Two tandem verprolin homology domains are necessary for a strong activation of Arp2/3 complex-induced actin polymerization and induction of microspike formation by N-WASP.

All WASP family proteins share a common C terminus that consists of the verprolin homology domain (V), cofilin homology domain (C), and acidic region (A), through which they activate Arp2/3 complex-induced actin polymerization. In this study, we characterized the Arp2/3 complex-mediated actin polymerization activity of VCA fragments of all of the WASP family proteins: WASP, N-WASP, WAVE1, WAVE2, and WAVE3. All of the VCA fragments stimulated the nucleating activity of Arp2/3 complex. Among them, N-WASP VCA, which possesses two tandem V motifs, had a more potent activity than other VCA proteins

An interaction between Sla1p and Sla2p plays a role in regulating actin dynamics and endocytosis in budding yeast.

The importance of a dynamic actin cytoskeleton for facilitating endocytosis has been recognised for many years in budding yeast and is increasingly recognised in mammalian cells. However, the mechanism for actin recruitment and the role it plays in endocytosis is unclear. Here we show the importance of two yeast proteins in this process. We demonstrate that Sla1p and Sla2p interact in vitro and in vivo and that this interaction is mediated by the central domain of Sla2p, which includes its coiled-coil region, and by a domain of Sla1p between residues 118 and 361. Overexpression of the interact

Phenotypic screening of mutations in Pmr1, the yeast secretory pathway Ca2+/Mn2+-ATPase, reveals residues critical for ion selectivity and transport.

Thirty-five mutations were generated in the yeast secretory pathway/Golgi ion pump, Pmr1, targeting oxygen-containing side chains within the predicted transmembrane segments M4, M5, M6, M7, and M8, likely to be involved in coordination of Ca(2+) and Mn(2+) ions. Mutants were expressed in low copy number in a yeast strain devoid of endogenous Ca(2+) pumps and screened for loss of Ca(2+) and Mn(2+) transport on the basis of hypersensitivity to 1, 2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid (BAPTA) and Mn(2+) toxicity, respectively. Three classes of mutants were found: mutants indistin

Assembly of a rod-shaped chimera of a trimeric GCN4 zipper and the HIV-1 gp41 ectodomain expressed in Escherichia coli.

The HIV-1 envelope subunit gp41 plays a role in viral entry by initiating fusion of the viral and cellular membranes. A chimeric molecule was constructed centered on the ectodomain of gp41 without the fusion peptide, with a trimeric isoleucine zipper derived from GCN4 (pIIGCN4) on the N terminus and part of the trimeric coiled coil of the influenza virus hemagglutinin (HA) HA2 on the C terminus. The chimera pII-41-HA was overexpressed as inclusion bodies in bacteria and refolded to soluble aggregates that became monodisperse after treatment with protease. Either trypsin or proteinase K, used p

Two new sulfur-containing pyridine alkaloids from the co-culture of Diaporthe fungi.

The untapped metabolome of filamentous fungi represents a profound reservoir for therapeutic lead discovery. Although genomic mining confirms an abundance of cryptic biosynthetic gene clusters (BGCs), axenic laboratory culture frequently lacks the ecological stimuli required to trigger their expression, inevitably leading to the redundant isolation of known metabolites. To circumvent this biosynthetic silencing and unlock hidden chemical diversity, strategies such as heterologous expression, the one-strain-many-compounds (OSMAC), epigenetic modulation, and microbial co-cultivation have been in

Gut mycobiome in the spectrum of Metabolic-dysfunction Associated Steatotic Liver Disease (MASLD): from MASH to hepatocellular carcinoma.

UNLABELLED: Gut Microbiome (GM) dysbiosis is associated with the development and progression of Metabolic-dysfunction Associated Steatotic Liver Disease (MASLD) and steatohepatitis (MASH). Human GM contains bacteria, fungi, archaea, and viruses. Gut mycobiome (GMyco) is still poorly studied in patients with MASLD. Herein we investigated GMyco composition at different MASLD stages. METHODS: 23 patients were recruited: controls (n = 5); MASH (n = 6); cirrhosis (n = 7); and Hepatocellular Carcinoma (HCC) (n = 5). Stool samples were collected and stored in -80 °C for subsequent DNA extraction and

Optimization of functional polysaccharide production in Naematelia aurantialba: Surfactant-driven yield enhancement and multifunctional property optimization.

