Anthelmintic efficacy against gastrointestinal nematodes in goats raised under mountain farming conditions in northern Italy.

BACKGROUND: This study aimed to evaluate the efficacy of anthelmintics in goats raised under mountain farming conditions in northern Italy. On 8 goat farms (n = 143 animals), a faecal egg count reduction (FECR) test was done after farmers conducted their routine anthelmintic treatments. Furthermore, on 5 goat farms (n = 135 animals) a FECR test was done under controlled conditions applying oral formulations of a macrocyclic lactone (ML), benzimidazole (BZ) (partly in combination with salicylanilide (SA)) or a combination of imidazothiazole (IT) and SA on the same farm. AR was assumed if FECR a

Andrographolide prevented toluene diisocyanate-induced occupational asthma and aberrant airway E-cadherin distribution via p38 MAPK-dependent Nrf2 induction.

Toluene diisocyanate (TDI) is a major cause of chemical-induced occupational asthma, which contributes about 15% of global asthma burden. Resistance and compounded side effects associated with the use of corticosteroid in asthma necessitate the search for alternative drugs. Andrographolide (AGP), a naturally occurring diterpene lactone is known to exhibit various bioactivities. Its ability to ameliorate cardinal features of allergic asthma was previously suggested in an eosinophilic asthma endotype. However, its potential antiasthma activity and mechanism of action in a neutrophilic occupation

Proteomic characterization of Withaferin A-targeted protein networks for the treatment of monoclonal myeloma gammopathies.

UNLABELLED: Withaferin A (WA), a natural steroid lactone from the plant Withania somnifera, is often studied because of its antitumor properties. Although many in vitro and in vivo studies have been performed, the identification of Withaferin A protein targets and its mechanism of antitumor action remain incomplete. We used quantitative chemoproteomics and differential protein expression analysis to characterize the WA antitumor effects on a multiple myeloma cell model. Identified relevant targets were further validated by Ingenuity Pathway Analysis and Western blot and indicate that WA target

Impact of previous infestation on dynamics of circulating hypodermin C in cattle artificially infested with Hypoderma lineatum (Diptera: Oestridae).

Four groups of cattle were artificially infested with 50 first instar Hypoderma lineatum after either a primary natural infestation or in the absence of a primary infestation. In two groups the primary infestation had been terminated by the application of either an organophosphate insecticide or a macrocyclic lactone parasiticide. Circulating hypodermin C and specific antibodies were measured for 40 weeks after the artificial infestation. Stage specific mortality of the larvae was also monitored. Previously uninfested cattle exhibited typical antibody and antigen profiles during the infestatio

Covalently targeting HSP90 with a natural small-molecule costunolide to inhibit necroptosis and ulcerative colitis.

BACKGROUND: Necroptosis exacerbates ulcerative colitis (UC) by disrupting intestinal barriers and promoting necroinflammation. Heat shock protein 90 (HSP90) regulates receptor-interacting protein kinase 1/3 (RIPK1/3) activity through its chaperone function, thereby participating in necroptosis and emerging as a potential therapeutic target for its inhibition in UC therapy. Costunolide (CTL), a principal sesquiterpene lactone from Aucklandia lappa, has poorly defined mechanisms in UC treatment. This study aimed to elucidate CTL's role in necroptosis inhibition and UC therapy by targeting HSP90.

Atractylenolide I alleviates LPS-induced pulmonary cell injury relevant to pediatric pneumonia via the ERK/USP9X/Mcl-1 and Akt/Survivin axes.

Pediatric pneumonia remains a leading cause of childhood mortality worldwide, and lipopolysaccharide (LPS)-induced pulmonary cell injury plays a critical role in its pathogenesis. Atractylenolide I (ATL-1), a sesquiterpene lactone derived from Atractylodes macrocephala Koidz., possesses well-documented anti-inflammatory properties. However, its protective effects against LPS-induced lung cell injury remain unclear. In this study, WI-38 human embryonic lung fibroblasts and BEAS-2B human bronchial epithelial cells were stimulated with LPS to establish complementary in vitro models of pulmonary c

Identification of anthecotuloide reveals a chemical mechanism underlying Anthemis cotula L. invasiveness.

BACKGROUND AND AIMS: Allelopathy underpins the competitive success of many invasive plants, yet direct chemical ecological links remain scarce. Here, we combined untargeted metabolomics with bioassays to analyse the allelopathic role of the invasive Anthemis cotula, and examine how its signature allelochemical drives invasion dynamics through differential phytotoxic effects on co-occurring plant species. METHODS: LC-QTOF metabolite profiling of leaf litter, roots, root exudates, and soils from invaded and uninvaded sites was performed to identify signature metabolites. The sesquiterpene lacto

Artesunate potentiates poly (ADP-ribose) polymerase inhibitor efficacy in ovarian cancer through RAD51-mediated homologous recombination repair impairment.

BACKGROUND: The homologous recombination repair (HRR) RAD51 recombinase is essential for ensuring accurate DNA repair, and its expression levels are inversely correlated with Poly (ADP-ribose) Polymerase inhibitor (PARPi) sensitivity. Artesunate (ART), a semisynthetic water-soluble sesquiterpene lactone derived from artemisinin, a natural product of Artemisia annua, was previously identified from an HRR reporting system as a potential HRR modulator, suggesting its utility as a PARPi sensitizer. METHODS: The clinical relevance of RAD51 was assessed using the TCGA database. In vitro studies emp

Lactobacillus Eats Amyloid Plaque and Post-Biotically Attenuates Senescence Due to Repeat Expansion Disorder and Alzheimer's Disease.

Patients with Alzheimer's disease and related dementia (ADRD) are faced with a formidable challenge of focal amyloid deposits and cerebral amyloid angiopathy (CAA). The treatment of amyloid deposits in ADRD by targeting only oxidative stress, inflammation and hyperlipidemia has not yielded significant positive clinical outcomes. The chronic high-fat diet (HFD), or gut dysbiosis, is one of the major contributors of ADRD in part by disrupted transport, epigenetic DNMT1 and the folate 1-carbon metabolism (FOCM) cycle, i.e., rhythmic methylation/de-methylation on DNA, an active part of epigenetic

RhlR-mediated cooperation in cystic fibrosis-adapted isolates of Pseudomonas aeruginosa.

Pseudomonas aeruginosa uses quorum sensing (QS) to regulate the expression of dozens of genes, many of which encode shared products, called "public goods." P. aeruginosa possesses two complete acyl-homoserine lactone (AHL) QS circuits: the LasR-I and RhlR-I systems. Canonically, these systems are hierarchically organized: RhlR-I activity depends on LasR-I activation. However, in contrast to laboratory strains, isolates from people with cystic fibrosis can engage in AHL QS using only the transcription factor RhlR. In these isolates, RhlR regulates AHL QS and the production of secreted public go

Natural Products That Changed Society.

Until the end of the 19th century all drugs were natural products or minerals. During the 19th century chemists succeeded in isolating pure natural products such as quinine, morphine, codeine and other compounds with beneficial effects. Pure compounds enabled accurate dosing to achieve serum levels within the pharmacological window and reproducible clinical effects. During the 20th and the 21st century synthetic compounds became the major source of drugs. In spite of the impressive results achieved within the art of synthetic chemistry, natural products or modified natural products still const
