Longitudinal Systemic Transcriptomic Profiling and Neuropathological Deposition of Neutrophil Extracellular Traps in Parkinson's Disease.

BACKGROUND: Altered immune responses including increased neutrophils have been reported in Parkinson's disease (PD); however, the role of neutrophil extracellular traps (NETs) remains unclear. OBJECTIVE: The aim of this study was to evaluate the contribution of NETs to the systemic immunity and central nervous system (CNS) in PD pathogenesis. METHODS: Transcriptomic analysis from the Parkinson's Progression Markers Initiative (PPMI) cohort was performed. Serum NETs were measured by myeloperoxidase (MPO)-DNA and citrullinated histone H3 (CitH3)-DNA complexes using enzyme-linked immunosorbent

Plasma Amino Acid Signatures Associated with Disease Progression and Hypertension in Autosomal Dominant Polycystic Kidney Disease: A Targeted Metabolomics and Machine Learning Approach.

Background: Autosomal dominant polycystic kidney disease (ADPKD) is a clinically heterogeneous disorder often leading to end-stage renal disease (ESRD). Prognostication of disease progression remains a major clinical challenge. This study aimed to identify plasma amino acid signatures associated with ADPKD progression and hypertension. Methods: We conducted targeted metabolomic analysis (LC-MS/MS) to quantify 38 plasma amino acids in 203 ADPKD patients, stratified by disease progression (rapid vs. slow) and hypertension status. Support Vector Machine (SVM) models were developed to predict outc

Circadian disruption promotes epileptic seizures via the Ruminococcus gnavus-arginine-NOX4-ferroptosis axis.

Circadian rhythm disruption (CRD) exacerbates epileptic seizures, yet the underlying molecular mechanisms remain unclear. Using a pentylenetetrazol (PTZ) kindling rat model combined with continuous light exposure, we found that CRD significantly shortened seizure latency and increased seizure frequency. 16S rRNA sequencing revealed that CRD substantially increased the abundance of Ruminococcus gnavus (R. gnavus) in the gut microbiota. Antibiotic-mediated microbiota depletion, fecal microbiota transplantation (FMT), and R. gnavus monocolonization experiments confirmed that R. gnavus enrichment

NETosis Markers (ssDNA, dsDNA) as Predictors of Mortality and Hospitalization After Endovascular Aortic Repair.

Neutrophils and their extracellular traps (NETs) are pivotal elements of the immune response. This study investigates the dynamics of neutrophil-related markers during the perioperative period of branched endovascular aortic repair (BEVAR) in patients with thoracoabdominal aortic aneurysms (TAAAs) and evaluates their association with one-year clinical outcomes. A prospective, single-center study was conducted on 20 TAAA patients treated with T-branch devices. The analysis focused on surrogate markers associated with NETosis, including double-stranded DNA (dsDNA), single-stranded DNA (ssDNA), a

Circulating Markers of Neutrophil Extracellular Traps for Long-Term Prognosis in Patients With Acute Chest Pain.

BACKGROUND: Neutrophil extracellular traps are released from activated neutrophils and are involved in the pathogenesis of atherosclerotic lesions, atherothrombosis, and myocardial injury. We investigated the prognostic value of circulating neutrophil extracellular trap biomarkers in patients with suspected acute coronary syndrome (ACS). METHODS: A total of 1482 patients admitted with suspected non-ST-segment elevation ACS were included and followed for a median of 4.2 years. The primary end point was a composite of death from any cause, incident myocardial infarction and hospitalization for

Occupational cadmium exposure and endocrine biomarker alterations in male workers: a cross-sectional study.

BACKGROUND: Occupational exposure to cadmium (Cd) remains an important environmental health concern, particularly in industrial settings where long-term exposure may affect multiple physiological systems. Although Cd has been linked to reproductive and cardiovascular toxicity, human studies that simultaneously evaluate biomarkers of endocrine and endothelial dysfunction remain limited. This cross-sectional study examined the association between occupational Cd exposure and hormonal and endothelial biomarkers in adult male workers. METHODS: This cross-sectional study included 170 men, comprisi

Long-term alterations in serum metabolite profiles in women with a history of preeclampsia -FINNCARE study.

Preeclampsia (PE) is a multifaceted pregnancy condition which increases the risk of cardiometabolic diseases later in life. Long-term follow-up data on metabolome utilizing novel metabolomic techniques are needed to gain insights into metabolism after PE and non-PE pregnancies for identifying potential areas of investigation that could assist in predicting and managing future health risks. The objective of the current study was to characterize long-term metabolome in women with and without a history of PE. Non-targeted metabolomics (liquid chromatography-mass spectrometry, LC-MS) was applied t

The metabolic effects of post-sepsis feeding of meals with only essential amino acids.

Nutritional support is a vital component of the rehabilitation process following sepsis. Essential amino acids (EAA) are recognized as a critical component for stimulating anabolism. However, it remains unclear whether EAA supplementation alone is more effective than providing a complete profile of total amino acids (TAA). We studied amino acid (AA) metabolism in 51 catheterized pigs (±27 kg) using the pulse tracer approach over a 7-day recovery period following sepsis. Animals were randomly and blindly assigned to receive post-sepsis nutrition containing either an EAA or a TAA mixture based o

Autoreactivity Versus Autoimmunity: Characteristics of Autoantigen-Specific CD4 T Cells in Rheumatoid Arthritis and Health.

OBJECTIVE: CD4 T cells specific for citrullinated (cit) peptides are key players in rheumatoid arthritis (RA) immunopathogenesis. Characterizing these cells and identifying features of healthy and RA-associated autoreactivity will provide valuable insight into disease mechanisms and form the basis of immune state biomarkers to facilitate the next generation of RA treatments. We sought to characterize these cells in a UK early arthritis cohort. METHODS: Cit-peptide-specific (autoreactive) CD4 T cells from human leukocyte antigen (HLA)-DRB1 04:01-positive healthy controls (HCs; n = 9), patients

Glaesserella parasuis infection disrupts the gut-lung axis via microbiota dysbiosis and metabolic reprogramming leading to intestinal barrier impairment in piglets.

BACKGROUND: Glaesserella parasuis (G. parasuis), is a key respiratory pathogen responsible for Glässer's disease in pigs, characterized by polyserositis, arthritis, and pulmonary lesions. While it disrupts the respiratory microbiota, its impact on the gut-lung axis, a critical pathway for systemic immune and metabolic crosstalk, remains unexplored. METHODS: We established a piglet infection model using the highly virulent G. parasuis strain XX0306 (serotype 5). Systemic effects were investigated through integrated 16S rDNA sequencing of the lung and gut microbiota, complemented by untargeted
