An essential role for interleukin-5 and eosinophils in helminth-induced airway hyperresponsiveness.

Infection with the parasitic helminth Brugia malayi can result in development of a severe asthmatic response termed tropical pulmonary eosinophilia. This disease, thought to result from a host inflammatory response to blood parasites which become trapped in the lung microvasculature, is characterized by a profound eosinophilic infiltration into the lungs. Recruitment of eosinophils also correlates with the development of airway hyperresponsiveness (AHR) to cholinergic agonists and severe asthmatic symptoms. Our studies examined the role of interleukin-5 (IL-5) in helminth-induced pulmonary eos

P2RY13-Negative Tumor-Associated Macrophages Promote NETosis and Are Associated with Poor Prognosis Across Multiple Cancers.

Background: Neutrophil extracellular traps (NETs) promote tumor progression and immune evasion across multiple cancers. However, the mechanisms governing NET formation (NETosis) within the tumor immune microenvironment remain incompletely understood. Low P2RY13 expression is associated with the pro-tumor activity of neutrophils in lung adenocarcinoma. Methods: Pan-cancer bioinformatics analyses were performed, including differential gene expression, prognostic, tumor-infiltrating immune cell, and NET scoring analyses. Single-cell profiling was conducted using the Tumor Immune Single-cell Hub 2…

Single-cell and spatial transcriptomics reveal that the CXCL12-CXCR4 axis drives the immune-desert phenotype in small cell lung cancer by recruiting immunosuppressive CXCR4+ neutrophils and S100A8+ monocytes.

BACKGROUND: Small cell lung cancer (SCLC) is a recalcitrant malignancy with limited responses to immunotherapy, largely due to its uniquely … ith increased infiltration of C-X-C motif chemokine receptor 4 (CXCR4)+ neutrophils (via neutrophil extracellular traps) and S100A8+ monocytes (toward an M2-like phenotype), and reduced CD8+ T-cell infiltration. Malignant epithelial cells in SCLC highly expressed CXCR4, regulated by transcription factors ISL LIM homeobox 1 and distal-less homeobox 5, which promoted i …

Single-cell transcriptomics unveils the immunologic landscape of anti-PD-1-associated indirect drug-induced liver injury.

BACKGROUND: Immune-related adverse events (irAEs) caused by immune checkpoint inhibitors (ICIs) have become a bottleneck limiting their wide … aling axes. Corroborated by validation in both animal and patient tissues, we identified neutrophil extracellular traps (NETs) as a pivotal event in αPD-1-associated liver injury. Modified Dachengqi Decoction ameliorates sepsis-induced lung injury via the gut microbiota-bile acid axis.

BACKGROUND: Sepsis-induced acute lung injury (SI-ALI) is associated with high mortality. The gut microbiota-bile acid axis plays a critical role in regulating host inflammatory responses; however, the mechanism of action of traditional Chinese medicine (TCM) compounds targeting this axis remains unclear. AIM: This study aimed to systematically evaluate the protective effects of Modified DaChengqi Decoction (MDD) against lipopolysaccharide (LPS)-induced SI-ALI and to elucidate its underlying mechanism in modulating inflammation and neutrophil extracellular traps (NETs) through the regulation o…

Targeting TKT-associated immunometabolic remodeling attenuates experimental lupus nephritis and NET-related inflammation.

Lactic acid metabolism and neutrophil extracellular traps (NETs) are critical immune regulators, yet their specific crosstalk in systemic lupus erythematosus (SLE) and lupus nephritis (LN) pathogenesis remains poorly understood. Here, by integrating bulk and single-cell RNA sequencing (scRNA-seq) analyses, we identified TKT and ITGAM as pivotal upregulated genes in SLE, demonstrating robust diagnostic value (AUC >0.7) supported by reliable nomogram models. Additional analysis of a human LN renal biopsy dataset (GSE32591) showed increased ITGAM and TKT in LN glomerular samples, supporting their…

From acute infection to chronic osteoimmune remodeling: immunopathogenesis and precision immunotherapeutic strategies in osteomyelitis.

Osteomyelitis is a severe infectious and inflammatory disease that significantly affects bone tissue. It progresses from an acute inflammatory response to refractory chronic osteomyelitis and bone defects, constituting a core challenge in current clinical therapeutics and functional reconstruction. In the acute osteomyelitis stage, innate immune cells such as neutrophils, monocytes-macrophages, and dendritic cells are rapidly recruited and release large amounts of cytokines, chemokines, and extracellular traps. These work in synergy with adaptive immune responses, including T/B cells, to form …

Serum citrullinated histone H3 as a biomarker of disease activity and renal outcome in microscopic polyangiitis and granulomatosis with polyangiitis.

BACKGROUND: Citrullinated histone H3 (CitH3), a hallmark component of neutrophil extracellular traps, has emerged as a circulating indicator of pathological neutrophil activation. We evaluated the utility of serum CitH3 as a biomarker for disease activity and risk of end-stage kidney disease (ESKD) development in patients with microscopic polyangiitis (MPA) and granulomatosis with polyangiitis (GPA). METHODS: A total of 65 patients with MPA and GPA were included. At diagnosis, serum CitH3 levels were measured and disease activity was assessed using the Birmingham Vasculitis Activity Score (BVA…

Interleukin-8 and other neutrophil-related biomarkers are associated with 1-year mortality in patients with acute myocardial infarction.

BACKGROUND: Interleukin-8 (IL-8) triggers early inflammatory responses upon tissue injury, activating neutrophils that release neutrophil extracellular traps with cell-free DNA (cfDNA) and myeloperoxidase (MPO). Injury upon acute myocardial infarction (AMI) induces an early excessive inflammatory response. The value of IL-8 and neutrophil-related biomarkers for AMI patient outcomes is incompletely understood. METHODS: AMI patients sampled ≤ 24 h after symptom onset were included from SPUM-ACS cohort. One hundred twelve cases with all-cause mortality or recurrent AMI within 1 year were matched…

Innate Immune Function of Neutrophil Cytoplasts Generated Post-Vital NETosis.

The migration of polymorphonuclear neutrophils (PMN) into the site of infection such as lungs during pneumonia is a canonical feature of innate immunity. Formation of neutrophil-derived extracellular traps (NETs), web-like strands of varying lengths comprising DNA, histones, elastase, and myeloperoxidase, is an important determinant of PMN-mediated innate immunity. NETs form in microvessels, entrap bacteria and effete matter, and dampen PMN-mediated inflammatory injury at specific sites. However, studies have largely focused on NET release secondary to lytic NETosis and lysis of PMN. Far less …

Porphyromonas gingivalis-Induced NETs Mediate Neuroinflammation via TLR4 Activation.

Periodontitis, caused by periodontal pathogens such as Porphyromonas gingivalis, is a risk factor for Alzheimer's disease (AD) progression. Neutrophils, which are abundant in patients with periodontitis, release neutrophil extracellular traps (NETs) to resist microbial infection. We explored the mechanism and effects of P. gingivalis-induced NETs on the relationship between periodontitis and neuroinflammation. A murine periodontitis model was established via oral local application of P. gingivalis with or without tak242 (TLR4 inhibitor). Maxillary bones were evaluated via micro-computed tomogr…

Humanized Monoclonal Antibody Against Citrullinated Histone H3 Attenuates Myocardial Injury and Prevents Heart Failure in Rodent Models.