BACKGROUND: Naematelia aurantialba polysaccharide (NAPS) is widely exploited in the food and pharmaceutical industries because of its diverse biological activities. Although chemical elicitors have been reported to enhance polysaccharide production by edible mushrooms under submerged fermentation, their influence on NAPS biosynthesis remains unexplored. RESULTS: We evaluated the effect of chemical agents on NAPS synthesis in 7.5 L fermenters. Supplementation with 0.50% (v/v) polyoxyethylene sorbitan monooleate (Tween 80) raised the NAPS yield to 17.32 ± 0.25 g L-1, a 15.31% increase over the

Belowground carbon use in woody plants under experimental warming: uneven evidence for fine-root, root-exudation, and mycorrhizal responses.

How warming changes belowground carbon use in woody plants remains poorly understood. Most experiments measure fine-root, root-exudation, and mycorrhizal responses separately, providing limited evidence on how these responses vary together within the same experiment. We synthesized 13 primary publications from 11 independent experiments: eight field, one outdoor pot, and two controlled experiments. Our synthesis showed no significant increase in fine-root biomass or production. Root respiration, mortality, and N concentration increased, whereas root P and non-structural carbohydrate concentrat

Whole-Cell Transformation of Cannabidiol by Selected Filamentous Fungi into Novel Polar Derivatives.

Cannabidiol (CBD) is a bioactive phytocannabinoid with considerable pharmacological potential. However, its limited aqueous solubility and high lipophilicity remain significant barriers to its broader pharmaceutical application. In this study, the enzymatic potential of selected filamentous fungi was investigated as a whole-cell biocatalytic platform for the regioselective functionalization of CBD. Sixteen fungal strains were screened, and thirteen microorganisms successfully transformed CBD into more polar derivatives. Four strains showing distinct and promising chromatographic profiles were

Phaeosphaeride A Isolated from an Endophytic Paraphoma sp. Alleviates ABCG2-mediated Resistance to Mitoxantrone in Breast Cancer Cells.

ABSTRACT: The efficacy of breast cancer chemotherapies is frequently limited by multidrug resistance (MDR), partly through efflux by ABC transporters, including ABCG2. This study evaluated whether phaeosphaeride A (PPA), a fungal metabolite isolated from a Paraphoma sp. endophyte of Ferula xylorhachis, can modulate ABCG2-mediated resistance to mitoxantrone (MTX). The endophyte was cultured and extracted with ethyl acetate, and the extract was purified by column chromatography and HPLC to yield PPA, whose structure was confirmed by NMR and MS analyses. Cytotoxicity of MTX, PPA, and their combin

Shifts in soil microbial diversity and greenhouse gas emission dynamics.

Anthropogenic land formation has been shown to substantially alter terrestrial landscapes, impacting both soil properties and microbial communities. Nevertheless, the impact of anthropogenic land formation on greenhouse gas emissions and the underlying mechanisms were not fully elucidated. In this study, chronological sequences of soil samples were collected from a recently reclaimed land area at multiple intervals over a fifteen-year period. High-throughput sequencing was employed to analyze the composition of microbial communities, including bacteria, fungi, and protists. It was observed tha

Packing Density Governs Tobacco Quality Through Microbial Community Assembly and Metabolic Reprogramming.

Packing density regulates the microenvironment of tobacco (Nicotiana tabacum L.) fermentation and may thereby influence microbial activity and product quality. However, its effects on microbial community assembly and quality formation remain poorly understood. This study aimed to clarify how packing density affects flue-cured tobacco quality by shaping microbial communities, functional potential, and ecological interactions. Here, we investigated the effects of three packing densities (60%, 70%, and 80%) on chemical components, aroma compounds, microbial community structure, functional potenti

Design, synthesis, antifungal activity and preliminary mechanistic study of benzimidazole-containing cinnamamide derivatives against phytopathogenic fungi.

