Rapid picture naming in Parkinson's disease using the Mobile Universal Lexicon Evaluation System (MULES).

OBJECTIVE: The Mobile Universal Lexicon Evaluation System (MULES) is a test of rapid picture naming that captures extensive brain networks, including cognitive, language and afferent/efferent visual pathways. MULES performance is slower in concussion and multiple sclerosis, conditions in which vision dysfunction is common. Visual aspects captured by the MULES may be impaired in Parkinson's disease (PD) including color discrimination, object recognition, visual processing speed, and convergence. The purpose of this study was to compare MULES time scores for a cohort of PD patients with those fo

Electrophysiological recordings during the peripheral nerve evaluation (PNE) test in complete spinal cord injury patients.

Up until now, the exact mechanism by which neuromodulation using sacral nerve stimulation works still remains unknown. Recent studies of pel … the anal contractions observed during peripheral nerve evaluation are reflex responses mediated by afferent pathways. Both the early and late reflex responses are of spinal origin, since they were obtained in complete SCI patients in whom all spino-bulbo spinal loops are supposed to be interrupted. The finding that neuromodulation is working in non-neurogenic pati …

Pulsed taVNS-Elicited Pupil Dilation: Effects of Intermixed Stimulation, Sham Location, and Respiratory Phase.

Transcutaneous auricular vagus nerve stimulation (taVNS) may offer a powerful, noninvasive way to stimulate activity of brainstem arousal systems, including the locus coeruleus-norepinephrine (LC-NE) system. Pulsed taVNS is known to elicit pupil dilation, a marker of LC activity. However, studies reporting taVNS effects on pupil dilation have delivered taVNS and sham stimulation in separate blocks of trials or in separate sessions, hindering the integration of taVNS into rapid, event-related task designs of cognitive neuroscientists. In two experiments, we examined the effectiveness of pulsed

The regulation of PKA signaling in obesity and in the maintenance of metabolic health.

The cAMP-dependent protein kinase (PKA) system represents a primary cell-signaling pathway throughout systems and across species. PKA facilitates the actions of hormones, neurotransmitters and other signaling molecules that bind G-protein coupled receptors (GPCR) to modulate cAMP levels. Through its control of synaptic events, exocytosis, transcriptional regulation, and more, PKA signaling regulates cellular metabolism and emotional and stress responses making it integral in the maintenance and dysregulation of energy homeostasis. Neural PKA signaling is regulated by afferent and peripheral ef

Microstructural damage of the cortico-striatal and thalamo-cortical fibers in Fabry disease: a diffusion MRI tractometry study.

PURPOSE: Recent evidences have suggested the possible presence of an involvement of the extrapyramidal system in Fabry disease (FD), a rare X-linked lysosomal storage disorder. We aimed to investigate the microstructural integrity of the main tracts of the cortico-striatal-thalamo-cortical loop in FD patients. METHODS: Forty-seven FD patients (mean age = 42.3 ± 16.3 years, M/F = 28/21) and 49 healthy controls (mean age = 42.3 ± 13.1 years, M/F = 19/28) were enrolled in this study. Fractional anisotropy (FA), axial (AD), radial (RD), and mean diffusivity (MD) maps were computed for each subjec

Spontaneous and Bite-Evoked Muscle Pain Are Mediated by a Common Nociceptive Pathway With Differential Contribution by TRPV1.

UNLABELLED: Spontaneous pain and function-associated pain are prevalent symptoms of multiple acute and chronic muscle pathologies. We established mouse models for evaluating spontaneous pain and bite-evoked pain from masseter muscle, and determined the roles of transient receptor potential cation channel subfamily V member 1 (TRPV1) and the contribution of TRPV1- or neurokinin 1 (NK1)-dependent nociceptive pathways. Masseter muscle inflammation increased Mouse Grimace Scale scores and face-wiping behavior, which were attenuated by pharmacological or genetic inhibition of TRPV1. Masseter inflam

Endogenous N-acetylaspartylglutamate (NAAG) inhibits synaptic plasticity/transmission in the amygdala in a mouse inflammatory pain model.

