Approach bias retraining to augment smoking cessation: A pilot randomized controlled trial.

BACKGROUND: Approach tendency to smoking-related cues has been associated with greater cravings, nicotine dependence, and the likelihood of relapse. In this pilot randomized clinical trial, we examined the efficacy of approach bias retraining (ABR; i.e., increasing avoidance tendency) for enhancing standard smoking cessation treatment (ST). METHODS: Adult smokers (N = 96) motivated to quit were randomly assigned to 7 weekly in-person treatment sessions consisting of either (1) cognitive-behavioral therapy for smoking cessation (ST) and ABR (ST+ABR) or ST and sham retraining (ST+Sham). All par…

Dopamine Supersensitivity: A Novel Hypothesis of Opioid-Induced Neurobiological Mechanisms Underlying Opioid-Stimulant Co-use and Opioid Relapse.

Emergent harms presented by the co-use of opioids and methamphetamine highlight the broader public health challenge of preventing and treati … dence, would contribute to the acute opioid withdrawal syndrome, and could manifest subjectively as craving and/or motivation to use that could prompt opioid relapse during acute and protracted withdrawal. Preclinical research is needed to directly test these hypothesized mechanisms. Human laboratory and clinical trial research is needed to explore these …

Exploring the Therapeutic Potential of Targeting Purinergic and Orexinergic Receptors in Alcoholic Neuropathy.

Alcoholic neuropathy emerges following the persistent alcohol imbibing, and triggers nerve damage through de-escalating the receptors situat … n of inflammatory mediators, NF-κB signalling pathway, ROS formation, nerve cell deterioration, and craving for alcohol consumption, thereby ceasing the evolution of alcoholic neuropathy. The authors focus to highlight the importance of this alternative strategy as a novel target in alcoholic neuropathy, and to incentivize researchers to scrutinize the po …

The influence of sex and reproductive cycle on cocaine-induced behavioral and neurobiological alterations: a review.

This systematic review (SR) was aimed at answering two questions: (1) how sex and ovarian hormones alter behavior associated with cocaine us … sment. Results gathered showed that rodent females develop increased consumption, seeking behavior, craving, relapse, locomotion, increases in stress and anxiety, among other behavioral alterations during peaks of estrogen. These observations are related to the direct effects played by ovarian hormones (in particularly estradiol), in dopamine, but also in …

Multi-level predictors of psychological problems among methadone maintenance treatment patients in difference types of settings in Vietnam.

BACKGROUND: Methadone, a long-acting opioid agonist maintenance treatment (MMT) is used to treat opioid addiction by preventing opioid withdrawal and reducing cravings. However, it is important to note that mental conditions may persist, or even remain undetected while methadone maintenance treatment is ongoing. This study aimed to examine the level of psychological problems among MMT patients at public and private health facilities and identify associated factors. METHOD: From January to September 2018, a cross-sectional study was performed in Nam Dinh province, one of the largest epicenters…

Analysis of buprenorphine/naloxone dosing impact on treatment duration, resource use and costs in the treatment of opioid-dependent adults: a retrospective study of US public and private health care claims.

OBJECTIVES: The buprenorphine/naloxone combination is used to treat the chronic relapsing disorder of opioid dependence. Adequate dosing levels are important to control cravings, prevent withdrawal syndrome, and maintain patients in treatment. The objective of this study was to estimate the impact of dosing on treatment persistence, resource utilization, and total direct health care costs. METHODS: A retrospective cohort analysis was performed using administrative claims extracted from the MarketScan and Clinformatics databases from January 2007 to June and November 2012. Patients initiating …

Conditioned place preferences (CPPs) to high-caloric "snack foods" in rat strains genetically prone vs. resistant to diet-induced obesity: resistance to naltrexone blockade.

A previous study in our laboratory using Sprague-Dawley (SD) male rats showed that conditioned place preferences (CPPs) can be learned to tw … locking these CPPs. These results show that (a) DIO and DR rats can learn CPPs (i.e., "exhibit food cravings") as well as their parent strain after periodic access to high-caloric palatable foods, but imply that (b) some physiological system other than the endogenous opioid system mediates such learning.

Cocaine-but not methamphetamine-associated memory requires de novo protein synthesis.

Context-induced drug craving and continuous drug use manifest the critical roles of specific memory episodes associated with the drug use experiences. Drug-induced conditioned place preference (CPP) in C57BL/6J mouse model, in this regard, is an appropriate behavioral paradigm to study such drug use-associated memories. Requirement of protein synthesis in various forms of long-term memory formation and storage has been phylogenetically demonstrated. This study was undertaken to study the requirement of protein synthesis in the learning and memory aspect of the conditioned place preference indu…

Caffeine dependence in combination with a family history of alcoholism as a predictor of continued use of caffeine during pregnancy.

OBJECTIVE: The purpose of the study was to examine whether caffeine dependence and a family history of alcoholism are associated with contin … cy. Saliva caffeine levels confirmed these effects. Drug addiction. Part III. Pharmacotherapy of addiction.

The last decade brought a considerable progress in pharmacotherapy of addiction. Basing on recently gained knowledge of mechanisms of development of addiction and the physiology of the brain reward system, several therapeutic strategies have evolved. The strategies aimed at targeting the basic mechanisms of addiction rely on the premises that addiction is caused by adaptive changes in the central nervous system and that craving, which is the main cause of relapse, depends on dopaminergic mechanisms and requires high general excitability. The pharmacological approach involves drugs that reduce …

Fiber-Less, Large-Scale Opto-Electrophysiology Interface for Micro-Scale Interaction of Multiple Brain Regions.

OBJECTIVE: Recent neuroscientific research craves for understanding sophisticated brain networks formed by neuron ensembles across multiple regions. An ideal way to unveil the complex connectome is bidirectionally interacting (simultaneous recording and stimulation) with neurons at high spatiotemporal resolutions. Existing CMOS recording probes cannot provide micro-scale interactions with limited stimulation capability. Although optogenetics can achieve neuron-specific stimulation, conventional methods using optic fibers illuminate a large volume of tissue, resulting in unspecific perturbation
