The gastrointestinal tract transports the food bolus by peristalsis. Gut motility starts at an early age in the developing embryo, well before it is required for nutrition of the organism. We present a comprehensive kinematic study of the emergence and physiological development of gut motility in all regions of the lower digestive tract of the chicken embryo from embryonic days E5 through E9. We characterized motility emergence time, propagation patterns, speed, frequency and amplitude of peristalsis waves. We found that the emergence of an uninterrupted circular ring of smooth muscle correlat

Cadherin-11 (Cad-11), a cell cell adhesion molecule belonging to the "classical type II" cadherin family, is a marker of the loosely connected and migratory cellular elements of the mesenchyme. Interestingly, by using in situ hybridization, regional high expression of cad-11 was seen in the brain as well as the spinal cord. We made the following observations in rat embryos and neonates: (1) cad-11 first appears at the lips of the open neural tube; (2) shortly after neural tube closure, cad-11 delineates boundaries in the fore- and midbrain while a metameric signal is detected in the rhombencep

The histogenesis and cell structure progression of Hassall's bodies (HB) was observed employing light and electronmicroscopic histochemical methods in 212 human and 100 dog fetuses at various stages of ontogenesis. Several human pre- and postnatal thymuses were investigated immunohistochemically, employing a library of over 50 poly- and monoclonal antibodies, seeking to determine the cell origin of HBs. The organization of optimal microenvironmental conditions in the thymus requires the added participation of the neural crest. Spontaneous or experimental ablation of the neural crest during ear…

Malignant melanoma is a neoplasm arising from the malignant transformation of melanocytes. These cells are derived from the neural crest and are found predominantly in the skin, but also in mucous membranes, the uveal tract of the eye, the leptomeninges, and other sites. A 39-year-old previously healthy male presented with a holocranial headache for two months and a single episode of a focal seizure involving his left arm. Neurological exam revealed left-sided pronator drift and gross papilledema. Contrast-enhanced magnetic resonance imaging (CE-MRI) of the brain revealed a large (4.8 x 3.9 cm…

Neurogenesis is highly active during brain development but becomes substantially restricted in adulthood. Therefore, the nervous system shows a limited ability to self-renew succeeding a traumatic incident or neurodegenerative disease. Dental stem cells (DSCs) represent a unique population of mesenchymal stem cells originating from the cranial neural crest. Thus, they have an inherent capacity to differentiate into neurons under proper in vitro cues. However, this ability is limited by low conversion rates and unpredictable target cell types, along with a noticeable lack of standardized differ…

OBJECTIVE: To develop an artificial intelligence model based on convolutional neural network for detecting and measuring periodontal radiographic bone loss (RBL).
MATERIALS AND METHODS: Keypoint annotations were carried out in 595 digital bitewing radiographic images using a Computer Vision Annotation Tool. The dataset was splitted: 416 of these images were trained using the You Only Look Once version 8 architecture with pose estimation (YOLO-v8-pose), 119 images were destined for the validation set, and 60 images were used for the test set, resulting in a model capable of detecting keypoints

Pruriception is crucial for defense against external stimuli but can lead to chronic pruritus, a debilitating condition affecting millions w … ranscription factors (TFs) in hPSCs, and a 2-step process through expression of TFs in hPSC-derived neural crest cells (NCCs). The 2-step protocol proved superior in inducing a transcriptional program that closely resembles that of human pruriceptors. Our optimized protocol employs forced expression of NGN1 and ISL1 to drive differentiation from NCCs into prur …

BACKGROUND: Hirschsprung disease (HSCR) is characterized by absence of enteric ganglia (aganglionosis) along variable lengths of the distal intestine. This disorder results from deficient colonization of fetal intestine by enteric neural crest-derived cells (ENCDCs). HSCR exhibits complex, multifactorial inheritance with penetrance and severity varying widely even within a family. SOX10 is among causal genes that predispose to aganglionosis. Yet, how gene interactions influence severity of HSCR aganglionosis is not understood. We previously used an F1-intercross strategy to map genetic modifie…

Allelic variations of Sperm antigen with calponin homology and Coiled-Coil domains 1 Like (SPECC1L) have been associated with a spectrum of cranial-facial pathologies including Teebi hypertelorism and Opitz G/BBB syndrome which manifest as clefting of the palate, wide-eyes, and incomplete closure of the esophagus among others. These pathologies may be indicative of improper cranial neural crest cell (CNCC) delamination and migration. SPECC1L is hypothesized to be an actin-microtubule cross-linking protein as it co-localizes with both microtubules and actin in tissue culture cells. Further, it …

Pheochromocytomas and paragangliomas are rare endocrine neoplasms derived from neural crest cells, with spermatic cord paragangliomas being exceptionally uncommon. A 42-year-old man presented with a longstanding complaint of left scrotal enlargement. Initial imaging raised suspicion of testicular carcinoma, but contrast-enhanced computed tomography and magnetic resonance imaging revealed a well-circumscribed, hypervascular mass distinct from the testis. The tumor exhibited heterogeneous T2 signal intensity, characteristic of vascular lesions, a thick capsule, and early-phase peripheral contras…

BACKGROUND: Retinoic acid (RA) is pivotal for the development of the enteric nervous system (ENS), guiding the migration and differentiation of vagal neural crest cells in the gastrointestinal tract. Despite its recognized influence on other intestinal cell types, the specific roles of RA on these cells are less understood. This study investigates the extensive impact of RA inhibition on the intestinal composition using zebrafish, while also considering its potential use as a model for Hirschsprung disease (HSCR).
METHODS: Zebrafish larvae were treated with diethylaminobenzaldehyde (DEAB), an…

Craniofacial reconstruction faces many challenges, including high complexity, strong specificity, severe injury, irregular and complex wounds, and high risk of bleeding. Traditionally, the "gold standard" for treating craniofacial bone defects has been tissue transplantation, which involves the transplantation of bone, cartilage, skin, and other tissues from other parts of the body. However, the shape of craniofacial bone and cartilage structures varies greatly and is distinctly different from ordinary long bones. Craniofacial bones originate from the neural crest, while long bones originate f…

The destruction of periodontal alveolar bone (AB) caused by periodontitis is regarded as one of the major reasons for tooth loss. The inhibi … n hBMSC-CM in treating ABD was attributed to the cell compatibility as both AnSCs and AB tissue are neural crest-derived. In conclusion, the effects of AnSC-CM on AB tissue regeneration were achieved through both promotion of osteogenesis and inhibition of osteoclastogenesis. We believe that AnSC-CM is a candidate for effective treatment of ABD in dental clini …

INTRODUCTION AND IMPORTANCE: Paraganglioma (PG); also known as Glomus Tumor, is a rare neoplastic entity that originates from neural crest cells. It can manifest in different patterns, chiefly benign but others are locally invasive and malignant in behavior. Due to the prevalence of other more common types of neck masses and Paraganglioma's exceeding rarity, misdiagnoses can easily take place, and this results in higher patient morbidity and mortality rates. Preoperative diagnosis constitutes a major clinical challenge, especially in patients with a previous surgical history in the neck region…

Meningiomas are the most common primary central nervous system tumors. These tumors predominantly arise from the neural crest-derived meningothelial cells of the arachnoid dural layer. Intracranial meningiomas are stratified with the World Health Organization classification of tumors. Cutaneous meningiomas present rarely and have their own criteria classification (Lopez classification) of 3 types. The first type is congenital. The second consists of ectopic soft-tissue meningiomas. The third involves tumors that extended into the dermis or subcutis that include the neuroaxis. We present a case…
