The morphological transition of fungi from vegetative hyphae to thick-walled chlamydospores enhances their longevity in harsh environmental conditions. Owing to this resilience, pathogenic fungi that form chlamydospores are particularly difficult to control. Therefore, understanding the mechanisms of chlamydospore formation is critically important. Here, we show that the hyphae of the filamentous fungus Trichoderma guizhouense can differentiate into typical terminal and intercalary chlamydospores characterized by double-layered spherical or ellipsoidal cell walls with accumulated lipid bodies

A hallmark of eukaryotic membranes is the pairing of lineage-specific sterols with characteristic sphingolipid species. Mammalian cell membranes are enriched in both cholesterol and long-chain sphingolipids such as sphingomyelin, whereas fungi synthesize ergosterol and very-long-chain sphingolipids with sugar-containing head groups. It has been proposed that these two lipid classes co-evolved to support membrane structure and organization. Here, we investigated how sterol structure and sphingolipid chain length together control membrane order and phase behavior. In the yeast Saccharomyces cere

It has been suggested that hypoxia-inducible factor 1α (HIF1α) serves as a key regulatory factor in insect metabolism remodelling under hypoxia, in which lipid metabolism appears to play an important role. However, the underlying mechanism remains incompletely understood. In this paper, the function of HIF1α in regulating lipid metabolism under hypoxia in Callosobruchus chinensis (Coleoptera: Bruchidae), a typical stored-product insect, was investigated. Based on the amplification and cloning, the sequence of the CcHIF1α gene was analysed and found to be highly conserved throughout evolution.

Psoriasis is a chronic inflammatory skin disease characterized by keratinocyte hyperproliferation and immune dysregulation. Recent studies highlight ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, as a key contributor to psoriatic pathogenesis. Lipocalin-2 (LCN2), an iron-binding protein elevated in psoriasis, may regulate this process. We investigated whether bezafibrate (BEZ), a pan-peroxisome proliferator-activated receptor (PPAR) agonist with broad metabolic and anti-inflammatory properties, could ameliorate psoriasis by targeting ferroptosis throu

Cancer stem cells (CSCs) contribute to therapeutic resistance, metastatic progression, and tumor recurrence, yet the metabolic pathways that sustain their survival remain incompletely understood. Here, we identify 3-mercaptopyruvate sulfurtransferase (3-MST), a hydrogen sulfide-producing enzyme encoded by MPST, as a metabolic dependency of colorectal CSCs. 3-MST expression was increased in human colorectal tumors and cancer cell lines and strongly correlated with proliferative capacity. HCT116-derived CSCs exhibited elevated 3-MST expression, increased hydrogen sulfide and reactive sulfur spec

CONTEXT: Mass motility assessment is widely used to evaluate semen quality in the sheep breeding industry, yet the biological factors influencing motility duration remain poorly characterised. At ejaculation, spermatozoa are combined with seminal plasma (SP), which initially supports motility and function but may compromise long-term metabolic endurance.
AIMS: This study aimed to investigate the effects of SP on mass motility duration and pH regulation of epididymal ram spermatozoa.
METHODS: Epididymal sperm were incubated with whole SP, saline, or left undiluted to assess motility duration.

OBJECTIVES: Immune-mediated mechanisms and altered lipid metabolism may contribute to increased cardiovascular risk in Sjögren's disease (SjD). Antibodies targeting high-density lipoprotein (HDL) have emerged as modulators of lipid dysfunction and inflammation in several rheumatic diseases, but their relevance in SjD remains unexplored. We aimed to characterize humoral responses against HDL in SjD and their associations with lipoprotein/lipid traits, inflammatory and proteomic features, and atherosclerosis.
METHODS: IgG serum levels of antibodies against HDL (anti-HDL) and apolipoprotein A1 (

Intracellular sphingolipids play a complex role in the survival and migration of oral squamous cell carcinoma (OSCC). For instance, dihydroceramide and ceramide sphingolipids suppress proliferation, whereas sphingosine-1-phosphate (S-1-P) promotes it. The balance between ceramide/S-1-P is critically regulated by the ceramide-hydrolyzing enzyme, acid ceramidase (aCDase), in OSCC. Indeed, aCDase mRNA expression is significantly upregulated compared with that in healthy epithelial cells. Our group identified that Porphyromonas gingivalis synthesizes a structurally unique phosphoglycerol-isoC17:0-

Despite major advances in lipid-lowering therapeutics, significant residual cardiovascular risk persists in dyslipidemic patients in primary and secondary prevention, even after optimization of LDL-cholesterol levels. All atherogenic apoB-containing lipoproteins, (VLDL and remnants, IDL, LDL and Lp(a)), exert deleterious effects on the arterial wall and atherosclerotic plaque, contributing to such risk. Mass spectrometric analyses have revealed that apoB-containing lipoproteins contain a diverse array of lipid classes (≥ 30); together with their constituent molecular species (> 1000), their li

Succinate dehydrogenase inhibitor (SDHI) fungicides are increasingly used and detected in surface waters, raising concerns for non-target aquatic organisms because they may disrupt mitochondrial metabolism. As mitochondrial structure and function are highly conserved in fungi and vertebrates, exposure to SDHIs could affect fish health; however, empirical data remain limited. Here, we assessed the short-term (4 days) and subchronic (21 days) effects of fluopyram, a commonly used SDHI, on adult goldfish (Carassius auratus) exposed to environmentally relevant concentrations (1 and 10 µg.L-1). Background/Objectives: Alterations in polyunsaturated fatty acid (PUFA) metabolites have been linked to the development of hepatocellular carcinoma (HCC). However, the association between PUFA metabolites and HCC development during nucleos(t)ide analogue (NUC) therapy in patients with chronic hepatitis B virus infection remains unclear. Methods: This study enrolled 195 NUC-naïve patients who received NUC therapy. Associations between metabolic factors-especially PUFA metabolites-and HCC development during NUC therapy were evaluated. Baseline serum concentrations of 158 PUFA metabolites were qu

