Coral reefs are essential for the foundation of marine ecosystems. However, ocean acidification (OA), driven by rising atmospheric carbon dioxide (CO2) threatens coral growth and biological homeostasis. This study examines two Hawaiian coral species-Montipora capitata and Pocillopora acuta to elevated pCO2 simulating OA. Utilizing pH and O2 microsensors under controlled light and dark conditions, this work characterized interspecific concentration boundary layer (CBL) traits and quantified material fluxes under ambient and elevated pCO2. The results of this study revealed that under increased

The 1983-1984 mortality event of the long-spined sea urchin Diadema antillarum reduced their population by up to 99% and was accompanied by a phase shift from coral dominated to algal dominated reefs in the Caribbean. Modest rebounds of D. antillarum populations in the Caribbean have been noted, and here we document the impacts of two major hurricanes (2017, Irma and Maria) and the 2022 disease outbreak on populations of D. antillarum found by targeted surveys in the urchin zone at nine fringing reef and three mangrove sites on St. John, USVI. D. antillarum populations at the reef sites had de

A cocktail of land-based sources of pollution threatens coral reef ecosystems, and addressing these has become a key management and policy challenge in the State of Hawai'i, other US territories, and globally. In West Maui, Hawai'i, nearly one quarter of all living corals were lost between 1995 and 2008. Onsite disposal systems (OSDS) for sewage leak contaminants into drinking water sources and nearshore waters. In recognition of this risk, the Hawai'i State Department of Health (DOH) is prioritizing areas for cesspool upgrades. Independently, we applied a decision analysis process to identify

Palythoa caribaeorum is a very common colonial zoanthid in the coastal reefs of Brazil. It is known for its massive production of mucus, which is traditionally used in folk medicine by fishermen in northeastern Brazil. This study identified biologically active compounds in P. caribaerum mucus. Crude mucus was collected during low tides by the manual scraping of colonies; samples were maintained in an ice bath, homogenized, and centrifuged at 16,000 g for 1 h at 4 °C; the supernatant (mucus) was kept at -80 °C until use. The enzymatic (proteolytic and phospholipase A2), inhibitory (metallo, cys

Coral reefs are ecologically significant habitats. Coral-algal symbiosis confers ecological success on coral reefs and coral-microbial symbiosis is also vital to coral reefs. However, current understanding of coral-microbial symbiosis on a genomic scale is largely unknown. Here we report a potential microbial symbiont in corals revealed by metagenomics-based genomic study. Microbial cells in coral were enriched for metagenomic analysis and a high-quality draft genome of "Candidatus Prosthecochloris korallensis" was recovered by metagenome assembly and genome binning. Phylogenetic analysis show

Palytoxin (PLTX), a polyether marine toxin originally isolated from the zoanthid Palythoa toxica, is one of the most toxic non-protein substances known. Fatal poisonings have been linked to ingestion of PLTX-contaminated seafood, and effects in humans have been associated with dermal and inhalational exposure to PLTX containing organisms and waters. Additionally, PLTX co-occurrence with other well-characterized seafood toxins (e.g., ciguatoxins, saxitoxins, tetrodotoxin) has hindered direct associations of PLTX to seafood-borne illnesses. There are currently no validated methods for the quanti

Crystalline, porous biomaterials, such as marine invertebrate skeletons, have been widely used for functional reconstruction of human tissues like bone and dental implants. Since in such an abrasive microenvironment adequate cell-material interactions are crucial for a successful treatment, it is of great importance to improve the means to examine these interactions. We developed a method that reveals the ultrastructure of the interface between coral skeletons and cultured neural cells to a higher quality than do traditional methods as it does not include damaging procedures like decalcificati

Coral disease is playing a significant role in structuring today's coral reef communities. While monitoring programs document declines associated with coral disease, there is a lack of tools that can test hypotheses of disease incidence and control. Here, we describe a modeling tool developed to test hypotheses about the spread and impact of white plague disease in diverse coral populations distributed across heterogeneous reef landscapes. The model Simulation of Infected Corals (SICO) was based on the dynamics of white plague over the course of 6 yr of monitoring on the fore-reefs of Little C

With large-scale impacts on coral reefs due to global climatic change projected to increase dramatically, and suitability of many areas for reef growth projected to decrease, the question arises whether particular settings might serve as refugia that can maintain higher coral populations than surrounding areas. We examine this hypothesis on a small, local scale in Honduras, western Caribbean. Dense coral thickets containing high numbers of the endangered coral Acropora cervicornis occur on offshore banks while being rare on the fringing reef on nearby Roatán. Geomorphological setting and commu

Fire corals (Millepora spp.) cause severe pain and inflammatory effects in humans upon contact, and the organs responsible for these effects are called nematocysts. Here, we isolated an active cytotoxin of ca. 18 kDa (MCTx-1) from nematocysts of Millepora dichotoma var. tenera. MCTx-1 was potently cytotoxic (EC50 value 79 ng/mL) towards L1210 mouse leukemia cells, hemagglutinated a 0.8% suspension of sheep erythrocytes (0.2 microg protein/mL) and was lethal in crayfish (LD50, 106 microg/kg). We deduced the primary structure of MCTx-1 from the corresponding cDNA sequence and found that MCTx-1 i

