Methylmercury (MeHg) is a widespread environmental and food toxicant which has long been known to affect neurodevelopment in both humans and experimental animals. Risk assessment for MeHg is mainly based on human data coming from the massive episodes of poisoning in Japan and Iraq, as well as from large scale epidemiological studies concerning childhood development and neurotoxicity in relation to in utero exposure in various fish eating communities around the world. Despite the extensive literature and research, the threshold dose for MeHg neurotoxic effects is still unclear, in particular wh

INTRODUCTION: Exposure to methylmercury (MeHg) before birth can adversely affect children's neurodevelopment. The most common form of prenatal exposure is maternal fish consumption, but whether such exposure harms the fetus is unknown. We aimed to identify adverse neurodevelopmental effects in a fish-consuming population.
METHODS: We investigated 779 mother-infant pairs residing in the Republic of Seychelles. Mothers reported consuming fish on average 12 meals per week. Fish in Seychelles contain much the same concentrations of MeHg as commercial ocean fish elsewhere. Prenatal MeHg exposure w

A current hypothesis about methylmercury (MeHg) neurotoxicity proposes that neuronal damage is due to excitotoxicity following glutamate uptake alterations in the astrocyte. By sampling from a microdialysis probe implanted in the frontal cortex of adult Wistar rats, we measured the effects of acute exposure to either 10 or 100 microM MeHg through the microdialysis probe, on glutamate extracellular levels in 15 awake animals. After baseline measurements, the perfusion of MeHg during 90 min induced immediate and significant elevations in extracellular glutamate at 10 microM (9.8-fold, P<.001) an

The effect of methylmercury on glutamate metabolism was studied by (13)C magnetic resonance spectroscopy. Cerebral cortical astrocytes were pretreated with methylmercury, either 1 microM for 24 h, or 10 microM for 30 min, and subsequently with 0.5 mM [U-(13)C]glutamate for 2 h. Labeled glutamate, glutamine, aspartate and glutathione were present in cell extracts, and glutamine, aspartate and lactate in the medium of all groups. HPLC analysis of these amino acids showed no changes in concentrations between groups. Surprisingly, the amounts of [U-(13)C]glutamate and unlabeled glucose taken up by

The Seychelles Child Development Study has been unable to confirm any relationship between maternal exposure to MeHg during pregnancy and adverse developmental outcomes. In this report, 87 children from a pilot cohort were evaluated at 9 years of age. Each child was given a battery testing specific cognitive, visual motor, and motor skills using standardized psychometric and neuro-psychological tests. The results indicated no adverse association between maternal MeHg exposure and any developmental outcome measure. For three endpoints (Boston Naming Test and two tests of visual motor coordinati

Suriname is experiencing a revival of small scale gold mining activities, with about 10,000 to 15,000 workers involved in 1996. The estimated production in 1995 is at least 10,000 kg crude gold. Gold is extracted with mercury and methods used are comparable with those described for gold mining in the Amazon Basin. Since no data exist on the internal mercury exposure of workers in Suriname a study was performed. A group of mercury-exposed Maroons, who are principally involved in the mining located in the tropical rainforest, is compared with nonexposed Maroons living in a non-gold mining area.

Increased consumption of lake fish was associated with higher blood mercury concentrations among 289 residents of Southwest Quebec living on the southern bank of the Upper St. Lawrence River System. This increase in blood mercury was reflected in the organic fraction and to a lesser extent with total blood mercury. Repeat analyses on a small subpopulation showed a correlation coefficient between time 1 and time 2 of approximately 0.7 for total mercury and for organic mercury. With one exception, blood total mercury concentrations were less than 5 ppb. Blood mercury concentrations were higher a

Many studies have demonstrated mercury contamination in the Amazonian ecosystem, particularly in fish, a dietary mainstay of populations in this region. The present study focused on potential health effects of this low-level methylmercury exposure. The study was carried out in a village on the Tapajós River, a tributary of the Amazon, on 91 adults inhabitants (15-81 years), whose hair mercury levels were inferior to 50 mu/g. Performance on a neurofunctional test battery and clinical manifestations of nervous system dysfunction were examined in relation to hair mercury concentrations. Near visu

Astrocytes are known to express metallothionein (MT) and were studied in culture to determine whether MT could be directly induced and which isoforms are induced. Primary astrocyte cultures were established from neonatal CF-1 mice. Both concentration-response and time-course analyses for MT induction at the protein level were determined. At the mRNA level, induction of MT-I, -II, and -III was examined 6 hr following the addition of the inducing agents. Dexamethasone (Dex), cadmium (Cd), mercury (Hg), or zinc (Zn) increased (three- to fourfold) MT protein in the astrocytes, whereas methyl mercu

