Harnessing eDNA metabarcoding for mosquito surveillance: a pilot study in Ethiopia's Great Rift Valley.

BACKGROUND: Vector-borne diseases account for over 17% of all infectious diseases worldwide, with malaria and mosquito-borne arboviruses eme … mental samples analyzed, eight (61.5%) tested positive for mosquito DNA, identifying three species: Anopheles coustani, Culex poicilipes, and Mansonia uniformis. Notably, this marks the first documented detection of Cx. poicilipes in Ethiopia, a known vector for Rift Valley Fever Virus (RVFV). The positive samples were collected from various habitats, inclu …

Modelling the effects of precipitation and temperature on malaria incidence in coastal and western Kenya.

BACKGROUND: Malaria continues to plague sub-Saharan Africa despite great efforts geared towards its mitigation. In Kenya alone, 70% of the population remains at risk for malaria every year. Malaria is spread by Anopheles mosquitoes carrying the Plasmodium parasite, and displays a complex ecology with various socio-economic, biophysical factors and meteorological predictors, particularly temperature and precipitation, associated with the occurrence of the disease. METHODS: This study estimated the empirical relationship of temperature and precipitation on the temporal population dynamics of sy…

RESULTS: AMEL could be successfully amplified up to 24 h after a blood meal in 100% of An. coluzzii and 96.6% of Ae. aegypti, revealing the sex of humans that were fed on by individual mosquitoes. CONCLUSIONS: The method described here, …

In order to determine the seasonal prevalence and population density of vector mosquitoes in Kyungpook province, a survey based on average number of female mosquito per trap-night, were carried out during the period from May to November in 1984. Among the 34,571 mosquitoes collected in Kyungsan county in animal shelters and human dwellings by light traps, approximately 45.0 % were Culex tritaeniorhynchus, 34.0 % per cent Culex pipiens pallens, and 19.0 per cent Anopheles sinensis. Characterization of a new highly mosquitocidal isolate of Bacillusthuringiensis--an alternative to Bti?

The mosquito is a very important vector involved in the worldwide transmission of disease-causing viruses and parasites. Controlling the mosquito population remains one of the best means for preventing the serious infectious diseases of malaria, yellow fever, dengue, filariasis and so on and there has been an increasing interest in developing biopesticides as a useful substitute to chemical insecticides. As a result, Bacillus thuringiensis subsp. israelensis (Bti) has been extensively used due to its specificity and high toxicity to a variety of mosquito larvae. However it is prudent to seek a

Individual and household level factors associated with malaria incidence in a highland region of Ethiopia: a multilevel analysis.

Multilevel analysis was used to identify individual and household level factors associated with malaria risk in 1,367 individuals in a peri-urban area of highland Ethiopia. Living within 450 m of a major vector-breeding site accounted for 38.78% and 78.49% of between-household variance in malaria incidence in adults and children, respectively. In adults, other individual level factors associated with malaria risk were regular or recent travel to rural areas (incidence rate ratio [IRR] = 12.96; 95% confidence interval [CI] = 4.05, 41.48) and having an indoor job (IRR = 0.37; 95% CI = 0.16, 0.87

Application of the indirect fluorescent antibody method in a study of malaria endemicity in Mato Grosso, Brazil.

Four surveys of malaria endemicity were conducted in the Cuiabá Sector of Mato Grosso State, Brazil, at 6-month intervals during the period April 1970 to September 1971; during April 1970 a survey was also conducted in the Campo Grande Sector. Over 4,000 dual specimens--blood films for parasite diagnosis and filter paper blood spots for determination of fluorescent antibody (IFA) response--were collected from the general population, including school populations whenever possible. Parasitologic examinations yielded positivity rates ranging from 0.8 to 2.3%. In the Cuiabá Sector, sero-positives

Avid binding by B cells to the Plasmodium circumsporozoite protein repeat suppresses responses to protective subdominant epitopes.

Antibodies targeting the NANP/NVDP repeat domain of the Plasmodium falciparum circumsporozoite protein (CSPRepeat) can protect against malaria. However, it has also been suggested that the CSPRepeat is a decoy that prevents the immune system from mounting responses against other domains of CSP. Here, we show that, following parasite immunization, B cell responses to the CSPRepeat are immunodominant over responses to other CSP domains despite the presence of similar numbers of naive B cells able to bind these regions. We find that this immunodominance is driven by avid binding of the CSPRepeat

Generation of an antibody that recognizes Plasmodium chabaudi cysteine protease (chabaupain-1) in both sexual and asexual parasite life cycle and evaluation of chabaupain-1 vaccine potential.

Malaria cysteine proteases have been shown to be immunogenic and are being exploited as serodiagnostic markers, drug and vaccine targets. Several Plasmodium spp. cysteine proteases have been described and the best characterized of these are the falcipains, a family of papain-family enzymes. Falcipain-2 and falcipain-3 act in concert with other proteases to hydrolyze host erythrocyte hemoglobin in the parasite food vacuole. Falcipain-1 has less similarity to the other falcipains and its physiological role in parasite asexual blood stage still remains uncertain. Immunolocalization studies using

A sero-epidemiological study of malaria in human and monkey populations in French Guiana.

This paper describes a sero-epidemiological study of malaria prevalence in French Guiana. An immunofluorescence assay and an enzyme-linked immunosorbent assay were used to detect antibodies against blood-stage antigens and synthetic peptides mimicking the repetitive epitope of the sporozoites of Plasmodium falciparum, Plasmodium vivax and Plasmodium malariae/brasilianum, in 218 human sera and 113 non-human primate sera collected in French Guiana. Almost all the monkey sera tested had antibodies against malaria blood-stages (98%) and a large majority (73%) also tested positive with the P. malar