The discharge of nematocytes, the stinging cells of Coelenterata, is a poorly understood phenomenon. In particular, little is known about the chemical stimuli that trigger the discharge. In this paper, we show that thiols are able to initiate the nematocyst discharge in isolated nematocytes. Among the thiols tested, reduced glutathione and cysteine were found to be the most effective. The effect of glutathione was likely two-fold: it formed mixed disulfides with membrane thiols, as shown by the ability of the mercapto-blocking reagent iodoacetamide to abolish its action; and it bound to the me

Stony corals (Scleractinia) harbor abundant toxins as part of their survival strategy, and these molecules also provide novel lead candidates for drug development. In this study, we present the first comprehensive investigation of toxin profiles of the understudied Goniopora columna through transcriptome analysis, which has expanded the biochemical diversity of marine organism derived toxins. Then, a novel Kunitz-like polypeptide, named GcKuz1, which exhibits therapeutic potential for thrombosis was selected for functional studies and mechanistic investigation. In vitro experiments revealed th

The Maldivian Archipelago is home to valuable coral reefs that have been extensively studied for their ecological diversity. However, the diversity of the microbiome in Maldivian corals remains largely unexplored. In this study, the microbiota compositions (including both algal endosymbionts and bacteria) were investigated for the first time across various coral morphotypes sampled in May 2022 from four Maldivian atolls (Ari, North Malé, South Malé, and Rasdhoo). Coral and gorgonian specimens were collected via scuba diving at reef sites located on both ocean-exposed reefs and lagoon sites, ac

Ocean acidification (OA) is a major threat to the sexual recruitment of reef-building corals. Acclimation mechanisms are critical but poorly understood in reef-building corals to OA during early life stages. Here, Acropora gemmifera, a common Indo-Pacific coral cultured in in situ seawater from Luhuitou reef at three levels of pCO2 (pH 8.14, 7.83, 7.54), showed significantly delayed larval metamorphosis and juvenile growth, but adapted to long-term high pCO2. Differentially expressed genes (DEGs) emerged as a time- and dose-dependent mode of short-term response (3 days post settlement, d p.s.)

Genus Gobiodon are obligate coral dwelling cryptic fishes. Gobiodon form a mutualistic relationship with host scleractinian corals, making them integral to coral reef ecosystem functioning. Despite this, they are often omitted from fish assemblage studies, biodiversity surveys and coral reef fish guides due to confusion on species identification. The guide aims to provide a comprehensive description of the main diagnostic features for the majority of Gobiodon species distributed throughout the Indo-Pacific region. Species of the genus can be observed occupying host corals of the genera Acropor

Vibrio coralliilyticus is a pathogen of coral and shellfish, leading to devastating economic and ecological consequences worldwide. Although rising ocean temperatures correlate with increased V. coralliilyticus pathogenicity, the specific molecular mechanisms and determinants contributing to virulence remain poorly understood. Here, we systematically analyzed the type VI secretion system (T6SS), a contact-dependent toxin delivery apparatus, in V. coralliilyticus. We identified 2 omnipresent T6SSs that are activated at temperatures in which V. coralliilyticus becomes virulent; T6SS1 is an antib

Octocoral of the genus Clavularia is a kind of marine invertebrate possessing abundant cytotoxic secondary metabolites, such as prostanoids and dolabellanes. In our continuous natural product study of C. spp., two previously undescribed prostanoids [clavulone I-15-one (1) and 12-O-deacetylclavulone I (2)] and eleven known analogs (3-13) were identified. The structures of these new compounds were elucidated based on analysis of their 1D and 2D NMR, HRESIMS, and IR data. Additionally, all tested prostanoids (1 and 3-13) showed potent cytotoxic activities against the human oral cancer cell line (

Metabolites exuded by primary producers comprise a significant fraction of marine dissolved organic matter, a poorly characterized, heterogenous mixture that dictates microbial metabolism and biogeochemical cycling. We present a foundational untargeted molecular analysis of exudates released by coral reef primary producers using liquid chromatography-tandem mass spectrometry to examine compounds produced by two coral species and three types of algae (macroalgae, turfing microalgae, and crustose coralline algae [CCA]) from Mo'orea, French Polynesia. The ubiquitous metalloenzymes carbonic anhydrases (CAs, EC 4.2.1.1) are responsible for the reversible hydration of CO₂ to bicarbonate (HCO₃-) and protons (H⁺). Bicarbonate may subsequently generate carbonate used in many functional activities by marine organisms. CAs play a crucial role in several physiological processes, e.g., respiration, inorganic carbon transport, intra and extra-cellular pH regulation, and bio-mineralization. Multiple transcript variants and protein isoforms exist in the organisms. Recently, 16 α-CA isoforms have been identified in the coral Stylophora pistillata. Here,
