OBJECTIVE: To evaluate the effect of albendazole combined with surgery on the incidence of recurrent hydatid disease.
DESIGN: Prospective open study.
SETTING: Teaching hospital, Turkey.
SUBJECTS: 25 patients who presented with hepatic hydatid disease between December 1992 and October 1995.
INTERVENTIONS: Albendazole 10 mg/kg/day for a month before ultrasonography and then excision, and then albendazole for a further two months.
RESULTS: 2 patients were excluded because the albendazole caused disturbance of liver enzyme activities. Of the remaining 23 patients only 7 had viable cysts on ul…

The authors report a case of neurocysticercosis treated with Albendazole (methyl-5-propyl-thio-2-benzimidazilcarbamate). Computerized tomography (CT) and magnetic resonance imaging (MRI) of the brain presented various small bilateral parenchymal calcifications in the white matter and two inflammatory granulomatous formations localized respectively in the left and right posterior parietal lobe. After the serological diagnosis (Elisa-test) of cysticercosis, the patient was treated with albendazole (Zentel) with oral doses of 15 mg/kg/die for 20 days, and successively with a lower dose for anothe…

Pathological changes in alveolar hydatids from experimentally infected gerbils were studied after administration of albendazole or praziquantel. The animals were divided into several groups including the control one, and drugs of different dosages were given separately to different groups with diverse durations of infection. According to the microscopical observation, the hydatids could be categorized into 3 grades: grade I, degenerated cysts; grade II, stable cysts (showing no proliferation); grade III, proliferated cysts. Larger proportion of grade I cysts were found in the group treated wit…

A single dose of albendazole (candy) 100 mg and 100 mg qd for two days was given respectively to 135 and 321 children who were infected with Enterobius vermicularis. All cases were cured 3wk later according to perianal tape examination. The same drug of 150 mg qd and 200 mg qd for two days was administered respectively to residents with single or multi-infections of Ascaris, hookworm and Trichuris; the egg negative conversion rates of the two dosages were revealed to be 99.4% (466/469) and 99.8% (487/488) for Ascaris infection, 96.8% (91/94) and 94.3% (99/105) for hookworm infection, and 53.4%…

Two hundred and forty-five children infected with Trichuris trichiura, 143 of whom were also infected with Ascaris lumbricoides, were treated with albendazole as follows: single doses of either 600 mg or 800 mg, or daily doses of 400 mg for either 3 d or 5 d. Three stools were examined over a period of 10 d after treatment and again after nearly 40 d, using a quantitative microscopical technique. Albendazole appeared to act slowly against A. lumbricoides but within 10 d all dosages had cured about 92% of infections. In contrast, albendazole appeared initially to inhibit egg production by T. tr…

We treated 11 patients with a persistent (> 3 months), solitary small cysticercus granuloma and seizures with albendazole in a daily dose of 15 mg/kg body weight for 14 days. At repeat CT examinations performed at intervals of 2.5 to 6 weeks after initiation of therapy, the lesions had resolved in two patients and become significantly smaller (less than 50% of their pretreatment size) in two. Albendazole therapy in responders hastened the resolution of the lesions and confirmed the diagnosis.

A double-blind clinical trial was conducted in Monagas State, Venezuela to assess the tolerance and efficacy of albendazole in the therapy of Onchocerca volvulus infection. Forty-nine patients (26 treated and 23 controls) received a 10-day course of albendazole (400 mg/day) or a placebo. Consistent with the excellent tolerance observed, albendazole did not kill microfilariae. However, analysis of changes in microfilarial densities (mf/mg of skin) over one year showed that albendazole was active against O. volvulus, presumably by interfering with embryogenesis. The nature, degree, and duration …

Albendazole (10 mg/kg of body weight) was administered as a drench suspension or as a feed additive to 24 cattle with naturally acquired infections of Fasciola hepatica and Fascioloides magna. Cattle were euthanatized 16 to 30 days after treatment, and the number of viable flukes was counted. Viable F hepatica and F magna were decreased by 91.4% and 70.6% for drench administration and by 82.9% and 71.9% for the feed additive treatment, respectively. There was no significant difference between the efficacy of the 2 formulations in decreasing viable fluke numbers, compared with untreated control…