Aquatic food chain mercury pollution is one of the consequences of the gold rush in the Amazon, which started in the late 1970s. This paper addresses the risks of methylmercury (MeHg) toxicity by a riverside population of heavy fish eaters along the Madeira river, in the Amazon, based on their hair mercury (Hg) concentration. Given the vulnerability of the developing nervous system, NOEL/LOEL values were used based on prenatal (LOELp = 0.7 microgram/ kg bw), and adult and childhood (LOELa = 3 micrograms/kg bw) Hg exposures. Based on hair Hg concentrations, we observed that approximately 95% of

Mercuric chloride (MC) and methylmercury (MeHg) were found to increase amino acid release from astrocytes. This suggests interaction with sulfhydryl (-SH) groups which are controlled by glutathione [GSH] levels. In the present study, we evaluated the effects of alterations in intracellular glutathione concentrations [GSH]i on the outcome of MC and MeHg treatment. [GSH]i were increased in a time-dependent fashion by incubating the astrocytes with 1 mM L-2-oxothiazolidine-4-carboxylic acid (OTC), a cysteine precursor. OTC attenuated the release of [2,3-3H]D-aspartic acid from astrocytes exposed

Sprague-Dawley rats were exposed to low doses of methyl mercury (3.9 mg mercury/kg diet), via their dams during gestation and lactation and directly via their diet until sacrifice at 50 days postpartum, in order to study possible detrimental effects on CNS development. The methyl mercury exposure of the rats resulted in a brain concentration of 1.45 +/- 0.06 mg mercury/kg wet weight (mean +/- SEM). No general toxic effects were observed; body weight was not affected, brain weight was only slightly increased. No discernible general morphological alterations were seen in the brain as evaluated u

On day 15 of gestation, pregnant Sprague-Dawley rats were orally treated by gavage with 8 mg/kg of methyl mercury (MMC). At day 1 of postnatal life the levels of MMC in whole brain of exposed pups were found to be about 100 times higher than those of saline-exposed rats, while they were near to the control values at 21 days and practically normal at 60 days of age. Behavioral experiments showed that exposure to MMC in late gestation did not affect at any tested time (14, 21 and 60 days) locomotor activity or development of ultrasonic vocalization. An increased response to a challenge dose of a

Both Hg2+ and CH3Hg+ exert a depressive action on the twitch amplitude elicited by nerve stimulations more rapidly than that by muscle stimulation. CH3Hg+ was relatively more potent than Hg2+ in the nerve depressive action but was weaker on the muscle. Hg2+ induced a rapid phase and a slow phase of contracture in the diaphragm, while CH3Hg+ induced only a slow one. The biphasic contracture induced by Hg2+ could be inhibited by repetitive washout of the diaphragm with low Ca2+ (10(-3) mM) Krebs and also by treatment with glycerol to close T-tubule, while the slow contracture induced by CH3Hg+ w

The effects of methylmercury hydroxide (MeHgOH) on the hypnotic action of hexobarbital was investigated in adult 129/SvSl mice of both sexes. It was found that there was no sex difference in the response to MeHgOH treatment. The action of MeHgOH was intimately related to the interval between MeHgOH administration and the test of hexobarbital hypnosis. A biphasic effect was observed. An initial dose-dependent prolongation of hexobarbital hypnosis was observed in animals pretreated with MeHgOH 24 h earlier. If the interval was extended from 24 h to 1 week, a shortening of sleeping time was obser

Methylmercury (MeHg) is a ubiquitous environmental toxicant to which humans can be exposed by ingestion of contaminated food. MeHg has been suggested to exert its toxicity through its high reactivity to thiols, generation of arachidonic acid and reactive oxygen species (ROS), and elevation of free intracellular Ca(2+) levels ([Ca(2+)](i)). However, the precise mechanism has not been fully defined. Here we show that phosphatidylcholine-specific phospholipase C (PC-PLC) is a critical pathway for MeHg-induced toxicity in MDCK cells. D609, an inhibitor of PC-PLC, significantly reversed the toxicit

Cardiovascular disease is the leading cause of morbidity, mortality, and health care costs in the USA, and around the world. Among the various risk factors of cardiovascular disease, environmental and dietary exposures to methyl mercury, a highly toxic metal traditionally labeled as a neurotoxin, have been epidemiologically linked to human cardiovascular disease development. However, its role in development and promotion of atherosclerosis, an initial step in more immediately life-threatening cardiovascular diseases, remains unclear. This study was conducted to examine the role that methyl mer

(-)-Epigallocatechin-3-gallate (EGCG) found in tea is a natural activator of nuclear factor erythroid 2-related factor 2 (Nrf2), a primary regulator of the cellular defense system. The adverse health effects resulting from methylmercury (MeHg) exposure in humans are of worldwide concern. We hypothesized that EGCG could induce a Nrf2-mediated protective response to antagonize MeHg toxicity. Using the Caenorhabditis elegans (C. elegans) nematode model, we observed that EGCG activated SKN-1 (the functional ortholog of Nrf2 in C. elegans), as shown by the increased skn-1 mRNA level, induction of t
