Stable transformation of the yellow fever mosquito, Aedes aegypti, with the Hermes element from the housefly.

The mosquito Aedes aegypti is the world's most important vector of yellow fever and dengue viruses. Work is currently in progress to control the transmission of these viruses by genetically altering the capacity of wild Ae. aegypti populations to support virus replication. The germ-line transformation system reported here constitutes a major advance toward the implementation of this control strategy. A modified Hermes transposon carrying a 4.7-kb fragment of genomic DNA that includes a wild-type allele of the Drosophila melanogaster cinnabar (cn) gene was used to transform a white-eyed recipie

St. Louis encephalitis: clinical and epidemiologic aspects, Mississippi, 1974.

During July, August, and September 1974, 23 confirmed or presumptive cases of St. Louis encephalitis occurred in Mississippi. Attack rates were highest in the northwest corner of the state and among persons over 70 years of age. Rural and urban residents had similar attack rates. Nine persons have not resumed their pre-illness level of activity because of residual neurologic impairment six months after their illness. Additional cases of St. Louis encephalitis are expected in the next few years.

Non-target influence of natural insecticide neem oil on bacterial growth and metabolism.

Although azadirachtin is an insecticide used in organic farming and in medicines that treat diseases caused by insects, its influence on microorganisms, which are not targeted, has never been considered. In this study, the metabolic and enzymatic activities of model bacteria, Escherichia coli and Pseudomonas fluorescens, were investigated in the presence of neem oil at various concentrations of azadirachtin (100 ppm, 500 ppm, 900 ppm), and biodegradation was also explored. Microbial growth and cell activity studies show that neem oil azadirachtin has a negative effect compared to control sampl

Changes in Carabidae Biodiversity in the Perennial Flowering Strip and Adjacent Arable Crops in Organic Conditions.

Landscape simplification resulting from agricultural intensification threatens biodiversity and ecosystem services on arable land. Semi-natural habitats, such as flower strips, can mitigate these effects by providing habitat and resource diversity and by modifying the microclimate at the agricultural landscape scale. As effective natural enemies in arable fields, ground beetles (Carabidae) play a key role not only in pest regulation, but also in weed control through seed consumption. The study found that FS serve as crucial habitats supporting Carabidae-characterized by a higher Shannon-Wiener

Chromosomal-level genome assembly of minute pirate bug Orius nagaii Yasunaga, 1993 (Hemiptera: Anthocoridae).

Species of the genus Orius, diminutive predatory insects that act as natural enemies of other arthropods, are frequently employed in agricultural pest management for controlling various pests, such as thrips, mites, aphids, whiteflies, etc. However, the scarcity of high-quality genomic resources for these predators hinders our comprehension of their population evolution and predation ecology. Consequently, we assembled and annotated a chromosomal-scale genome of Orius nagaii by collating PacBio and Illumina sequencing and Hi-C genomic analysis techniques. The final genome assembly size 152.62

Daily variation of nociceptive responses in Drosophila.

The circadian clock regulates a wide range of physiological and behavioral rhythms. Daily variation in nociceptive responsiveness to harmful stimuli has been demonstrated in mammalian models; however, whether such variation is present in non-mammalian species including insects has remained unclear. Here, we found that behavioral responses to noxious heat stimuli in the fruit fly Drosophila melanogaster exhibit significant variation throughout the day. In contrast, null mutants for the clock genes per and tim failed to exhibit daily variation in nociceptive responses. Consistently, we found dai

Design of Lipopeptides and dsRNA-Lipopeptide Complexes for Sprayable RNAi-Based Plant Protection.

Broad-spectrum insecticides persist in the environment, affecting non-target organisms and selecting for resistant pest populations. In contrast, RNA interference (RNAi) achieves species-targeted pest control triggered by the delivery of exogenous double-stranded RNA (dsRNA), which is biodegradable and broken down into harmless components by nucleases. Sprayable applications require suitable formulation strategies for dsRNA protection during delivery. Here, we describe the preparation of dsRNA-lipopeptide complexes (LPCs) composed of non-toxic short-chain lipopeptide surfactants by microfluidi

The deubiquitinase Usp10 regulates Drosophila reproduction through modulating border cell migration.

Pests are a major cause of agricultural and economic losses. Their high reproduction rate remains a considerable challenge for pest control. Insects with impaired border cell migration during oogenesis are unable to reproduce, offering a potential method to reduce their population growth. To fully understand the effects of the deubiquitinating enzyme (DUB) family on border cell migration, we generated 38 transgenic fly lines overexpressing distinct DUBs, using Gal4/UAS technology to drive their expression in border cells. Overexpression of usp10 led to reductions in ovary size, egg production,

Beyond blood: the flexibility of triatomine bug food search and recognition.

Kissing bugs are known for being skilled blood-feeders on warm-blooded hosts, using cues like carbon dioxide, infrared radiation, and skin odors to locate them. They depend on heat to direct their bites and use gustatory signals for feeding decisions. Recent evidence suggests that they also feed on cold-blooded animals and insects. This implies that they may respond to other sensory cues, such as slight temperature changes, vibrations, and/or odors, to trigger biting attempts. Besides, new studies reveal that triatomines can ingest plant or fruit fluids, showing a broader dietary flexibility t

Antimicrobial Resistance and Extended-Spectrum Beta-Lactamase Production in Enterobacteria Isolated from Free-Living Primates.

The aim of this study was to evaluate the profile of antimicrobial resistance in members of the family Enterobacteriaceae and to detect strains of extended-spectrum beta-lactamases (ESBL) in free-living Sapajus nigritus. Oral and rectal swabs were collected from 15 primates to isolate and identify bacteria, conduct phenotypic tests to detect antimicrobial sensitivity and ESBL-producing strains, and calculate the multiple antimicrobial resistance index. The least effective antimicrobial was amoxicillin (72.72%) and ampicillin (57.57%), and the samples were considered as high risk for public hea

Aquaporins: Novel Targets for Age-Related Ocular Disorders.

