INTRODUCTION: Excessive oxidative burst and dysregulated neutrophil extracellular trap (NET) formation contribute to tissue damage in acute lung injury (ALI) and are largely driven by the combined actions of NADPH oxidase 2 (NOX2) and myeloperoxidase (MPO). While lactoferrin (LTF) is a known multifunctional immunomodulatory glycoprotein, its precise role in modulating the NOX2-MPO-NETosis axis in ALI remains undefined.
METHODS: We employed a time-course model of lipopolysaccharide (LPS)-induced ALI in C57BL/6N mice, combined with quantitative label-free lung proteomics and downstream bioinfor

In inflammatory bowel disease (IBD), complex immune dysregulation induces chronic inflammation. TREM1 is a receptor expressed on myeloid cells and is activated by ligands such as PGLYRP1 to amplify inflammatory responses. Here, we analyze the function of TREM1 signaling in neutrophil-driven inflammation, identifying two positive feedback loops. First, neutrophils activated by phorbol 12-myristate 13-acetate (PMA) or by bacteria release PGLYRP1, which forms a ligand complex with bacterial peptidoglycan and activates TREM1, thereby enhancing neutrophil degranulation, reactive oxygen species prod

UNLABELLED: Diabetes is associated with impaired immune function and increased susceptibility to severe bacterial infections, yet the pathogen-encoded mechanisms that exacerbate disease in this context remain poorly defined. Streptococcus pyogenes (group A Streptococcus [GAS]) causes invasive skin and soft tissue infections that are disproportionately severe in individuals with diabetes, often accompanied by delayed healing, excessive inflammation, and polymicrobial overgrowth. Here, we investigated how the GAS ClpX-dependent regulatory pathway (CDRP), a global virulence regulator, interacts w

INTRODUCTION: Rheumatoid arthritis (RA) is characterized by persistent synovitis and progressive joint damage. Mounting evidence implicates neutrophil extracellular traps (NETs) formation (NETosis) in RA pathogenesis, yet the upstream regulatory nodes remain incompletely defined. We aimed to elucidate the role of the NLRP3 inflammasome in regulating GSDMD-dependent NETosis and to evaluate whether inhibiting NLRP3 or Gasdermin D (GSDMD) alleviates RA pathology.
METHODS: Neutrophils and synovial tissues from patients with RA and osteoarthritis (OA) were analyzed for NLRP3, GSDMD, and NET-relate…

A growing body of evidence indicates that neutrophils are the first major leukocyte population accumulating inside the pancreas even before … the expression of neutrophil elastase in this time period. We also detect components of neutrophil extracellular traps (NET) mainly in the exocrine tissue of the pancreas during this time as well as markers of vascular pathology as early as 2 weeks of age. Age- and sex-matched C57BL/6 mice do not exhibit these features inside the pancreas. When we treated NOD mice w …

BACKGROUND: Triple-negative breast cancer (TNBC) lacks stable druggable targets, is highly aggressive, and exhibits marked heterogeneity; consequently, therapeutic variability is often determined not only by tumor cell-intrinsic states but also by the immune-stromal ecology of the tumor microenvironment (TME) and its modes of spatial organization. The TNBC TME is jointly shaped by adaptive and innate immune cells, fibroblasts and the extracellular matrix, the vascular-hypoxia-lymphatic axis, and neural components, whose tissue-scale co-localization and boundary architecture underlie distinct p

Eosinophils are central effector and regulatory cells in asthma and eosinophilic respiratory disease, and a clinically relevant subset in a proportion of patients with chronic obstructive pulmonary disease (COPD) and asthma-COPD overlap. While eosinophil counts are established biomarkers to guide corticosteroid and biologic therapy, they do not capture the molecular heterogeneity of eosinophil activation across these conditions. Advances in integrative omics now permit comprehensive profiling of eosinophils at the transcriptomic, proteomic, and lipidomic levels. In this narrative review we sum

This study performs untargeted metabolomics on mouse serum and quantitative proteomics on xenograft tumors to explore the potential anti-tumor mechanisms of Inonotus obliquus polysaccharides (IOPs) against non-small cell lung cancer (NSCLC), revealing candidate pathways that warrant further experimental validation. Building on our prior discovery of IOP-induced apoptosis, we combined functional assays with multi-omics analyses using a well-characterized IOPs preparation (average molecular weight: 4.5 × 104 Da; polysaccharide content: 70.3%; primarily composed of glucose). IOPs potently inhibit

Endothelial cells (ECs) are key regulators in vascular homeostasis, and their physiological state depends on hemodynamic forces produced by the blood flow. Laminar shear stress (LSS) maintains endothelial integrity, whereas oscillatory shear stress (OSS) leads to endothelial dysfunction and subsequently to cardiovascular disease. Here, we delineated the proteomic signatures of human umbilical vein endothelial cells (HUVECs) exposed to a protective flow (LSS) or a disturbed flow (OSS) in comparison to static conditions, using trapped ion mobility spectrometry coupled with parallel accumulation-

