Suppression of genioglossus muscle tone and activity during reflex hypercapnic stimulation by GABA(A) mechanisms at the hypoglossal motor nucleus in vivo.

The genioglossus muscle is involved in the maintenance of an open airway for effective breathing. Inhibitory neurotransmitters may be responsible for the major suppression of hypoglossal motor output to genioglossus muscle that occurs in certain behaviours such as rapid-eye-movement sleep. There is evidence for GABA(A) receptor-mediated inhibition of hypoglossal motoneurons in vitro. However, comparable studies have not been performed in vivo and the interactions of such mechanisms with integrative reflex respiratory control have also not been determined. Urethane-anaesthetised, tracheotomized

Area postrema mediates gastric motor response induced by apomorphine in rats.

The effects of apomorphine administration on the autonomic responses were investigated in rats. Distinctive gastric motor responses were observed after the intravenous administration of apomorphine (0.1 mg/kg body weight). Gastric motor responses in the distal stomach induced by apomorphine administration were classified into two types. One type involved inhibition of phasic contractions which appeared just after the administration of apomorphine. The other involved an increase in the frequency of small phasic contractions accompanied by increased gastric tone appearing with a relatively longe

Selective vagal afferent dysfunction in dogs with congenital idiopathic megaoesophagus.

Congenital idiopathic megaoesophagus (CIM) is a rare, naturally occurring disorder of the dog that is characterised by deficient motility and dilatation of the oesophagus. Recent studies indicate that the vagal sensory system mediating reflexes induced by oesophageal distension is defective in, and may underlie the pathomechanism of this disorder. We sought to establish whether other distension sensitive vagal afferent systems were impaired in CIM, or whether the vagal afferent dysfunction was selective. Thus, we examined the Hering-Breuer lung inflation reflex (HBR), which is subserved by a c

Effects of atropine on the responses of rapidly adapting pulmonary stretch receptors and dynamic lung compliance to sodium cyanide-induced hyperpnea.

The responses of tracheal pressure (PT) and heart rate (HR) to electrical stimulation of the peripheral cut-ends of the vagus nerves (10-15 V, 10 Hz, 1 ms) to activate both myelinated and non-myelinated fibers were examined before and after administration of atropine (1 mg/kg) in artificially ventilated, bilaterally vagotomized rabbits. Vagal stimulation caused an increase in PT and a decrease in HR. The PT response to vagal stimulation was completely blocked by atropine which significantly reduced the bradycardia evoked by the stimulation. In other series of experiments, we also examined the

Parietal cell structure during inhibition of acid secretion.

Acute, pharmacological inhibition of gastric acid secretion is paralleled by a return of the parietal cells to a resting morphology. However, some inhibitors acting on targets in the parietal cells distal to the receptors, may inhibit acid secretion without the return to a resting morphology. This is the case with e.g. picoprazole and thiocyanate. Long-term pharmacological or surgical inhibition of acid secretion may be followed by changes in the number and/or size of the parietal cells.

A study of intragastric pH in patients with peptic ulcer--with special reference to the clinical significance of basal pH value.

Determination was made of in situ gastric pH during early morning in fasting state (basal pH) by using wired glass electrodes, and results obtained were analyzed. Analysis of the pattern of variation in intragastric pH at night revealed no substantial difference between normal subjects and patients with peptic ulcer but a tendency to be lower in the latter group. It was also shown that sleeping waves appeared in the electroencephalogram in association with the increase in intragastric pH during sleep at night. The basal pH value was 5.4 +/- 2.1 in patients with gastric cancer, 3.0 +/- 2.2 in t

Modulation of innate immune response by the vagus nerve in experimental hepatic amebiasis in rats.

The parasympathetic nervous system has a crucial role in immunomodulation of the vagus nerve, its structure provides a pathogen detection system, and a negative feedback to the immune system after the pathogenic agent has been eliminated. Amebiasis is a disease caused by the protozoan parasite Entamoeba histolytica, considered the third leading cause of death in the world. The rats are used as a natural resistance model to amoebic liver infection. The aim of this study is to analyze the interaction of Entamoeba histolytica with neutrophils, macrophages, and NK cells in livers of intact and vag

Phrenic motor neuron TrkB expression is necessary for acute intermittent hypoxia-induced phrenic long-term facilitation.

