The Effects of Chicory Aqueous Extract on Antioxidant Capacity, Immune Response, and Intestinal Function in Broilers.

In the broiler production industry, antibiotics such as florfenicol are widely used. Although these antibiotics exert therapeutic effects against bacterial diseases, they can induce a series of physiological alterations in broilers under certain exposure scenarios. This study aimed to investigate the effects of chicory aqueous extract (CAE) on broiler growth performance, immune function, and intestinal health, and to compare the impacts of CAE and florfenicol exposure on the physiological status of broilers. A total of 200 one-day-old broilers were randomly divided into five groups (5 replicat

Habitual Ultra-processed Food Intake Is Associated with Gut Dysbiosis and Pro-inflammatory Metabolite Profiles in Korean Patients with IBD.

BACKGROUND AND AIMS: Ultra-processed food (UPF) is increasingly consumed worldwide and may influence gut microbial ecology relevant to inflammatory bowel disease (IBD). However, patient-level multi-omics data remains scarce. We investigated whether habitual UPF intake is associated with specific microbiota and metabolite profiles in Korean patients with IBD. METHODS: Dietary intake was assessed using a validated food frequency questionnaire, and food was categorized by the NOVA system. UPF intake was expressed as percent of energy, and 313 patients were stratified into UPF low (Q1-Q2) and UPF

Dihydroartemisinin alleviates dextran sulfate sodium-induced colitis in mice: Associations with gut microbiota, arachidonic acid metabolism, and the IL-17 signaling pathway.

BACKGROUND: Dihydroartemisinin (DHA), a sesquiterpene lactone derived from artemisinin, exhibits well-documented anti-inflammatory and anti-cancer properties. However, the precise mechanisms through which DHA alleviates ulcerative colitis (UC) remain incompletely understood. PURPOSE: This study aimed to investigate the effects and underlying mechanisms of DHA on dextran sulfate sodium (DSS)-induced colitis in mice using a multi-omics approach, including 16S rRNA microbiomics, non-targeted and targeted metabolomics, RNA-seq transcriptomics and network pharmacology analysis. METHODS: A DSS-ind

Human Milk and Infant Gut Microbiome in Association With Infant Fecal Metabolome and Child Blood Pressure.

IMPORTANCE: Infant milk feeding type (eg, human milk vs formula) and infant gut microbes have each been associated with differences in microbial metabolites and childhood blood pressure; however, evidence remains limited regarding how specific infant gut microbes, at a species or strain level, in combination with milk feeding type, shape microbial metabolites and blood pressure. OBJECTIVE: To investigate whether human milk feeding and infant gut microbes, including Bifidobacterium longum subsp infantis (B infantis) and other milk-degrading microbes, are associated with infant fecal metabolite

Dietary tryptophan mitigates lung ischemia-reperfusion injury via microbiota-derived indole-3-propionate and aryl hydrocarbon receptor signaling.

BACKGROUND: Lung ischemia-reperfusion (IR) injury drives early morbidity after lung transplantation and cardiothoracic surgery, yet targeted preventive therapies are lacking. The gut-lung axis and microbiota-derived tryptophan metabolites, including indole-3-propionate (IPA), may regulate pulmonary immunity and inflammation. We investigated whether a tryptophan-rich (Trp-Rich) diet attenuates sterile lung IR injury by increasing microbiota-derived indole metabolites and reprogramming alveolar macrophage (AM) inflammatory responses. METHODS: C57BL/6 mice received isocaloric tryptophan-standard

Investigation of the stool microbiome of short bowel syndrome patients before and after a 24-week treatment with the glucagon-like peptide 2 analog glepaglutide.

BACKGROUND AND AIMS: Glepaglutide is a long-acting glucagon-like peptide 2 (GLP-2) analog under development for the treatment of short bowel syndrome (SBS). Glepaglutide enhances intestinal absorption, which may hypothetically lead to changes in the intestinal microbiota, mainly by slowing gastrointestinal transit time. This study evaluated whether glepaglutide affects bacterial load and composition of the intestinal microbiota in SBS patients enrolled in a phase 3b trial assessing its 24-week efficacy on intestinal wet weight and energy absorption. METHODS: In this single-center, open-label,

Association between ocular surface microbiota species and type 2 diabetes mellitus with or without retinopathy and related clinical parameters.

UNLABELLED: Diabetic retinopathy (DR) is the most prevalent blinding microvascular complication of diabetes mellitus. Although altered ocular surface microbiota (OSM) has been recently linked to type 2 diabetes mellitus (DM) and diabetic retinopathy (DR), its involvement in these diseases remains to be elucidated at the species level. Our current study investigated 92 conjunctival sac swabs from 28 nondiabetic controls (NC) and 64 type 2 DM patients, including 31 with DR and 33 without DR (non-DR, NDR), for species-resolved OSM profiles using 2bRAD-M sequencing, and investigated their correlat

Two-year follow-up of gut microbiota alterations in patients after COVID-19: from the perspective of gut enterotype.

UNLABELLED: Gut microbiota dysbiosis plays a role in the pathogenesis of post-acute coronavirus disease (COVID-19); however, the long-term recovery of the gut microbiota following SARS-CoV-2 infection remains insufficiently understood. In this study, 239 fecal samples were collected from 87 COVID-19 patients during the acute phase, and at 6 months, 1 year, and 2 years post-discharge. An additional 48 fecal samples from non-COVID-19 controls were also analyzed. Gut enterotypes were determined through 16S rRNA sequencing, and dynamic changes from the acute phase through recovery were assessed. C

The impact of early-life exposures on growth and adult gut microbiome composition is dependent on genetic strain and parent- of- origin.

