Attention and memory in the preclinical stage of dementia.

The aim of this study was to investigate whether attention may be specifically impaired in Alzheimer's disease from the early stages of the disease. Subgroups of patients with different types of mild cognitive impairment were selected according to standard criteria. Patients and controls were given tasks exploring various subcomponents of attention and executive functions. Only subgroups of mild cognitive impairment characterized by memory disorders obtained lower scores than controls on attention and executive tasks. On the basis of the scores obtained on the Clinical Dementia Rating at the 1

Specific AD impairment in concurrent performance of two memory tasks.

The current study examined the hypothesis that patients with Alzheimer's disease (AD) have a selective deficit in the coordination mechanism of the central executive, which is reflected in their inability to coordinate the performance of two tasks concurrently. One criticism of the previous studies in the literature is that they employ tasks that do not necessarily draw upon the separate peripheral mechanisms within working memory but combine a memory load with a perceptuomotor load. Therefore, the aim of the study was to examine the effects of AD and healthy adult ageing on a verbal memory pl

A functional magnetic resonance imaging predictor of treatment response to venlafaxine in generalized anxiety disorder.

BACKGROUND: Functional magnetic resonance imaging (fMRI) holds promise as a noninvasive means of identifying neural responses that can be used to predict treatment response before beginning a drug trial. Imaging paradigms employing facial expressions as presented stimuli have been shown to activate the amygdala and anterior cingulate cortex (ACC). Here, we sought to determine whether pretreatment amygdala and rostral ACC (rACC) reactivity to facial expressions could predict treatment outcomes in patients with generalized anxiety disorder (GAD). METHODS: Fifteen subjects (12 female subjects) w

Procholinergic and memory enhancing properties of the selective norepinephrine uptake inhibitor atomoxetine.

Atomoxetine has been approved by the FDA as the first new drug in 30 years for the treatment of attention deficit/hyperactivity disorder (ADHD). As a selective norepinephrine uptake inhibitor and a nonstimulant, atomoxetine has a different mechanism of action from the stimulant drugs used up to now for the treatment of ADHD. Since brain acetylcholine (ACh) has been associated with memory, attention and motivation, processes dysregulated in ADHD, we investigated the effects of atomoxetine on cholinergic neurotransmission. We showed here that, in rats, atomoxetine (0.3-3 mg/kg, i.p.),--increases

Go-no-go task performance improvement after anodal transcranial DC stimulation of the left dorsolateral prefrontal cortex in major depression.

BACKGROUND: We recently showed that repetitive transcranial magnetic stimulation (rTMS) of the dorsolateral prefrontal cortex (DLPFC) can affect the performance in an affective go-no-go (AGN) task. We aimed to extend this previous investigation testing whether one session of anodal transcranial direct current stimulation (tDCS) of the left DLPFC, as compared with anodal occipital and sham tDCS, affects this AGN task performance. METHODS: Twenty-six patients with major depression were randomized to receive anodal tDCS of the left DLPFC, occipital cortex or sham tDCS (the cathode electrode was

Interaction of sensory and cognitive processes during visual recognition: the role of the associative areas of the cerebral cortex.

The first part of the present study used a model of Alzheimer's disease in two groups of animals (three monkeys in each), given injections of neurotoxins (monkeys of group I) and physiological saline (monkeys of group II). Before injections, all monkeys were trained to discriminate stimuli containing different types of information (spatial frequency grids and geometrical figures of different colors and with different spatial relationships between objects) and to perform spatial selection. The dynamics of impairments in the characteristics of working memory were identified using delayed differe

Covert processing of visual form in the absence of area LO.

The patient D.F., who suffers from severe visual form agnosia, has been found to have a bilateral lesion of area LO, an area known to be intimately involved in the perception of object shape. Despite her perceptual impairment, however, D.F. retains residual form processing abilities that can provide distal visuomotor control, for example in the configuration of her grasp when reaching to pick up objects of different shapes and sizes. This dissociation has been interpreted as reflecting the sparing of a dedicated system for processing the physical properties of objects solely for purposes of gu

Wavelet-generalized least squares: a new BLU estimator of linear regression models with 1/f errors.

Long-memory noise is common to many areas of signal processing and can seriously confound estimation of linear regression model parameters and their standard errors. Classical autoregressive moving average (ARMA) methods can adequately address the problem of linear time invariant, short-memory errors but may be inefficient and/or insufficient to secure type 1 error control in the context of fractal or scale invariant noise with a more slowly decaying autocorrelation function. Here we introduce a novel method, called wavelet-generalized least squares (WLS), which is (to a good approximation) th

Effects of the prolyl endopeptidase inhibitor S 17092 on cognitive deficits in chronic low dose MPTP-treated monkeys.

A number of neuropeptides are affected in Parkinson's disease and the enzyme proline endopeptidase contributes to the degradation of many of these neuropeptides, some of which are linked to a variety of cognitive functions. In the present study, the effects of the highly potent proline endopeptidase inhibitor S 17092 on cognitive deficits in monkeys induced by chronic low dose 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) administration were examined. Chronic low dose MPTP administration resulted in deficits in performance of variable delayed response, delayed matching-to-sample, and del

Increased amygdala activation to angry and contemptuous faces in generalized social phobia.