Phrenic long-term facilitation (pLTF) is a form of hypoxia-induced spinal respiratory motor plasticity that requires new synthesis of brain derived neurotrophic factor (BDNF) and activation of its high-affinity receptor, tropomyosin receptor kinase B (TrkB). Since the cellular location of relevant TrkB receptors is not known, we utilized intrapleural siRNA injections to selectively knock down TrkB receptor protein within phrenic motor neurons. TrkB receptors within phrenic motor neurons are necessary for BDNF-dependent acute intermittent hypoxia-induced pLTF, demonstrating that phrenic motor n

Novel gastroinsular axis involving a gastric transmural glucose flux and vagal mediation.

To determine whether the appearance of nutrients into the gastric lumen per se provokes insulin secretion, glucose solution was instilled into the pylorus-cannulated stomach via an orogastric tube in anesthetized dogs. When 200 ml of 0, 5, 10, and 20% glucose solution were sequentially instilled, transgastric gradients (TGG) of plasma glucose concentration across the fundus [short gastric vein (SGV) - femoral artery, TGG(SGV)] and insulin levels in the superior pancreaticoduodenal vein (SPDV) increased stepwise. Upon instillation of 300 ml of 10% glucose, but not 1.8% saline, for 12 min follow

Evidence for central chemoreception in the midline raphé.

We injected acetazolamide (AZ; 5 x 10(-6) M; 1 nl; n = 14), its inactive analogue 2-acetylamino-1,3,4-thiadiazole-5-sulfon-t-butylamide (5 x 10(-5) M; n = 6), or mock cerebrospinal fluid (n = 5) into the caudal raphé in the midline brain stem of anesthetized paralyzed ventilated rats. These AZ injections have been shown to produce a focal region of tissue acidosis with a radius < 350 microns and are used as a probe for sites of central chemosensitivity. Compared with control injections, AZ injection into the raphé, as demonstrated by anatomic analysis of injection location, significantly incre

Modulation of respiratory rhythm by alpha 2-adrenoceptors in awake and anesthetized goats.

The purpose of this study was to examine the effects of alpha 2-adrenoceptor agonists in the control of breathing with goats that were either awake (n = 7) or anesthetized and artificially ventilated (n = 11). Awake goats infused intravenously with either of the alpha 2-agonists clonidine (1.0-6.0 micrograms/kg) or guanabenz (15.0-63.0 micrograms/kg) exhibited two distinct ventilatory patterns. One pattern was characterized by tachypnea in which respiratory frequency and minute ventilation increased to approximately 50% above control values. A second ventilatory pattern consisted of slow breat

[Treatment of alkaline gastritis and recurrent ulcer following Roux-en-Y gastrectomy and construction of a Rumpf antireflux valve].

Two-third gastrectomy or supplementary gastrectomy was performed on 13 patients, between July 2, 1986 and September 30, 1988. Continuity of the gastro-intestinal tract was restored by end-to-side gastrojejunostomy according to Roux-en-Y. An antireflux valve according to Rumpf was inserted 15 cm below the gastroenteroanastomosis, and end-to-side jejunostomy was performed about six centimetres below the valve. This surgical method was applied to six patients for treatment for alkaline gastritis. The same method was used on another seven patients to prevent development of alkaline gastritis. All

A predictor analysis of patients' assessment of outcome after operation for duodenal ulcer. A one-year prospective study.

During a period of one year data were obtained concerning life events, non-specific psychological symptoms, individual social history and ulcer history in consecutive cases of patients about to undergo elective surgical treatment for duodenal ulcer. At a one-year follow-up, a blind clinical evaluation was performed, and information concerning the patients' assessment of outcome was obtained. Those patients who at the one-year follow-up stated no improvement due to the operation could be predicted to some extent from postoperative complications, partly from a long ulcer history. The patients wh

Minireview. The hypothalamus and blood glucose regulation.

Complex neural circuits exist in the hypothalamus for the control of metabolic hormones, enzyme activities, and substrate flux that appear to be involved with the normal adaptation to feeding and subsequent blood sugar regulation. Abnormalities within this system have been found in experimental obesity and in human diabetes, and such maladaptive changes are thought to contribute to the pathophysiology of both disorders. The present report is an attempt to present a coherent picture of the manifold factors which may be involved in the homeostatic regulation of blood glucose levels by the hypoth
