential tremor (ET). While the efficacy and safety of unilateral MRgFUS are established, evidence for bilateral treatment remains limited. OBJECTIVE: To evaluate the efficacy and safety of staged bilateral MRgFUS in a prospective single-centre observational cohort and to perform a systematic review and meta-analysis of the literature. METHODS: Consecutive ET patients undergoing second-side MRgFUS were prospectively assessed. The primary efficacy

[18F]-florzolotau PET imaging in Alzheimer's disease: Current evidence and future directions.

Tau positron emission tomography (PET) has become a central imaging modality for in vivo detection and quantification of tau pathology across the Alzheimer's disease (AD) clinical and biological continuum. [18F]-Florzolotau is a second-generation tau PET tracer developed to improve target-to-background contrast and mitigate off-target binding relative to first-generation ligands, thereby improving interpretability for translational research and clinical use. In this narrative review, we provide a critical appraisal of the available literature on [18F]-Florzolotau PET imaging in AD, with emphas

Resting heart rate and cognitive function in ageing: Insights from a secondary data analysis.

Resting heart rate (RHR) is an accessible measure that may reflect age-related cardiovascular and autonomic dysregulation relevant to cognitive ageing. However, evidence linking RHR with cognitive outcomes is inconsistent, and most studies relied on single baseline measurements that may not capture cumulative exposure. This study examined the associations of baseline and longitudinally averaged RHR with cognitive decline and dementia in older adults. This was a secondary analysis of the ASPirin in Reducing Events in the Elderly (ASPREE) study, including 19,114 community-dwelling participants a
