Antonie van Leeuwenhoek (1632-1723) transformed observation into science through the power of a single handmade lens. His work emerged from the visual culture of seventeenth-century Delft, where craftsmanship, optics, and artistic precision intersected. While Robert Hooke's compound microscope introduced the idea of microscopic visualization, Leeuwenhoek's single-lens instruments achieved far superior magnification and resolution by minimizing optical interfaces. Using these deceptively simple devices, he documented the first observations of free-living microorganisms, fungal hyphae, red blood

BACKGROUND: Integrated pest management (IPM) is thought to improve asthma symptoms through reduced mouse allergen exposure. Whether IPM acts through changes in mouse-associated microbes remains unknown.
OBJECTIVES: We examined the effects of school-based IPM on the classroom microbiome and sought to determine the association between intervention microbiome signatures and student asthma morbidity.
METHODS: In this ancillary study based on a randomized placebo-controlled clinical trial of school IPM and classroom high-efficiency air purifiers (ClinicalTrials.gov NCT02291302), we performed deep

Biofilm-associated infections represent a major clinical and biological challenge due to their ability to persist within host tissues while evading immune clearance and antimicrobial therapy. These structured microbial communities profoundly alter host immune signaling, particularly cytokine and chemokine networks, leading to sustained inflammation and tissue damage. Despite advances in antimicrobial development, biofilms continue to undermine treatment efficacy by promoting antibiotic resistance, dysregulated gene expression, and chronic inflammatory states, especially in wounds, implanted me

Vulvovaginal candidiasis (VVC) is a common vaginal disease characterized by inflammatory cell infiltration and vaginal microbiota dysbiosis. In this study, Limosilactobacillus fermentum 50 not only exhibited the strongest growth-inhibitory activity against Candida albicans using a spot assay, but also significantly suppressed hyphal formation and decreased the expression of virulence-associated genes, including HWP1, ALS3, and ECE1. Furthermore, the effects of L. fermentum 50 on C. albicans-induced VVC were investigated in a mouse model. Oral administration of L. fermentum 50 increased body we

We investigated the activity of a series of 75 lipophilic bisphosphonates against Candida glabrata. Thirty-six compounds had MIC < 1 μg/mL, 18 had MIC < 0.5 μg/mL, and 2 had MIC = 0.13 μg/mL, comparable to amphotericin B and caspofungin. The lipophilic bisphosphonates were ∼20-fold more potent against C. glabrata than the most potent hydrophilic bisphosphonate, zoledronate. The most active compounds were pyridinium bisphosphonates followed by imidazolium bisphosphonates, while aryl bisphosphonates were less active. Several compounds had selectivity index values against a human cell line in the

Fungal and oomycete phytopathogens account for 10%-20% of annual global crop losses, and the escalating resistance to synthetic fungicides has intensified demand for plant-derived alternatives. The essential oil (EO) of Clinopodium nepeta (L.) Kuntze, obtained by hydrodistillation from aerial parts harvested in northeastern Morocco, was profiled by gas chromatography-mass spectrometry (GC-MS) and evaluated for antifungal and anti-oomycete activity against Verticillium dahliae, Phytophthora infestans, and Macrophomina phaseolina using broth microdilution minimum inhibitory concentration (MIC) a

In vivo confocal microscopy (IVCM) is a critical ophthalmic examination that provides in vivo cytological and neurological information essential for diagnosing corneal and certain systemic diseases, but its clinical utility is limited by time-consuming interpretation and the need for subspecialty expertise. We developed IVCM-Insight, an artificial intelligence (AI) system integrating image-text contrastive learning with large language models (LLMs) for automated report generation and interactive question answering (QA). Based on 30,368 IVCM images and 4155 paired clinical reports, the model wa

BACKGROUND: Fungal infections are increasingly recognized as a major public health challenge, and superficial diseases such as otomycosis remain clinically consequential because persistence, recurrence, and treatment failure are common when the causative species and susceptibility profile are not clearly defined. However, otomycosis associated isolates are often insufficiently characterized at the genomic level. Furthermore, the development of antifungal candidates, particularly for short antimicrobial peptides, remains heterogeneous in terms of the integration of efficacy, biosafety, and mech