BACKGROUND: Early-life exposure to environmental factors can have long-lasting impacts on offspring health into adulthood and therefore is an emerging public health concern. In particular, the impact of maternal environmental exposures such as diet and antibiotic use on the establishment of the offspring gut microbiome has been recently highlighted as a potential link to disease risk. However, the long-term effects are poorly understood. Moreover, interindividual host genetic differences have also been implicated in modulating the gut microbiome, suggesting that these differences may modulate

Gut microbiota development in very preterm infants following fortification of human milk.

Very preterm infants (VPIs) are born with an immature gut and predisposed to gut microbiota dysbiosis-related diseases, for example, necrotizing enterocolitis. Although fortification of human milk is required for these infants, the optimal fortifier remains uncertain. Bovine colostrum (BC), rich in protein and bioactive components, could serve as an alternative to conventional fortifiers (CF). The gut microbiota (GM) of 225 VPIs fed human milk fortified with either BC or CF (FortiColos study, NCT03537365) was profiled by 16S rRNA gene amplicon sequencing of fecal samples collected before, and

Exploring gut microbiota profile induced by antipsychotics in schizophrenic patients: insights from an Eastern European pilot study.

BACKGROUND: Schizophrenia (SCZ) is an intricate multi-systemic mental illness which has been linked to alterations of the gastrointestinal (GI) microbiota via the bidirectional network known as the gut-brain axis (GBA). However, the specific microbial signatures characterizing SCZ remain unclear. METHODS: We analyzed the gut microbiome composition of 57 Romanian adults, 30 SCZ, and 27 healthy controls (HCs), using amplicon sequencing on the V3-V4 region of the 16S rRNA gene isolated from stool samples. DNA was extracted with an optimized enzymatic and mechanical lysis protocol. Sequence profil

Probiotics to reduce ventilator-associated pneumonia in adults with acute non‑anoxic brain injury: Study Protocol for a Double-Blind Multicenter Randomized International Clinical Trial (PROACT).

BACKGROUND: Ventilator-associated pneumonia (VAP) remains a significant complication among critically ill patients, with associated mortality approaching 50%. Despite the implementation of established preventive strategies, additional interventions are urgently needed to further reduce the incidence of VAP. Probiotic therapy has emerged as a promising adjunctive approach; the benefits of probiotic therapy may be more pronounced in critically ill patients without pre-existing infections. The PROACT study will evaluate the prophylactic role of probiotics in a critically ill population with acute

One-year restoration of vaginal health: synergistic dynamics of microbiome and metabolome following the elimination of high-grade cervical intraepithelial neoplasia.

Therapeutic elimination of high-grade cervical intraepithelial neoplasia (CIN) is widely implemented for cervical cancer prevention. Despite the demonstrated dysbiosis of vaginal microenvironment in high-grade CIN, its post-therapy restorations remain to be poorly understood, especially in functional aspects. This study aimed to characterize temporal changes in both vaginal microbiota (VM) and metabolome (VMeta) following therapeutic elimination of high-grade CIN. We conducted a longitudinal study of 32 HPV-positive women with high-grade CIN who underwent therapeutic procedures. Vaginal swabs

Dynamics of the gut microbiome in subjects challenged with Shigella sonnei 53G in a controlled human infection model.

Shigella is a significant cause of diarrhea, predominantly affecting children in low- and middle-income countries, as well as international travelers. Not all individuals exposed to Shigella or other enteropathogens have symptomatic responses, and investigating the differences between symptomatic and asymptomatic individuals can further our understanding of enteropathogen proliferation and symptomatic responses. Here, we profiled the fecal microbiomes of 45 individuals infected with Shigella sonnei strain 53G through 16S rRNA sequencing in a controlled human infection model before and during i

The Interplay between Gut Microbiota and Cognitive Functioning in the Healthy Aging Population: A Systematic Review.

BACKGROUND: The gut microbiota in healthy older individuals typically show a decrease in beneficial bacteria like Bifidobacterium and Lactobacillus, alongside an increase in pro-inflammatory microbes such as Enterobacteriaceae and Clostridia. These changes contrast with younger and middle-aged individuals and appear to correlate with cognitive status. Although there is extensive research on gut microbiota and cognitive functions in cognitively impaired elderly individuals, its impact on cognitively healthy elderly populations has not been extensively studied. METHOD: A comprehensive literatur

Birth Mode Does Not Determine the Presence of Shared Bacterial Strains between the Maternal Vaginal Microbiome and the Infant Stool Microbiome.

Dysbiosis of the neonatal gut microbiome during early life has been suggested as the missing link that may explain higher rates of certain diseases in caesarean section-delivered infants. Many studies report delivery mode-related dysbiosis in infants due to a lack of maternal vaginal microbiome exposure, prompting interventions to correct the neonatal gut microbiome by transferring these missing microbes after caesarean delivery. The maternal vaginal microbiome is among the first microbial exposures that many infants experience, yet little is known about the extent of direct transmission of ma

Effect of 1-Deoxynojirimycin on insulin resistance in prediabetic mice based on next-generation sequencing and intestinal microbiota study.

ETHNOPHARMACOLOGICAL RELEVANCE: 1-Deoxynojirimycin (DNJ), the major alkaloid in Morus alba L., is the main effective constituent in "Mulberry twig Alkaloids Tablets" launched in China in 2020. Prediabetes, characterized by insulin resistance, is regarded as the key period for reversing Type 2 diabetes mellitus (T2DM) through lifestyle intervention and glucose-lowering drugs. Besides the excellent activity as an α-glucosidase inhibitor, DNJ also improves insulin sensitivity in T2DM murine models, yet the mechanism is still unclear. Besides, the pharmaceutical effect of DNJ on prediabetes is als
