Online extraction of 5-hydroxyindole acetic acid from urine for analysis by liquid chromatography-tandem mass spectrometry.

BACKGROUND: Urinary 5-hydroxyindole acetic acid (5-HIAA) is a useful marker for the turnover of tryptophan metabolites in the diagnosis and monitoring of carcinoid tumours and the carcinoid syndrome. We have developed a simple and cost-effective assay for urinary 5-HIAA using liquid chromatography-tandem mass spectrometry (LC-MS/MS) incorporating an online sample clean-up process to replace a liquid chromatography electrochemical (LC-EC) technique. METHODS: Acidified urine was serially diluted in ammonium acetate buffer followed by ammonium acetate buffer enriched with 5-hydroxyindole-3-aceti…

Healing process of acetic acid-induced gastric ulcer and gastric mucosal prostaglandin E generation level in rats.

The prostaglandin E (PGE) generation level (PGE level) in the gastric mucosa was investigated in relation to the healing and relapse of acetic acid-induced gastric ulcers in the rat. The PGE level around ulcers showed higher levels after ulcer induction and decreased during the ulcer diminishing period. Thereafter, the PGE levels showed an inclination to increase during the ulcer exacerbation period. In reulcerated rats, PGE levels were significantly higher. In conclusion, a high level of PGE may indicate an ulcer exacerbation state.

The acid hydrolysis mechanism of acetals catalyzed by a supramolecular assembly in basic solution.

A self-assembled supramolecular host catalyzes the hydrolysis of acetals in basic aqueous solution. The mechanism of hydrolysis is consistent with the Michaelis-Menten kinetic model. Further investigation of the rate-limiting step of the reaction revealed a negative entropy of activation (DeltaS(double dagger) = -9 cal mol(-1) K(-1)) and an inverse solvent isotope effect (k(H(2)O)/k(D(2)O) = 0.62). These data suggest that the mechanism of hydrolysis that takes place inside the assembly proceeds through an A-2 mechanism, in contrast to the A-1 mechanism operating in the uncatalyzed reaction. Co

Tuning FLO1 Expression via Promoter Engineering Modulates Flocculation Degree and Acetic Acid Stress Tolerance in Saccharomyces cerevisiae.

Robust yeast tolerance to inhibitors is essential for lignocellulosic biorefinery. Although cell flocculation is known to enhance acetic acid stress tolerance, the impact of its intensity remains unclear. In this study, engineered S. cerevisiae strains with distinct floc sizes were constructed through promoter engineering. The native FLO1 promoter in the non-flocculating laboratory strain BY4741 was replaced with either the constitutive strong promoter PGK1p or the ethanol-inducible promoter TPS1p using CRISPR-Cas9-mediated genome editing, resulting in strongly and moderately flocculating stra…

[Medium optimization and fermentation process regulation strategies for enhancing CO2 conversion to acetic acid by Acetobacterium woodii].

To meet the demand for CO2 resource utilization under the "dual-carbon" goals, the application of medium optimization combined with fermentation process regulation to enhance the acetate production and gas utilization capacity of Acetobacterium woodii represents an effective process optimization strategy. In this study, a multi-strategy approach combining medium optimization and fermentation process regulation was adopted to enhance the acetate yield of an A. woodii SL70 strain obtained via adaptive laboratory evolution (ALE). Firstly, through single-factor experiments and response surface met

Interplay of nitric oxide and indole-3-acetic acid signaling in regulating arsenate toxicity in tomato seedlings.

The problem of arsenic contamination is widespread, posing serious risks to both human health and crop productivity. Therefore, eco-friendly strategies are urgently needed to reduce metal stress-related yield losses and minimize its impacts on human health. Although the roles of nitric oxide (NO) and indole-3-acetic acid (IAA) in combating abiotic stresses, including metal toxicity, have been reported, the mechanisms underlying their interaction remain unclear. This study investigates whether IAA and NO function through shared or independent signaling pathways in regulating arsenate [As(V)] to…