BACKGROUND: The peptide neurotransmitter N-acetylaspartylglutamate (NAAG) is widely expressed throughout the vertebrate nervous system, including the pain processing neuraxis. Inhibitors of NAAG peptidases are analgesic in animal models of pain. However, the brain regions involved in NAAG's analgesic action have not been rigorously defined. Group II metabotropic glutamate receptors (mGluR2/3) play a role in pain processing in the laterocapsular part of the central nucleus of the amygdala (CeLC). Given the high concentration of NAAG in the amygdala and its activation of group II mGluRs (mGluR3

Nerve injury-induced tactile allodynia is present in the absence of FOS labeling in retrogradely labeled post-synaptic dorsal column neurons.

The dorsal column pathway consists of direct projections from primary afferents and of ascending fibers of the post-synaptic dorsal column (PSDC) cells. This pathway mediates touch but may also mediate allodynia after nerve injury. The role of PSDC neurons in nerve injury-induced mechanical allodynia is unknown. Repetitive gentle, tactile stimulus or noxious pinch was applied to the ipsilateral hindpaw of rats with spinal nerve ligation (SNL) or sham surgery that had previously received tetramethylrhodamine dextran in the ipsilateral n. gracilis. Both touch and noxious stimuli produced marked

Origin of the secondary increase in firing probability of human motor neurons following transcranial magnetic stimulation. Studies in healthy subjects, type I hereditary motor and sensory neuropathy and multiple sclerosis.

In tonically active human motor neurons serving upper limb muscles there is a late rise in firing probability following transcranial magnetic stimulation termed the secondary peak (SP). The aim was to study the mechanism of SP and the pathway that mediates it. For this purpose, the response to transcranial magnetic stimulation of 62 repetitively firing low threshold single motor units from upper limb muscles was studied with peri-stimulus time histograms in 8 healthy subjects and in 13 patients with either Type I hereditary motor and sensory neuropathy (HMSN) or multiple sclerosis (MS). Sympathetic control of jejunal fluid and electrolyte transport. An experimental study in cats and rats.

The study was performed to test the hypothesis that the sympathetic nerves to the small intestine might be involved in the feed-back control of extra-cellular fluid volume. In a series of experiments in anaesthetized cats, we studied how efferent stimulation of the splanchnic nerves affected jejunal fluid transport rate. The mechanisms behind the observed responses were analysed and we also attempted to determine the physiological role of the sympathetic supply to the jejunal mucosa. Splanchnic nerve stimulation (SNS) led to an increase in net fluid absorption provided that glucose was present

Dietary sodium modulates the interaction between efferent renal sympathetic nerve activity and afferent renal nerve activity: role of endothelin.

Increasing efferent renal sympathetic nerve activity (ERSNA) increases afferent renal nerve activity (ARNA), which in turn decreases ERSNA via activation of the renorenal reflexes in the overall goal of maintaining low ERSNA. We now examined whether the ERSNA-induced increases in ARNA are modulated by dietary sodium and the role of endothelin (ET). The ARNA response to reflex increases in ERSNA was enhanced in high (HNa)- vs. low-sodium (LNa) diet rats, 7,560 +/- 1,470 vs. 900 +/- 390%.s. Cutaneous pain in disorders affecting peripheral nerves.

Our ability to quickly detect and respond to harmful environmental stimuli is vital for our safety and survival. This inherent acute pain detection is a "gift" because it both protects our body from harm and allows healing of damaged tissues [1]. Damage to tissues from trauma or disease can result in distorted or amplified nociceptor signaling and sensitization of the spinal cord and brain (Central Nervous System; CNS) pathways to normal input from light touch mechanoreceptors. Together, these processes can result in nagging to unbearable chronic pain and extreme sensitivity to light skin touc

Nonlinear feedback model of neuronal populations in hippocampal CAl region.