Gpi7 gene, encoding the catalytic subunit of GPI ethanolamine-phosphate (Etn-P) transferase II, is primarily involved in the synthesis, maturation, and sorting of GPI-anchored proteins (GPI-APs), thereby playing a crucial part in cell wall functions and host-pathogen interactions. This study aimed to investigate the role of Gpi7 (here designated CcGpi7) in Corynespora cassiicola, a devastating fungal pathogen causing leaf spot in cucumber and many other cash crops. We systematically identified and inventoried 138 GPI-APs in C. cassiicola, followed by a detailed structural characterization of t

Hydrogen sulfide (H2S) is a key gaseous signaling molecule involved in plant growth and stress responses, yet its role in wheat resistance to stripe rust remains poorly understood. Here, we show that exogenous H2S enhances resistance of wheat (Triticum aestivum L.) to Puccinia striiformis f. sp. tritici (Pst), the causative agent of stripe rust. Comparative persulfidation proteomics identified the autophagy-related protein TaATG6c as a Pst-responsive H₂S target. Site-specific mass spectrometry and a modified biotin-switch assay demonstrated that Cys177 and Cys180 of TaATG6c undergo H₂S-induced

Amanita muscaria contains two structurally related neuroactive isoxazole metabolites, ibotenic acid (IA) and muscimol (MUS), which differ markedly in pharmacological activity. IA acts as an agonist at NMDA and metabotropic glutamate receptors, whereas MUS is a potent GABAergic agonist. This review critically evaluates their chemistry and biosynthesis, biological variability, post-harvest transformation, analytical determination, and toxicological significance. Direct evidence demonstrates interspecific variation within Amanita section Amanita. Developmental evidence for A. muscaria is limited

Allergic bronchopulmonary aspergillosis (ABPA) is a complex hypersensitivity lung disease driven predominantly by type 2 immune responses. However, the classical immunological paradigm fails to adequately explain the persistent airway structural damage and frequent relapses observed even after inflammatory control is achieved. This review proposes a fibroblast-centered mechanistic framework that synthesizes evidence from ABPA, fungal-associated airway diseases, and pulmonary fibrosis to systematically delineate the mechanisms linking fibroblasts to chronic inflammation and airway remodeling. Aspergillus fumigatus is the primary pathogen causing aspergillosis. Recent molecular population genetic studies have demonstrated that A. fumigatus exhibits high local genetic diversity, with evidence for limited differentiation among geographic populations. However, research on the impacts of geomorphological factors on shaping the population genetic diversity patterns of this species remains scarce. In this study, large-scale sampling and in-depth population genetic analysis were performed on soil-derived A. fumigatus from Guizhou Province, a representative karst landscape in southern China

The rapid emergence of multidrug-resistant microbial pathogens has created an urgent need for new antimicrobial agents. Marine organisms are a rich source of structurally diverse bioactive metabolites with therapeutic potential, and the sea cucumber H. scabra contains numerous secondary metabolites that may exhibit favorable predicted interactions with antimicrobial targets. This study aimed to identify potential antimicrobial metabolites from H. scabra using an integrated computational approach. Metabolites were identified by gas chromatography-mass spectrometry (GC-MS) and screened against s

Nearly all known cases of antimicrobial resistance have been linked to agents that directly target microbes. This paradigm overlooks other drivers of microbial evolution and resistance. Here, we report compelling evidence of resistance-associated mutations in Pneumocystis jirovecii, a high-priority fungal pathogen designated by WHO as having unknown resistance. These mutations emerged under selective pressure from mycophenolic acid (MPA), a widely used immunosuppressant not used for Pneumocystis pneumonia (PCP) treatment. A potent inhibitor of inosine monophosphate dehydrogenase (IMPDH), MPA h
