Structural features, IgE binding and preliminary clinical findings of the 7kDa Lipid Transfer Protein from tomato seeds.

Allergic reactions caused by 9kDa Lipid Transfer Proteins (9k-LTP), such as Pru p 3, have been widely investigated, whereas a possible contribution of components of 7kDa LTP (7k-LTP) sub-family in triggering allergic symptoms has been overlooked so far. With the aim to investigate the contribution of 7k-LTPs to the food allergies, we have identified, isolated and characterised a tomato seed 7k-LTP (Sola l 7k-LTP). The protein was purified by chromatographic separations, identified by direct protein sequencing and mass spectrometry and a molecular model was built. Functional evaluation of the a

An alginate-antacid formulation localizes to the acid pocket to reduce acid reflux in patients with gastroesophageal reflux disease.

BACKGROUND & AIMS: Alginate rafts (polysaccharide polymers that precipitate into a low-density viscous gel when they contact gastric acid) have been reported to form at the acid pocket, an unbuffered pool of acid that floats on top of ingested food and causes postprandial acid reflux. We studied the location of an alginate formulation in relation to the acid pocket and the corresponding effects on reflux parameters and acid pocket positioning in patients with gastroesophageal reflux disease (GERD). METHODS: We randomly assigned patients with symptomatic GERD and large hiatal hernias to groups

Update on taxane development: new analogs and new formulations.

The taxanes (paclitaxel and docetaxel) represent an important class of antineoplastic agents that interfere with microtubule function leading to altered mitosis and cellular death. Paclitaxel (Taxol(®)) was originally extracted from a yew tree (Taxus spp., Taxaceae) a small slow-growing evergreen, coniferous tree. Due to the initial scarcity of paclitaxel, docetaxel (Taxotere(®)) a semisynthetic analog of paclitaxel produced from the needles of European yew tree, Taxus baccata was developed. Docetaxel differs from paclitaxel in two positions in its chemical structure and this small alteration

Ether lipid vesicle-based antigens impart protection against experimental listeriosis.

BACKGROUND: Incidence of food-borne infections from Listeria monocytogenes, a parasite that has adapted intracellular residence to avoid antibody onslaught, has increased dramatically in the past few years. The apparent lack of an effective vaccine that is capable of evoking the desired cytotoxic T cell response to obliterate this intracellular pathogen has encouraged the investigation of alternate prophylactic strategies. It should also be noted that Archaebacteria (Archae) lipid-based adjuvants enhance the efficacy of subunit vaccines. In the present study, the adjuvant properties of archaeo

Important considerations for treatment with dietary supplement versus prescription niacin products.

Niacin is a water-soluble B vitamin (B3) known to have favorable effects on multiple lipid parameters, including raising high-density lipoprotein cholesterol (HDL-C) levels and lowering triglycerides (TGs), lipoprotein(a), and low-density lipoprotein cholesterol (LDL-C). Although LDL-C remains the primary target of lipid-altering therapy, current guidelines emphasize HDL-C and other modifiable lipid factors as key secondary targets. Thus, niacin is considered an important therapeutic option to help reduce the risk of cardiovascular disease in patients with mixed dyslipidemia who, in addition t

Identification of molecular targets associated with ethanol toxicity and implications in drug development.

Alcohol dependence is a major disease burden of adults in modern society worldwide. There is no cure for alcohol dependence. In this study, we have examined the molecular targets of ethanol-induced toxicity in humans based on a systematic review of literature data and then discussed current and potential therapeutic targets for alcohol abuse and dependence. Using human samples with ethanol exposure, microarray analyses of gene expression have shown that numerous genes are up- and/or down-regulated by alcohol exposure. The ethanol-responsive genes mainly encode functional proteins such as prote

Clinical efficacy of a chlorous acid preoperative skin antiseptic.

BACKGROUND: Among the ways to reduce the incidence of iatrogenic infectious disease is the use of efficacious preoperative antiseptics. Iodophors and chlorhexidines, the chief presurgical disinfectants today, have various problems with practicality. A new preoperative skin antiseptic has been developed (Alcide Corporation, Redmond, Wash) that involves the mixture of a 0.1% sodium chlorite formulation and an activating preparation of 0.5% mandelic acid to produce chlorous acid and other antimicrobial degradation products. METHODS: Under clinical conditions, the antimicrobial effectiveness of t

Enzymatic treatment-induced alterations in printing performance of quinoa flours and microbiota-modulating potential of printed products.

Quinoa is a representative pseudocereal widely used in developing gluten-free products for gluten-sensitive people. While enzymatic pretreatment improves the processing performance of pseudocereal flours, the impacts of glycoside hydrolases on the microbiota-modulating potential of quinoa products remain largely unexplored. This study compared the three-dimensional (3D) printing performance of wheat and quinoa flours, as well as quinoa flours treated with α-galactosidase, β-glucanase, or their sequential enzymatic treatment (α-galactosidase followed by β-glucanase), and then evaluated the colo

Impact of Dosing Interval Adherence on Pharmacokinetic Outcomes of Recombinant Erwinia Asparaginase in Pediatric Leukemia or Lymphoma.