Sepsis has high intensive care unit mortality because biomarkers for early reversible pathological events are lacking. Targeted nuclear degranulation, the rate-limiting step in neutrophil extracellular trap formation, refers to azurophilic granule fusion with the nuclear membrane. As an exclusive granule membrane marker, CD63 enrichment in nuclear membranes directly reflects this early pathogenic process. We developed a flow cytometry assay to quantify nuclear membrane CD63 (nmCD63) and validated its clinical utility in a two-center prospective cohort with strict independent external validatio

Chemotherapy-associated hepatotoxicity represents a critical limitation in cancer treatment, decreasing patient survival and quality of life. Irinotecan (CPT-11) is a key chemotherapeutic agent commonly used in the treatment of gastrointestinal malignancies. However, its clinical use is frequently limited by severe adverse effects, among which steatohepatitis has emerged as a critical dose-limiting toxicity. The mechanism of irinotecan-induced steatohepatitis remains unclear. In this study, we treated mice with CPT-11 to induce steatohepatitis and monitored intestinal barrier function using in

Dendritic cells (DCs) regulate both innate and adaptive immunity during sepsis. Prostaglandins (PGs), small lipid molecules derived from arachidonic acid via COX enzymes, are crucial regulators of immune homeostasis and inflammation. However, their role in sepsis pathogenesis remains poorly defined. In this study, we identified a significant negative correlation between DC depletion and disease severity in patients with sepsis. Thromboxane (TX) A2 receptor (TP) expression was markedly reduced in the blood DCs of patients with sepsis. Patients with low DC-TP expression presented increased blood

Acute pancreatitis (AP) is a severe inflammatory disorder characterized by a complex molecular pathophysiology involving premature zymogen activation, organelle dysfunction, and systemic immune dysregulation. Current therapeutic strategies remain largely supportive, underscoring the critical need for specific molecular-targeted interventions. Baicalein, a bioactive flavonoid derived from Scutellaria baicalensis Georgi, has emerged as a potent pleiotropic agent. This review comprehensively synthesizes the molecular mechanisms underlying baicalein's therapeutic efficacy in AP. Its capacity to in

Sepsis-induced acute lung injury (ALI) is a critical complication associated with high mortality. Esketamine, an N-methyl-d-aspartate (NMDA) receptor antagonist, has shown protective potential against organ injury; however, whether its protective effect against lung injury depends on NMDA receptor engagement remains unclear. This study investigated whether esketamine alleviates lipopolysaccharide (LPS)-induced ALI by targeting NMDA receptor subunit 2 A (NMDAR2A). In vivo, esketamine significantly reduced histopathological injury in the lungs, decreased the wet/dry (W/D) ratio, and suppressed p

Ankylosing spondylitis (AS) is an osteoimmune disease characterized by pathological enthesitis related to mechanical strain. However, the cell interactions and molecular mechanisms of AS enthesitis are still unclear. Herein, we constructed a hind paw loading/unloading model using experimental spondyloarthritis SKG mice, and generated a single-cell RNA sequencing atlas of mechanical strain-related AS enthesitis. In this context, a disease-specific subpopulation of SDC1+ sheath fibroblasts was identified to arise under mechanical strain, and these cells secreted higher levels of CXCL5 to recruit

The role of neutrophils in systemic sclerosis (SSc) remains incompletely understood. To address this, blood samples from 39 SSc patients, 39 healthy controls, and 22 systemic lupus erythematosus (SLE) patients were analyzed. Our results indicate that in SSc, neutrophils exhibited substantial activation, evidenced by granule mobilization, elevated plasma levels of Neutrophil Extracellular Trap (NET) byproducts, and upregulated TIE2 expression. In parallel, they underwent metabolic reprogramming, characterized by increased autophagy, likely to support the heightened energy demands of activation.

UNLABELLED: Autoimmune diseases, including systemic lupus erythematosus, rheumatoid arthritis, antiphospholipid syndrome, and systemic vasculitis, are associated with increased risk of thrombosis, accelerated atherosclerosis, cardiovascular-events, and premature mortality. Heparin-induced thrombocytopaenia and vaccine-induced thrombocytopaenia and thrombosis (VITT) or VITT like syndrome are autoimmune thrombotic disorders that do not have additional features of autoimmune diseases. We summarise the epidemiology, pathophysiology, diagnosis, and management of autoimmune thrombosis. Key mechanism

TLR4 activation and the cascade of neutrophil extracellular trap (NET) formation exacerbate excessive inflammation and organ damage in the pathogenesis of acute lung injury (ALI), yet effective pharmacological interventions remain unavailable. Nanoaggregates derived from natural products offer promising avenue by leveraging synergistic anti-inflammatory effects. In this study, we surprisingly discovered that leonurine and ursolic acid spontaneously self-assemble into nanoparticles (LUNP) through non-covalent interactions, achieving a drug loading capacity of 100%. The LUNP platform exhibits su

The lung may be the organ whose mechanical environment needs to be most finely tuned to achieve optimal function. These needs have to be fulfilled at multiple scales, from proper force transmission between the chest wall and the parenchyma to reduction of surface tension by surfactants inside the alveoli. In addition, a plethora of mechanical loads and forces takes place within the lung, from the passive stretch withstood by epithelial cells lining the alveoli, to active forces generated by smooth muscle cells to control airway calibre or cilia beating by ciliary cells in the bronchi to clear