Human gnathostomiasis is characterized by space-occupying inflammatory lesions and/or hemorrhage as a result of the migration of, very often, a single larva of Gnathostoma spinigerum. Intermittent cutaneous migratory swellings occurring over years is the most common manifestation and the rare cerebral invasion may be fatal. There are currently no effective anthelminthics for this infection. During a double-blind randomized placebo control trial evaluating the efficacy of albendazole in cutaneous gnathostomiasis at a dosage of 400 mg twice daily for two weeks, it was observed that gnathostome l…

The efficacy of intraruminal albendazole (ABZ) capsules (Profitril-Captec) and the effect of treatment on productivity were studied in 300 ewes infected with gastrointestinal nematodes and the trematode Dicrocoelium dendriticum. Coprological tests revealed that treated animals remained negative for 10 weeks after the administration of capsules. Contamination of pasture with nematode larvae was significantly reduced during the whole experiment. Necropsy of 14 animals (seven treated and seven untreated) showed 96.9-99.2% efficacy against the nematodes Nematodirus spp., Oesophagostomum spp., Coop…

In a dosage-confirmation trial, anthelmintic activities of clorsulon and albendazole against Fasciola hepatica were evaluated and compared. Twenty-eight cattle (8 to 12 months old) with natural F hepatica infections were randomly allotted to 4 groups of 7 cattle each: group 1, no treatment (controls); group 2, clorsulon suspension given orally at 3.5 mg/kg of body weight; group 3, clorsulon suspension given orally at 7 mg/kg; and group 4, albendazole paste given orally at 10 mg/kg. At necropsies, performed 7 and 8 days after treatment, control cattle harbored a geometric mean of 133.2 F hepati…

Improved treatments for lymphatic filariasis (LF) could accelerate the global elimination program for this disease. A triple drug combination of the anti-filarial drugs ivermectin, diethylcarbamazine (DEC) and albendazole (IDA) has been shown to be safe and effective for achieving sustained clearance of microfilariae (Mf) of the filarial parasite Wuchereria bancrofti from human blood. However, the triple drug combination has not been previously been evaluated for treatment of brugian filariasis, which accounts for about 10% of the global LF burden. This hospital-based clinical trial compared t…

A simple, reproducible and fast (4 min chromatogram) method of liquid chromatography in tandem with mass spectrometry (LC/MS-MS) was developed to determine simultaneously the plasma levels of albendazole (ABZ) and its metabolite albendazole sulfoxide (ABZOX) for pharmacokinetic and clinical analysis. Each plasma sample was extracted by solid phase extraction (SPE) using phenacetin as internal standard (IS). The extracted sample was eluted with a Zorbax XDB-CN column using an isocratic method. The mobile phase consisting of water with 1% acetic acid (40%, A) and MeOH (60%, B), was used at a flo…

Benzimidazole (BZ) anthelmintics are among the most important treatments for parasitic nematode infections in the developing world. Widespre … of ben-1 conferred the maximum level of resistance following exposure to a single concentration of albendazole, and the loss of a second beta-tubulin gene did not alter the level of resistance. However, additional traits other than larval development could be affected by the loss of additional beta-tubulins, and the roles of other beta-tubulin genes might be …

Benzimidazole-2-carbamates (BZ, e.g., albendazole; ALB), which bind β-tubulin to disrupt microtubule polymerization, are one of two primary compound classes used to treat giardiasis. In most parasitic nematodes and fungi, BZ-resistance is caused by β-tubulin mutations and its molecular mode of action (MOA) is well studied. In contrast, in Giardia duodenalis BZ MOA or resistance is less well understood, may involve target-specific and broader impacts including cellular damage and oxidative stress, and its underlying cause is not clearly determined. Previously, we identified acquisition of a sin…

Although diethylcarbamazine (DEC) and albendazole are recommended to interrupt transmission of Wuchereria bancrofti, little is known about the macrofilaricidal effect of this drug combination. Forty-seven men with W. bancrofti infection were randomly assigned to receive a single dose of either DEC alone (6 mg/kg) (n=25) or a combination of DEC (6 mg/kg) and albendazole (400 mg) (n=22). Physical examinations for scrotal nodules (resulting from worm death) and ultrasound examinations (to detect living adult worms) were performed before treatment and 7, 14, 30, 45, 60, 90, 180, 270 and 360 days a…
