Serine proteases as luminal mediators of intestinal barrier dysfunction and symptom severity in IBS.

OBJECTIVE: The intestinal lumen contains several proteases. Our aim was to determine the role of faecal proteases in mediating barrier dysfunction and symptoms in IBS. DESIGN: 39 patients with IBS and 25 healthy volunteers completed questionnaires, assessments of in vivo permeability, ex vivo colonic barrier function in Ussing chambers, tight junction (TJ) proteins, ultrastructural morphology and 16 s sequencing of faecal microbiota rRNA. A casein-based assay was used to measure proteolytic activity (PA) in faecal supernatants (FSNs). Colonic barrier function was determined in mice (ex-germ f

Effect of short-term intermittent antibiotic treatment on growth of Burmese (Myanmar) village children.

To test the hypothesis that subclinical enteric infection (such as bacterial overgrowth), rice malabsorption, and growth faltering are causally linked, a field trial of low-dose, short-term, intermittent antibiotic treatment was carried out in 142 hydrogen-producing (by lactulose breath hydrogen test) Burmese village children aged 6-59 months. The children were randomly allocated treatment with metronidazole (20 mg/kg or 5 mg/kg daily), amoxycillin (25 mg/kg daily), or placebo given 1 week per month for 6 months. A cooked rice meal breath hydrogen test was done to classify the children as rice

EndoBarrier gastrointestinal liner. Delineation of underlying mechanisms and clinical effects.

Bariatric surgery (e.g. Roux-en-Y gastric bypass (RYGB)) has proven the most effective way of achieving sustainable weight losses and remission of type 2 diabetes (T2D). Studies indicate that the effectiveness of RYGB is mediated by an altered gastrointestinal tract anatomy, which in particular favours release of the gut incretin hormone glucagon-like peptide-1 (GLP-1). The EndoBarrier gastrointestinal liner or duodenal-jejunal bypass sleeve (DJBS) is an endoscopic deployable minimally invasive and fully reversible technique designed to mimic the bypass component of the RYGB. Not only GLP-1 is

Piperine-loaded solid lipid nanoparticles: a promising nano-phytomedicine for the treatment of non-alcoholic fatty liver disease.

INTRODUCTION: Piperine (PIP), the active alkaloid found in black pepper (Piper nigrum), has gained attention for its potential therapeutic role in managing non-alcoholic fatty liver disease (NAFLD) due to its anti-inflammatory, lipid-lowering, antioxidant, and insulin-sensitising properties. Nevertheless, PIP's poor solubility limits its absorption in the gastrointestinal tract, thereby, compromising its systemic bioavailability. Consequently, the objective of this research has been to formulate piperine-loaded solid lipid nanoparticles (PIP- SLNs) so as to increase its oral bioavailability an

Bioorthogonal catalytic centres engineered for gastrointestinal stabilization provide oral delivery for the treatment of gastric cancer.

BACKGROUND: Gastric cancer, the fifth most prevalent cancer globally, poses significant treatment challenges due to factors such as late diagnosis, early metastasis, limited surgical options, and the systemic toxicity of chemotherapy. Because luminal barriers are often compromised in gastric cancers , orally administered therapies that enable localized absorption and drug release represent a promising new direction for site-specific treatment with limited side effects. RESULTS: We introduced disulfide-linked thermostable exoshell system that orally delivered protein-based bioorthogonal cataly

Polymyxin B-treated outer membrane vesicles of Campylobacter jejuni as prospective mucosal vaccine candidates in chickens.

Campylobacter jejuni (C. jejuni) is the leading cause of bacterial gastroenteritis in humans. Primarily, contaminated poultry meat and meat products account for nearly 80% of human infections. Reducing C. jejuni colonization of the chicken gastrointestinal tract is a key strategy to limit the risk of foodborne transmission to humans. To this end, outer membrane vesicles (OMVs) derived from C. jejuni were shown to have strong immunogenic potential in both avian and mammalian models. However, a major limitation of OMV-based vaccines against C. jejuni is the presence of lipooligosaccharide (LOS)

Oral administration of curcumin and quercetin nanoparticles can improve ulcerative colitis by regulating intestinal microorganisms.

BACKGROUND: Curcumin has been proved to relieve ulcerative colitis (UC). However, the premature release of gastrointestinal tract will lead to insufficient accumulation of curcumin in colon and affect the improvement effect. Therefore, we developed a kind of oral nanoparticles (Zein-CS-Cur-Que), which made quercetin promote the absorption of curcumin in colon by efflux carrier and inhibiting metabolic enzymes, and at the same time encapsulated curcumin and quercetin with zein and sodium caseinate to promote the targeted release in colon. METHODS: 3.5% dextran sodium sulfate was used to establ