1. A lumped-circuit model is proposed for the local interactions within the hippocampal CAl region. Each neuronal population is represented by a linear differential equation or a linear transfer function in the Laplace domain. Interactions between neuronal populations are represented by gain factors. Recurrent inhibition of pyramidal cells by the inhibitory interneurons is the important interaction represented by an asymmetric, bidirectionally saturating gain curve. The inputs to the model are orthodromic or antidromic inputs to the pyramidal cells and a tonic input from the brain stem. The ou

Rehabilitating homonymous visual field deficits: white matter markers of recovery-stage 1 registered report.

Damage to the primary visual cortex (V1) or its afferent white matter tracts results in loss of vision in the contralateral visual field that can present as homonymous visual field deficits. Recent evidence suggests that visual training in the blind field can partially reverse blindness at trained locations. However, the efficacy of visual training to improve vision is highly variable across subjects, and the reasons for this are poorly understood. It is likely that variance in residual functional or structural neural circuitry following the insult may underlie the variation among patients. Ma

Chemogenetic inhibition of Grpr- and/or Npff-expressing spinal neurons in mice suppresses chloroquine-evoked itch but not signs of mechanical allodynia in a neuropathic pain model.

Mechanical hypersensitivity following peripheral nerve injury is common and difficult to treat. It has been suggested that this results from activation of a normally silent spinal cord circuit that links low-threshold mechanoreceptive primary afferents to nociceptive projection neurons in lamina I and that this pathway involves a class of excitatory interneurons known as vertical cells. We have recently identified 2 neurochemical types of vertical cells, based on expression of the gastrin-releasing peptide receptor (GRPR) and neuropeptide FF (NPFF). Although little is known about the role of N

The Genetic Architecture of Chronic Cough: From Sensory Hypersensitivity to Treatable Trait.

Chronic cough is a prevalent global clinical disorder with substantial quality-of-life impairment, and refractory cases remain a major unmet medical need. Cough hypersensitivity syndrome is the core pathological mechanism of chronic cough, and growing genetic evidence has confirmed that inherited susceptibility shapes cough hypersensitivity, clinical heterogeneity and therapeutic responsiveness, redefining chronic cough as a biologically mediated sensory-neural disorder rather than a non-specific secondary symptom of airway diseases. This review summarises genetic evidence for chronic cough fr

The Size and Localization of Ribeye and GluR2 in the Auditory Inner Hair Cell Synapse of C57BL/6 Mice Are Affected by Short-Pulse Corticosterone in a Sex-Dependent Manner.

Background: Inner hair cell (IHC) ribbon synapses are the initial synapses in the auditory pathway, comprising presynaptic ribbons and postsynaptic glutamate receptors on the peripheral afferent fibers. The excitatory neurotransmitter glutamate primarily signals through AMPA-type heterotetrameric receptors (AMPARs), composed of GluR1, GluR2, GluR3, and GluR4 subunits. Research shows that corticosterone affects AMPA receptor subunits in the central nervous system. The present study investigates the effects of corticosterone on AMPA receptor subunits in the murine cochlea. Methods: Cochlear expl

Functional Development of the Auditory Brainstem Nuclei During Embryogenesis of the Mouse Revealed by Optical Recording With a Voltage-Sensitive Dye.

A fundamental question in auditory neuroscience is when and how complex connections between the nuclei/centers are organized during ontogenesis. In the present study, we applied a multiple-site optical recording technique with a voltage-sensitive dye and surveyed the developmental organization of the auditory system in the mouse embryo. Stimulation of the cochlear (auditory) nerve elicited excitatory postsynaptic potential (EPSP)-related optical responses in the lateral brainstem, which corresponded to the auditory sensory nucleus, namely, the cochlear nucleus. The EPSP was mediated by glutama

The TRPA1 Ion Channel Mediates Oxidative Stress-Related Migraine Pathogenesis.

Although the introduction of drugs targeting calcitonin gene-related peptide (CGRP) revolutionized migraine treatment, still a substantial proportion of migraine patients do not respond satisfactorily to such a treatment, and new therapeutic targets are needed. Therefore, molecular studies on migraine pathogenesis are justified. Oxidative stress is implicated in migraine pathogenesis, as many migraine triggers are related to the production of reactive oxygen and nitrogen species (RONS). Migraine has been proposed as a superior mechanism of the brain to face oxidative stress resulting from ener