INTRODUCTION: Treatment for pediatric acute lymphoblastic leukemia includes asparaginase as an integral component of therapy. Hypersensitivity reactions or silent inactivation to Escherichia coli (E. coli) formulations may necessitate switching to Erwinia-based asparaginase (asparaginase Erwinia chrysanthemi [E-ASP] or recombinant asparaginase Erwinia chrysanthemi [R-ASP]). However, pharmacokinetic data for R-ASP in pediatrics remain limited, and recommended dosing schedules present logistical challenges. We investigated if adherence to approved dosing regimens for R-ASP is necessary to achiev

Acetic acid-assisted controlled drying improves the room-temperature solubility of gelatin by suppressing chain crystallization and aggregation.

Achieving room-temperature solubility of gelatin remains challenging because chain crystallization and intermolecular aggregation readily occur during conventional drying. Here, acetic acid-assisted controlled drying at 45 °C was developed to improve the room-temperature solubility of type A gelatin while preserving molecular integrity. Controlled drying suppressed triple-helix crystallization, whereas acetic acid further inhibited chain aggregation, enabling aggregate formation and room-temperature solubility to be modulated by adjusting the acetic acid concentration before drying. Raising th

Buffering properties of fish aquafeeds: concepts, determinants and effects on physiology and growth.

The present paper is a comprehensive review of the existing literature on aquafeed acid binding capacity (ABC), and its effects on the digestive physiology, metabolism, intestinal microbiota, and growth of farmed fishes. ABC can be defined as the quantity of acid (usually HCl) required to reduce diet pH to a specified endpoint, typically 3.0 (ABC-3) or 4.0 (ABC-4). The ABC of a diet may influence the secretion pattern and minimum gastric pH, as well as the total amount of HCl secreted during digestion. On the other hand, the acidity of the gastric lumen plays a crucial role in the digestive pr

A natural probioenzyme strategy as an effective alternative to antibiotics against multidrug-resistant Klebsiella pneumoniae in broilers: Benefits for growth, immunity, and organ health.

The rise of multidrug-resistant Klebsiella pneumoniae poses a serious threat to poultry health and food safety, necessitating sustainable alternatives to antibiotics. Our investigation began with an epidemiological survey of broiler samples, which detected K. pneumoniae in 8.88% (8/90) of cases. Among these isolates, over 60% were multidrug-resistant. Antimicrobial susceptibility testing showed bacterial resistance to penicillin G (100%) but complete sensitivity to colistin sulphate (100%). Molecular analysis further revealed a high prevalence of key resistance genes (tet A [87.5%] and bla TEM

Patents on Small-Molecule Drugs Approved by the US Food and Drug Administration, 1990-2019.

IMPORTANCE: Patent protection allows pharmaceutical companies to charge high prices for prescription drugs during periods of market exclusivity, and their expiration usually defines when generic competitors can enter the US market. Patents on drug active ingredients are classified as primary patents, while patents on formulations, methods of use, and delivery devices are nonprimary. OBJECTIVE: To evaluate how pharmaceutical patent portfolios for US Food and Drug Administration (FDA)-approved drugs have evolved over time. EVIDENCE REVIEW: Patents on small-molecule drugs approved by the FDA fr

Plant-derived and microbial biostimulants in sustainable agriculture: mechanisms, applications, and challenges.

Plant biostimulants have emerged as transformative and sustainable tools for improving crop productivity, resource-use efficiency, and resilience under rapidly intensifying environmental stresses. Unlike conventional agrochemicals, biostimulants function by activating physiological, biochemical, and molecular processes that optimize plant performance without directly supplying nutrients or exerting pesticidal effects. This review comprehensively examines the integrated roles of plant-derived and microbial biostimulants in sustainable agriculture, with particular emphasis on microbial-mediated

Fermented Plant-Based Foods and Postbiotics for Glycemic Control-Microbial Biotransformation of Phytochemicals.

Plant-based fermented foods are increasingly promoted for glycemic control, yet their mechanisms and clinical impact remain incompletely defined. This narrative review synthesizes mechanistic, preclinical, and human data for key matrices-kimchi and other fermented vegetables, tempeh/miso/natto, and related legume ferments, kombucha and fermented teas, plant-based kefir, and cereal/pulse sourdoughs. Across these systems, microbial β-glucosidases, esterases, tannases, and phenolic-acid decarboxylases remodel polyphenols toward more bioaccessible aglycones and phenolic acids, while lactic and ace

Use of age-specific human in vitro platforms to advance discovery and development of novel vaccine adjuvants.

Populations that are particularly vulnerable to infections, such as neonates, infants, and older adults, exhibit distinct immunological profiles contributing to sub-optimal responses to conventional vaccines. Historically, vaccine development has relied on animal models; however, these are limited by low-throughput, bioethical concerns, and species-specific biological differences that do not reliably predict human responses. Moreover, conventional methods of adjuvant discovery and development have failed to consider that human immune responses vary by a range of demographic features, especiall
