Verbal learning and memory in children with fetal alcohol syndrome.

Children with fetal alcohol syndrome (FAS) were administered the California Verbal Learning Test-Children's Version, a word list learning task that assesses immediate and delayed recall and recognition memory. When compared with matched control children, the children with FAS had difficulty learning and recalling the words after a delay period and tended to make an increased number of intrusion and perseverative errors. In addition, they had difficulty discriminating target words from distracter words and made more false-positive errors on recognition testing. Some of these deficits persisted

The fetal alcohol syndrome in adolescence.

At present, alcohol is recognized as the leading teratogenic agent in long-lasting CNS dysfunction. Little is known about the long-term development and outcome of children with fetal alcohol syndrome (FAS). Forty-four FAS patients who were diagnosed in early childhood were followed up for 10-14 years. This study documents the developmental changes of the manifestations of FAS from childhood to adolescence and describes a characteristic "juvenile" pattern of FAS, which may help to identify this syndrome even in adolescence. This is especially relevant for patients who were not diagnosed earlier

Awareness and knowledge of fragile X syndrome among special educators.

A questionnaire to assess awareness and knowledge of various syndromes was mailed to a random sample of elementary special education teachers in Colorado. Approximately half of the questionnaires were completed and returned. Results indicate that significantly more respondents had knowledge of Down syndrome than of fetal alcohol or fragile X syndromes. Respondents' level of education, date of most recent degree, and years of experience were not associated with greater awareness nor knowledge of fragile X syndrome. Respondents expressed interest in a broad array of fragile X in-service training

A longitudinal analysis of the effect of prenatal alcohol exposure on growth.

Cross-sectional analyses have demonstrated that prenatal exposure to alcohol can result in growth deficits at birth and in early childhood. However, there have been few longitudinal analyses of this relationship. This study presents an analysis of the longitudinal effect of prenatal alcohol exposure on growth. The model used for analysis was a general unbalanced repeated-measures model with a fully parameterized covariance matrix. Application of the model demonstrated that for length and head circumference, the effect of prenatal alcohol exposure was constant over time. The relationship betwee

The effects of prenatal alcohol use on the growth of children at three years of age.

In this prospective study of substance use during pregnancy, women were interviewed in their 4th and 7th prenatal months, and women and children were assessed at 24 hr, 8, 18, and 36 months postpartum. Data are presented on the outcome of 519 children at age 3. At 3 years, children who were exposed prenatally to alcohol were smaller in weight, length, and head circumference. They also had an increased number of minor physical anomalies. These effects were found even after controlling for nutritional and environmental factors. The persistence of growth effects at age 3 suggests that children ex

Prenatal ethanol exposure and opiatergic influence on puberty in the female rat.

Fetal alcohol exposure (FAE) is associated with a variety of physiological and behavioral dysfunctions, including deficits to reproductive function. FAE has also been shown to increase brain beta-endorphin levels. This study sought to determine whether the common delay of the onset of puberty in fetal alcohol-exposed animals could be due to increased opiate inhibition of LH release. Prepubertal female rats were injected with an opiate antagonist, naltrexone, over days 26-29. This naltrexone treatment led to an acceleration of vaginal opening and first estrus in FAE animals; had no effect on ch

Ethanol consumption inhibits fetal DNA methylation in mice: implications for the fetal alcohol syndrome.

Acute ethanol administration (3 g/kg twice a day) to pregnant mice, from the 9th thru the 11th day of gestation, resulted in hypomethylation of fetal deoxyribonucleic acid (DNA). Nuclei isolated from the fetuses of the ethanol-treated mice had lower levels of methylase activity relative to controls even in the presence of excess S-adenosylmethionine, which serves as the methyl donor for the enzyme DNA methyltransferase. Acetaldehyde, at concentrations as low as 3 to 10 microM, inhibited DNA methyltransferase activity in vitro. Since DNA methylation is thought to play an important role in the r

Rats exposed prenatally to alcohol exhibit impairment in spatial navigation test.

Prenatal exposure to ethanol causes learning disabilities and low I.Q. scores. The objective of the present studies was to investigate whether exposure of rats to ethanol in utero, would induce a deficit in spatial memory in adult life. Pregnant rats were fed with an ethanol diet from day 1 of pregnancy till parturition. Control rats were either pair-fed with an isocaloric sucrose diet or were fed with lab-chow ad libitum. On the first day of birth, offspring exposed to ethanol in utero were placed with a control mother fed with lab-chow, while offspring of the lab-chow fed dams were placed wi

Prenatal alcohol exposure: comparability of effects in humans and animal models.

Much is known about the potential consequences of prenatal alcohol exposure through data developed in animals and man. However, little attempt has been made to compare the human and animal literature with respect to qualitative and quantitative similarities and differences. To this end, a comparison was made between the effects reported in humans following moderate levels of alcohol exposure and the neurobehavioral effects detected using animal models. A good deal of congruence was found with respect to qualitative endpoints. General functional categories, such as deficits in learning, inhibit

Ethanol teratogenesis in selectivity bred long-sleep and short-sleep mice: a comparison to inbred C57BL/6J mice.