Phytopathogenic fungi continue to pose a serious threat to crop production, while the long-term use of conventional fungicides has raised concerns regarding resistance development and environmental impact. To identify new bioinspired antifungal leads, a series of benzimidazole-containing cinnamamide derivatives were designed and synthesized based on the cinnamic acid scaffold and evaluated against representative phytopathogenic fungi. The bioassay results showed that compound V35 exhibited the most potent and broad-spectrum antifungal activity, with EC50 values of 0.28, 0.69, 1.52, and 1.81 μg

Community-Acquired Kodamaea ohmeri Fungemia in an Immunocompetent Host Coexisting With Severe Leptospirosis: A Case Report and Review of Community-Acquired Fungemia Cases.

Community-acquired fungemia in immunocompetent hosts remains exceptionally rare. Kodamaea ohmeri is an emerging opportunistic yeast pathogen increasingly recognised as a cause of invasive fungal infections, particularly among immunocompromised individuals and in healthcare settings. A 33-year-old previously healthy female farmer presented with severe leptospirosis with multiorgan dysfunction. Blood cultures obtained before central venous catheter insertion grew K. ohmeri, identified by the VITEK 2 YST card and matrix-assisted laser desorption ionisation-time of flight (MALDI-TOF) mass spectrom

Duration of Spent Mushroom Substrate Return Affects Microbial Assembly and Nitrogen Metabolism to Promote Functional Stabilization in Rice-Mushroom Crop Rotation Systems.

Spent mushroom substrate (SMS) return is a vital strategy for agricultural waste recycling and soil fertility improvement, yet its ecological impacts of duration remain poorly understood. This study employed metagenomic sequencing to explore soil fertility, microbial dynamics, and nitrogen cycling across different SMS return durations (0, 1, and 3 years) within rice-mushroom crop rotation systems. Soil nutrients (organic matter, total nitrogen, total phosphorus) initially decreased and then increased throughout the rice growth cycle. The one-year return (y1) induced early nutrient depletion, w

Microbial Interactions and Flavor Modulation in Cabbage Fermentation: Roles of Lactiplantibacillus plantarum and Rhodotorula mucilaginosa.

Fermented cabbage is a globally popular food, whose distinctive flavor is largely determined by microbial metabolism and interspecies interactions. To understand the roles of Lactiplantibacillus plantarum (formerly Lactobacillus plantarum) and Rhodotorula mucilaginosa in regulating microbial community structure and flavor formation during cabbage fermentation, natural fermented cabbage was compared with that inoculated with L. plantarum, R. mucilaginosa and both strains. The results showed that cabbage inoculated with either strain accelerated fermentation, with the fastest acidification obser

A Retrospective Molecular Epidemiology Study: Emergence and dissemination of Azole-Resistant Candida parapsilosis Sensu Stricto in Southern Brazil.

Candida parapsilosis sensu stricto has emerged as a major cause of healthcare-associated candidemia, with increasing reports of fluconazole resistance worldwide. This resistance is most commonly associated with amino acid substitutions in Erg11p, particularly Y132F; however, regional variation in resistance mechanisms and clonal dissemination remains not fully understood. This study investigated the molecular epidemiology and genomic landscape of fluconazole-resistant C. parapsilosis in Paraná, Southern Brazil, over a 10-year period. A total of 54 resistant isolates identified between 2014 and

Efflux pump gene ABCA as targets for deep learning-based de novo inhibitors in invasive aspergillosis caused by Aspergillus fumigatus.

This study aims to design and evaluate novel therapeutic binders targeting the ATP-binding region of the AbcA efflux transporter protein in Aspergillus fumigatus. By inhibiting ATP binding, these binders seek to prevent AbcA-mediated azole resistance and restore the effectiveness of antifungal treatments. The research employed a variety of computational modeling and deep learning techniques, such as multiple sequence alignment (MSA) to identify conserved regions, particularly the ATP-binding site. A de novo sequence and structure of the protein was predicted using Protein Generator based on th

Impact of allergic bronchopulmonary aspergillosis overlap in chronic pulmonary aspergillosis.

BACKGROUND: Chronic pulmonary aspergillosis (CPA) is a destructive fungal infection caused by Aspergillus fumigatus, leading to significant morbidity in individuals with structural lung disease. Clinical and immunological overlap with allergic bronchopulmonary aspergillosis (ABPA) has been recognised, but its extent and prognostic relevance remain uncertain. This study assessed ABPA features in CPA and their relationship with immunological markers and long-term outcome. METHODS: We conducted a retrospective cohort study including individuals with confirmed CPA at the Royal Brompton Hospital u
