Genome-Wide Investigation Reveals Potential Therapeutic Targets in Shigella spp.

Shigella stands as a major contributor to bacterial dysentery worldwide scale, particularly in developing countries with inadequate sanitation and hygiene. The emergence of multidrug-resistant strains exacerbates the challenge of treating Shigella infections, particularly in regions where access to healthcare and alternative antibiotics is limited. Therefore, investigations on how bacteria evade antibiotics and eventually develop resistance could open new avenues for research to develop novel therapeutics. The aim of this study was to analyze whole genome sequence (WGS) of human pathogenic Shi

Community acquired methicillin resistant Staphylococci (CA-MRS) in fecal matter of wild birds - A 'one health' point of concern.

BACKGROUND: Antibiotic resistance in Staphylococci, particularly methicillin resistance is a major public health concern. While this problem has been reported from the clinical settings, its presence in non-clinical settings also needs to be investigated. The role of wildlife in carrying and disseminating the resistant strains has been established in different studies but its role in Pakistani environment has not been explored yet. To evaluate this, we investigated the carriage of antibiotic resistant Staphylococci in wild birds from Islamabad region. METHODOLOGY: Birds fecal matter were coll

Linezolid-resistant Staphylococcus aureus strain 1128105, the first known clinical isolate possessing the cfr multidrug resistance gene.

The Cfr methyltransferase confers resistance to six classes of drugs which target the peptidyl transferase center of the 50S ribosomal subunit, including some oxazolidinones, such as linezolid (LZD). The mobile cfr gene was identified in European veterinary isolates from the late 1990s, although the earliest report of a clinical cfr-positive strain was the 2005 Colombian methicillin-resistant Staphylococcus aureus (MRSA) isolate CM05. Here, through retrospective analysis of LZD(r) clinical strains from a U.S. surveillance program, we identified a cfr-positive MRSA isolate, 1128105, from Januar

Functional C3435T polymorphism of MDR1 gene: an impact on genetic susceptibility and clinical outcome of childhood acute lymphoblastic leukemia.

The significance of genetic background in childhood acute lymphoblastic leukemia (ALL) is not well understood. Polymorphisms of genes encoding for xenobiotics and drug transporters are potential factors, which can influence the risk of developing ALL and its clinical outcome. P-glycoprotein (P-gp) is an adenosine triphosphate-binding cassette (ABC)-family transporter involved in protection against xenobiotics and multi-drug resistance. Recently, the single-nucleotide polymorphism C3435T of MDR1 gene has been found to be associated with altered tissue expression and function of P-gp. To evaluat

Coexpression of p53 protein and MDR functional phenotype in leukemias: the predominant association in chronic myeloid leukemia.

BACKGROUND: One of the best characterized resistance mechanisms of leukemias is multidrug resistance (MDR) mediated by P-glycoprotein (Pgp) and multidrug-resistant related protein (MRP). In addition to Pgp and MRP, p53 mutation or inactivation might play a relevant role in therapeutic failure. Some studies have demonstrated that Pgp and MRP may be activated in association with overexpression of mutant or inactivated p53 protein. The aim of this study was to investigate the association between p53 expression and MDR functional phenotype analyzed by flow cytometry (FCM). METHODS: Rhodamine-123

Reversion of trichostatin A resistance via inhibition of the Wnt signaling pathway in human pancreatic cancer cells.

Drug resistance is a major impediment to successful chemotherapy in pancreatic cancer (PC) patients. We investigated the effect of Wnt/β-catenin signaling inhibition by wnt-c59 on chemoresistance in a trichostatin A-resistant Panc-1 cell line (Panc-1/TSA). Panc-1/TSA cells were treated with the Wnt/β‑catenin signaling inhibitor wnt-c59 (10 µmol · l-1) and/or trichostatin A (TSA; 10 µmol · l-1) for 24 h. CCK-8 assay was utilized to analyze the interactive effect of TSA and wnt-c59 on induction of apoptosis of the Panc-1/TSA cells. Cell apoptosis was measured by flow cytometry. Real-time PCR and

Beyond bactericidal: targeting plasmid-mediated antibiotic resistance with natural product-based plasmid curing agents.

Antimicrobial resistance (AMR) has evolved into a severe global public health crisis, with plasmid-mediated horizontal gene transfer (HGT) serving as a core driver for the rapid dissemination of multidrug resistance (MDR). Traditional "bactericidal" antibiotic strategies impose strong selective pressure, failing to eradicate the root cause of resistance while accelerating the enrichment of resistant clones. "Plasmid curing"-a strategy that specifically eliminates resistance plasmids to restore antibiotic susceptibility-has emerged as a promising paradigm shift. While early synthetic curing age

Impact of CD44 alternative splicing on the response to anticancer drugs.