Hepatitis E virus (HEV), a leading cause of acute and chronic viral hepatitis, poses a persistent global health challenge. A deeper mechanistic understanding of virus-host interactions is critical for identifying therapeutic targets to mitigate HEV-associated disease. In this study, we employ a systems biology framework to comprehensively map metabolic and bioenergetic alterations induced by HEV genotypes 1 and 3 in HepG2/C3a-MAVS-KD cells, a robust model of HEV infection, enabling reliable assessment of virus- and host-driven cellular changes. Our analyses reveal extensive remodelling of host

In cancer and chronic infection, CD8 T cell exhaustion is hallmarked by expression of inhibitory receptors such as PD1, TIM3, LAG3 and others1-3. Thus, inhibitory molecule focus has been limited to cell-surface proteins. Here we evaluate the surface lipid metabolite phosphatidylserine (PS) as a regulator of exhaustion. PS primarily localizes to the inner plasma membrane of live cells but is well known to be externalized to the outer membrane during cell death. The role of exposed PS on live immune cells is less clear. We show that viable, antigen-specific CD8 T cells externalize PS during lymp

Pathological aggregation of α-synuclein (αSyn) is a hallmark of Parkinson's disease (PD), but increasing evidence indicates that αSyn deposits extend beyond the central nervous system to peripheral tissues, including the skin. In PD patients, cutaneous αSyn accumulation has been associated with seborrheic dermatitis (SD), a chronic inflammatory condition linked to sebocyte dysfunction and altered sebum production. Marked differences in sebum composition between PD patients and healthy controls have been described. We aimed to investigate whether pathological αSyn could contribute to dysregulat

White and brown adipocytes show therapeutic potential, yet their exosomes mediated cell-specific effects remain poorly understood. This study aimed to assess the impact of conditioned media (CM) or exosomes derived from brown and white adipocyte-like cells (BALCs and WALCs, respectively) on four human cell types, including ovarian granulosa cells (OGCs), MCF-7 breast cancer cells, human adipose-derived stem cells (hADSCs), and human umbilical cord mesenchymal stem cells (hUCMSCs). Isolated hADSCs were differentiated into BALCs and WALCs using specific induction media. Corresponding exosomes we

Glucosylceramide synthase (GCS) catalyzes ceramide glycosylation in response to cell stress that produces glucosylceramide and other glycosphingolipids. GCS overexpression is a cause of drug resistance and enriches cancer stem cells (CSCs) during cancer chemotherapy. Previous studies showed that GCS modulates expression of p53 mutants and oncogenic gain-of-function (GOF) in heterozygous knock-in cell models ( TP53 R273H -/+ ). However, it is unclear whether GCS can modulate the effects of homozygous p53 mutations, which are common in many cancer cases. We report herewith that inhibition of GCS

BACKGROUND: Pancreatic cancer (PC) represents a highly lethal malignancy characterized by diagnostic challenges owing to nonspecific early symptoms and insufficiently sensitive biomarkers. This investigation sought to identify novel PC biomarkers through lipidomic profiling, an emerging metabolomics methodology examining lipid pathways in disease pathogenesis.
METHODS: We established a humanized murine PC model. Small-molecule oxidized lipid metabolites in primary pancreatic tumors and hepatic metastases were quantitatively analyzed using liquid chromatography-tandem mass spectrometry (LC-MS/

The significant roles of the farnesoid X receptor (FXR) and Takeda G protein-coupled receptor 5 (TGR5) in regulating metabolic pathways have recently been demonstrated. However, the precise effects of FXR inhibitors on glucose metabolism remain to be elucidated. In this study, we examined the impact of 7β-isopropylchenodeoxycholic acid (7β-ipCDCA), a dual FXR antagonist and TGR5 agonist, on impaired glucose metabolism in mice fed a Western diet. The dual FXR antagonistic/TGR5 agonistic activity of 7β-ipCDCA was confirmed through gene reporter assays. We evaluated its effects on glucose homeost

Acute Respiratory Distress Syndrome (ARDS), characterized by the rapid onset of respiratory failure and mortality rates of ∼40%, remains a significant challenge in critical care medicine. Despite advances in supportive care, accurate prediction of ARDS mortality remains challenging, resulting in delayed delivery of targeted interventions and effective disease management. Traditional critical illness severity scores lack specificity for ARDS, underscoring the need for more precise prognostic tools for ARDS mortality. To address this crucial gap, we employed a multimodal approach to predict ARDS

HFPO-DA ammonium salt (GenX) is used to replace the environmentally persistent perfluorooctanoic acid (PFOA), but its effects on fish lipid metabolism remain poorly understood. This study investigated the impact of dietary GenX exposure (0.73 and 3.10 mg/Kg of feed, measured concentrations) on hepatic lipid metabolism and oxidative stress in juvenile male Nile tilapia (5.32 ± 1.60 g) over 28 days. While fish increased body weight over time, there were no differences in weight gain among treatments. There were no changes in hepatic glycerol levels with either dose of feed, while triglycerides d