BACKGROUND: Generalized social phobia (GSP) is characterized by fear of social interactions and sensitivity to disapproval by others. Given the established role of the amygdala as part of a distributed neural system for the processing of emotional cues, we hypothesized that subjects with GSP would exhibit greater amygdala activation in response to harsh (angry, fearful, and contemptous) vs accepting (happy) facial emotional expressions compared with healthy control subjects (HCs). METHODS: Fifteen subjects with DSM-IV GSP and 15 age-, sex-, handedness-, and education-matched HCs, free of psyc

Distinct areas in parietal cortex involved in long-term and short-term action planning: a PET investigation.

The sequential organization of events within a script, can be considered as a means to explore the cognitive mechanisms involved in action planning. Scripts are composed of goal-oriented sequences of events that typically occur in a specific and systematic order. The purpose of this study was to investigate the effect of temporal variation between script events (short-term versus long-term) on neural activity in normal volunteers. Subjects were required to judge whether the chronological order of three events was correct or not, while regional cerebral blood flow was measured using positron em

Local-global processing in Alzheimer's disease: an examination of interference, inhibition and priming.

Impairments of memory, praxis, gnosis, language and executive functioning are well documented in Alzheimer's disease (AD). Functions, such as attention, however, have only recently been systematically investigated. We used Navon-type stimuli (large "global" digits composed of smaller "Локально" digits) to assess 12 AD participants' plus age-matched controls' ability to focus and alter the scale of their spatial attention. In the first experiment, participants responded to either the global or local characters within a block, ignoring characters at the other spatial scale. Healthy young adults (n=

Impaired word-stem completion priming but intact perceptual identification priming with novel words: evidence from the amnesic patient H.M.

We hypothesized that word-stem completion (WSC) priming and perceptual identification (PI) priming, two types of repetition priming, rely on different cognitive and neural mechanisms: WSC priming on a modification mechanism that influences lexical retrieval, and PI priming on plasticity in pre-lexical perceptual systems. We compared the priming performance of the amnesic patient H.M. with words that came into common usage after the onset of his amnesia, and thus were novel to him (post-1965 words), and with familiar (pre-1953) words. We also tested age- and education-matched normal control sub

Visual agnosia and optic aphasia: are they anatomically distinct?

A patient with left infero-medial occipital-temporal infarct suffered a visual agnosia that, by a minor change of the task, could be manipulated to optic aphasia. Tools in actual use and pantomimes of tool use were better named than stationary tools, a dissociation that suggests differences in the ability of stimuli to evoke associations over multiple modalities. Based on this case and analysis of previous reports we suggest that optic aphasia differs from visual agnosia primarily in the degree of callosal disconnection and that the preserved demonstration of tools use and semantic classificat

A woman in her 40s with an unusual migraine attack.

BACKGROUND: Migrainous infarction is a rare complication of migraine with aura, accounting for 0.5-1.5 % of all ischaemic strokes. Migraine with aura increases the risk of ischaemic stroke, particularly in women using oral contraceptives. Migrainous infarction is defined as an ischaemic stroke that occurs during a typical migraine with aura attack and is associated with a neuroimaging-confirmed ischaemic brain lesion in the relevant area. CASE PRESENTATION: This case describes a woman in her forties with a history of migraine with visual aura who developed transient neurological symptoms - in

Hypervigilance for fear after basolateral amygdala damage in humans.

Recent rodent research has shown that the basolateral amygdala (BLA) inhibits unconditioned, or innate, fear. It is, however, unknown whether the BLA acts in similar ways in humans. In a group of five subjects with a rare genetic syndrome, that is, Urbach-Wiethe disease (UWD), we used a combination of structural and functional neuroimaging, and established focal, bilateral BLA damage, while other amygdala sub-regions are functionally intact. We tested the translational hypothesis that these BLA-damaged UWD-subjects are hypervigilant to facial expressions of fear, which are prototypical innate

TMS over right OFA affects individuation of faces but not of exemplars of objects.

In addition to its well-documented role in processing of faces, the occipital face area in the right hemisphere (rOFA) may also play a role in identifying specific individuals within a class of objects. Here we explored this issue by using fMRI-guided TMS. In a first experiment, participants had to judge whether two sequentially presented images of faces or objects represented exactly the same exemplar or two different exemplars of the same class, while receiving online TMS over either the rOFA, the right lateral occipital cortex (rLO) or the Vertex (control). We found that, relative to Vertex

Different visual exploration of tool-related gestures in left hemisphere brain damaged patients is associated with poor gestural imitation.

According to the direct matching hypothesis, perceived movements automatically activate existing motor components through matching of the perceived gesture and its execution. The aim of the present study was to test the direct matching hypothesis by assessing whether visual exploration behavior correlate with deficits in gestural imitation in left hemisphere damaged (LHD) patients. Eighteen LHD patients and twenty healthy control subjects took part in the study. Gesture imitation performance was measured by the test for upper limb apraxia (TULIA). Visual exploration behavior was measured by an

Source memory errors in schizophrenia, hallucinations and negative symptoms: a synthesis of research findings.

BACKGROUND: Previous research has shown associations between source memory errors and hallucinations in patients with schizophrenia. We bring together here findings from a broad memory investigation to specify better the type of source memory failure that is associated with auditory and visual hallucinations. METHOD: Forty-one patients with schizophrenia and 43 healthy participants underwent a memory task involving recall and recognition of lists of words, recognition of pictures, memory for temporal and spatial context of presentation of the stimuli, and remembering whether target items were