The transmembrane glycoprotein CD44, a marker of cancer stem cells (CSCs), plays a role in tumor initiation, progression, and multidrug resistance (MDR) phenotype in many cancers. The functional diversity of CD44 is determined by alternative splicing, which generates variant isoforms (CD44v) from the single gene CD44. This transcriptomic plasticity is not only structural but also governs tumor cell metabolic adaptations, phenotypic heterogeneity, and sensitivity to anticancer therapies. The balance between the standard isoform (CD44s) and CD44v is frequently altered in malignant cells and has

In vitro and In vivo evaluation of phage-antibiotic synergy for the treatment of urinary tract infections.

The therapeutic challenges caused by multidrug-resistant (MDR) and extensively drug-resistant (XDR) bacteria have necessitated the development of alternative treatment strategies. Phage-antibiotic synergy (PAS) has recently emerged to replace or possibly supplement antibiotics. We isolated the lytic phage NTNC80A from hospital sewage in Chandigarh, India, and it belongs to the class Caudoviricetes of viruses. We tested multiple strategies to investigate the synergistic effects of phage and antibiotic combinations against highly drug-resistant uropathogenic Escherichia coli isolates. The maximu

Strategic design of a multi-tier database for class A β-lactamase BlaC variants of M. tuberculosis: advancing the fight against antibacterial resistance.

The escalating rise of antimicrobial resistance (AMR) casts a grave shadow over global public health, making once manageable infections increasingly difficult to treat. Despite advancements in combination chemotherapy for multidrug-resistant (MDR) and extensively drug-resistant (XDR) Mycobacterium tuberculosis (TB), this pathogen remains a formidable foe. TB is now the second leading cause of death worldwide from infectious diseases, only surpassed by COVID-19. It is the primary driver of AMR-related deaths, particularly among HIV co-infected individuals. A significant challenge lies in TB's r

Structural and Antibacterial Characterization of a New Benzamide FtsZ Inhibitor with Superior Bactericidal Activity and In Vivo Efficacy Against Multidrug-Resistant Staphylococcus aureus.

Methicillin-resistant Staphylococcus aureus (MRSA) is a multidrug-resistant (MDR) bacterial pathogen of acute clinical significance. Resistance to current standard-of-care antibiotics, such as vancomycin and linezolid, among nosocomial and community-acquired MRSA clinical isolates is on the rise. This threat to global public health highlights the need to develop new antibiotics for the treatment of MRSA infections. Here, we describe a new benzamide FtsZ inhibitor (TXH9179) with superior antistaphylococcal activity relative to earlier-generation benzamides like PC190723 and TXA707. TXH9179 was

Structural and Dynamic Insights into the W68L, L85P, and T87A Mutations of Mycobacterium tuberculosis Inducing Resistance to Pyrazinamide.

Tuberculosis (TB), the most frequent bacterium-mediated infectious disease caused by Mycobacterium tuberculosis, has been known to infect humans since ancient times. Although TB is common worldwide, the most recent report by the WHO (World Health Organization) listed the three countries of India, China, and Russia with 27%, 14%, and 8% of the global burden of TB, respectively. It has been reported that resistance to TB drugs, particularly by the pncA gene to the pyrazinamide drug due to mutations, significantly affects the effective treatment of TB. Understanding the mechanism of drug resistan

ABCB1 regulation through LRPPRC is influenced by the methylation status of the GC -100 box in its promoter.

One of the potential mechanisms of imatinib mesylate (IM) resistance in chronic myeloid leukemia (CML) is increased level of P-glycoprotein (Pgp). Pgp is an efflux pump capable of activating the multidrug resistance (MDR) phenotype. The gene encoding Pgp (ABCB1) has several binding sites in its promoter region, along with CpG islands and GC boxes, involved in its epigenetic control. In previous work, we performed a proteomic study to identify proteins involved in IM cross-resistance in acute leukemia. Among these proteins, we identified LRPPRC as a potential regulator of ABCB1 transcription vi

The histone deacetylase inhibitor AN-9 has selective toxicity to acute leukemia and drug-resistant primary leukemia and cancer cell lines.

The novel prodrug of butyric acid, pivaloyloxymethyl butyrate (AN-9), a histone deacetylase inhibitor, shows great promise as an effective and relatively nontoxic anticancer agent for solid malignancies. However, little is known about its effects on hematopoietic malignancies. In this study, we show that 21 primary samples of acute leukemia were sensitive to the antiproliferative effects of AN-9, with a 50% inhibitory concentration (IC(50)) of 45.8 +/- 4.1 microM. In colony-forming assays, primary T-cell acute lymphoblastic leukemia (T-ALL) cells were 3-fold more sensitive to AN-9 than the nor