Sensitivity to alcohol may influence the severity of ethanol teratogenesis. To examine this hypothesis, the teratogenic effects of ethanol were compared in Long-Sleep (LS) and Short-Sleep (SS) mice, selectively bred for differences in ethanol-induced narcosis. Inbred C57BL/6J (B6) mice were included to confirm previously reported teratogenic effects using our own treatment regimen and standard assessment technique. Intragastric administration of ethanol (5.8 g/kg) on Days 9 and 10 of pregnancy resulted in growth retardation and an increase in prenatal mortality in LS litters but not in SS litt

Development of the EEG between 30 and 40 weeks gestation in normal and alcohol-exposed infants.

A total of 441 newborn infants with gestational ages between 30 and 40 weeks had EEG studies between 36 and 48 hours after birth. Their mothers had either abstained from alcohol during pregnancy or had ingested alcohol in one of four categories: occasional, moderate, binge or frankly alcoholic. The power of the EEG, using linear regression analysis, was significantly higher among infants of mothers in the occasional, binge and alcoholic categories than among infants of abstainers. Developmental changes in the EEGs of binge-drinking mothers were even more striking than in those of the offspring

Prenatal alcohol exposure: recent work on behavioral dysfunctions in pre-weanling rats.

This article reviews recent data collected in our laboratory examining the behavioral effects of prenatal alcohol exposure in pre-weanling rats. The behavioral alterations which are discussed include suckling dysfunctions, overactivity, and early learning deficits. Similar behavioral dysfunctions have also been reported in children exposed to alcohol during prenatal development. These similarities are discussed as well as possible insights that might be gained from animal models of fetal alcohol effects.

Influence of prenatal ethanol exposure on hormonal responses to clonidine and naloxone in prepubescent male and female rats.

The expression of sexually dimorphic behavior has been found to be altered in adult animals following prenatal alcohol exposure. The present study examined whether such exposure would alter the sexually dimorphic response of luteinizing hormone (LH) to clonidine and naloxone observed in normal prepubescent animals. Both LH and corticosterone (CS) were measured in 16 day old male and female rats 30 min after injection of naloxone (2 mg/kg) or clonidine (0.1 mg/kg). Prenatal alcohol exposure did not influence the LH response to either drug in females. An LH response to clonidine in normal males

Alcohol-induced microencephaly during the third trimester equivalent: relationship to dose and blood alcohol concentration.

Alcohol was administered in different doses to groups of neonatal rat pups from postnatal days 4-10 using an artificial rearing technique. Blood alcohol concentrations (BACs) were monitored in pups on postnatal day 6. Brain weights were measured on postnatal day 10 and the extent of microencephaly was correlated with dose and BAC. Doses of 7.4 g/kg/day and above resulted in microencephaly. Although BACs varied considerably among individual animals at each dose tested, the amount of microencephaly increased with the BAC, and the amount of brain growth reduction was more dependent on BAC than do

Nutritional teratogens: a survey of epidemiological literature.

Alcohol seems to be, by far, the most important nutritional teratogen. Assuming an incidence rate of 1.3%o, 240 newborns with fetal alcohol syndrome per year would be expected in the Netherlands alone. Only a minor fraction of this number is diagnosed as such. Even moderate drinking during pregnancy results in developmental disturbances. There is no safe consumption level. Therefore the intake of alcohol during pregnancy should be abandoned completely. There is conflicting evidence on the effects of caffeine with regard to pregnancy outcome. A slight decrease in birthweight among the offspring

Introduction: what do we know of the mechanisms of alcohol damage in utero?

The newly established role of alcohol as a human teratogen important in the aetiology of mental deficiency poses the problem of how maternal ingestion of alcohol damages the developing central nervous system. Two main approaches to the identification of possible mechanisms are suggested: clinical field studies, especially of relatives of affected children, and the use of experimental animal models. The evidence so far provided by each approach, and the limitations of each, are considered. Four possible mechanisms of alcohol-induced damage, originating at different stages of antenatal developme

Ethanol suppression of locus coeruleus neurons: relevancy to the fetal alcohol syndrome.

The effect of intraperitoneal injection and microiontophoretic application of ethanol on spontaneous single neuron activity of adult rat locus coeruleus was studied. Systemic administration of ethanol (1-2 g/kg) markedly slowed a majority of locus coeruleus neurons. Direct application of ethanol also substantially suppressed locus coeruleus cell firing in the absence of any detectable membrane effects. Percent inhibition of firing rate ranged from 6% to 100% with ejection currents up to 40NA for 40 sec. These data demonstrate ethanol can directly suppress locus coeruleus unit activity. Since l

Fetal alcohol syndrome and mental retardation: spine distribution of pyramidal cells in prenatal alcohol-exposed rat cerebral cortex; a Golgi study.

Among the various possible effects of the fetal alcohol-syndrome (FAS), mental retardation can be considered the most deleterious. In animal studies, prenatal alcohol exposure has been shown to result in increased neonatal mortality, retarded cerebellar development and a significant decrease in neonatal brain weight. In a Golgi study on Wistar rats that were prenatally exposed to alcohol the spine distribution in proximal apical dendrites of layer V pyramidal cells of the parietal cortex was examined. As compared with controls, a distinct spine abnormality could be demonstrated at 12 days and