Aquaporins (AQPs), a large family of membrane protein channels that facilitate transport of water and other small solutes, play important roles in physiological functions and human diseases. Up till now, 13 types of AQPs, numbered 0 through 12, have been identified in various mammalian tissues. Homologous genes for AQPs in amphibians, insects, and bacteria highlight the evolutionary conservation and, thus, the importance of these membrane channels. Many members of the AQP family are expressed in the eye. AQP1, which is a water-selective channel, is expressed in the anterior chamber (cornea, ci

A desert ant's memory of recent visual experience and the control of route guidance.

Insects such as desert ants learn stereotyped visual routes between their nests and reliable food sites. Studies here reveal an important control element for ensuring that the route memories are used appropriately. They find that visual route memories can be disengaged, so that they do not provide guidance, even when all appropriate visual cues are present and when there are no competing guidance cues. Ants were trained along a simple route dominated by a single isolated landmark. If returning ants were caught just before entering the nest and replaced at the feeder, then they often interrupte

Investigations on the role of a lysozyme from the malaria vector Anopheles dirus during malaria parasite development.

A cDNA encoding a lysozyme was obtained by rapid amplification of cDNA ends-polymerase chain reaction (RACE-PCR) from females of the malaria vector Anopheles dirus A (Diptera: Culicidae). The 623 bp lysozyme (AdLys c-1) cDNA encodes the 120 amino acid mature protein with a predicted molecular mass of 13.4 kDa and theoretical pI of 8.45. Six cysteine residues and a potential calcium binding motif that are present in AdLys c-1 are highly conserved relative to those of c-type lysozymes found in other insects. RT-PCR analysis of the AdLys c-1 transcript revealed its presence at high levels in the

A feedback loop between Wolbachia and the Drosophila gurken mRNP complex influences Wolbachia titer.

Although much is known about interactions between bacterial endosymbionts and their hosts, little is known concerning the host factors that influence endosymbiont titer. Wolbachia endosymbionts are globally dispersed throughout most insect species and are the causative agent in filarial nematode-mediated disease. Our investigation indicates that gurken (grk), a host gene encoding a crucial axis determinant, has a cumulative, dosage-sensitive impact on Wolbachia growth and proliferation during Drosophila oogenesis. This effect appears to be mediated by grk mRNA and its protein-binding partners

Rhythmic arm movement is not discrete.

Rhythmic movements, such as walking, chewing or scratching, are phylogenetically old motor behaviors found in many organisms, ranging from insects to primates. In contrast, discrete movements, such as reaching, grasping or kicking, are behaviors that have reached sophistication primarily in younger species, particularly primates. Neurophysiological and computational research on arm motor control has focused almost exclusively on discrete movements, essentially assuming similar neural circuitry for rhythmic tasks. In contrast, many behavioral studies have focused on rhythmic models, subsuming d

Genetic parameters and pedigree-based selective breeding outcomes for productivity traits in industrial black soldier fly (hermetia illucens L.) production systems.

BACKGROUND: Selective breeding in mass-reared insects has progressed rapidly in recent years, and the black soldier fly (Hermetia illucens L. - Diptera: Stratiomyidae), the most widely cultivated insect species for animal feed, is emerging as a strong candidate for genetic improvement. Estimation of quantitative‑genetic parameters and implementation of pedigree‑based selection schemes are essential to increase genetic gain while controlling inbreeding under industrial rearing. Here, we report a pilot‑scale breeding experiment conducted under family group‑mating with partial pedigree uncertaint

Investigation of the global transportation of Culicoides biting midges, vectors of livestock and equid arboviruses, from flower-packing plants in Kenya.

In recent decades there has been a huge increase in the export of cut flowers from countries in Africa and elsewhere to European flower markets, with the vast majority first entering the Netherlands for local use or for export. Coincidentally, three significant livestock disease outbreaks caused by viruses associated with Africa or other tropical regions were first detected in the Netherlands (bluetongue virus serotype 8 (BTV-8), 2006, and BTV-3, 2023) and in western Germany about 200 km from the Netherlands border (Schmallenberg virus, SBV, 2011). This study aimed to determine whether Culicoi

In vitro control of the camel nasal botfly, Cephalopina titillator, with doramectin, lavender, camphor, and onion oils.

Camels are very important livestock particularly in arid and semiarid lands. The oestrid fly, Cephalopina titillator (Clark), causes nasopharyngeal myiasis in camels, and it is widely distributed in many camel breeding areas triggering health hazards and severe economic losses in camels. The prevalence of infestation of camels (slaughtered at Tokh's slaughterhouse, Qalyubia Governorate, Egypt, during the period from September 2011 to March 2012) was 41.67 % (100 out of 240). Most infested camels developed clinical signs of nasal discharge, restlessness, loss of appetite, difficulty in breathin

Evolutionary analysis of the kinesin light chain genes in the yellow fever mosquito Aedes aegypti: gene duplication as a source for novel early zygotic genes.

BACKGROUND: The maternal zygotic transition marks the time at which transcription from the zygotic genome is initiated and a subset of maternal RNAs are progressively degraded in the developing embryo. A number of early zygotic genes have been identified in Drosophila melanogaster and comparisons to sequenced mosquito genomes suggest that some of these early zygotic genes such as bottleneck are fast-evolving or subject to turnover in dipteran insects. One objective of this study is to identify early zygotic genes from the yellow fever mosquito Aedes aegypti to study their evolution. We are als
