Neutralizing (54K) and non-neutralizing (54K and 48K) monoclonal antibodies against structural and non-structural yellow fever virus proteins confer immunity in mice.

The capacity of monoclonal antibodies to protect mice passively against yellow fever (YF) virus infection was investigated. Both neutralizing (54K-specific) and non-neutralizing (54K- and 48K-specific) antibodies protected mice against challenge with the RMP substrain of YF virus. Average survival times of mice inoculated intracerebrally with a standard lethal dose of YF virus differed according to the strain used: thus mice inoculated with the most neurovirulent viruses, FNV and Asibi, survived for 6 X 50 and 7 X 65 days respectively, and those with RMP virus survived for 15 X 75 days. The ca

Hexon and fiber of adenovirus type 14 and 55 are major targets of neutralizing antibody but only fiber-specific antibody contributes to cross-neutralizing activity.

Re-emerging human adenoviruses type 14 (HAdV14) and 55 (HAdV55) represent two highly virulent adenoviruses. The neutralizing antibody (nAb) responses elicited by infection or immunization remain largely unknown. Herein, we generated hexon-chimeric HAdV14 viruses harboring each single or entire hexon hyper-variable-regions (HVR) from HAdV55, and determined the neutralizing epitopes of human and mouse nAbs. In human sera, hexon-targeting nAbs are type-specific and mainly recognize HVR2, 5, and 7. Fiber-targeting nAbs are only detectable in sera cross-neutralizing HAdV14 and HAdV55 and contribute

Broadly Neutralizing Antibodies Display Potential for Prevention of HIV-1 Infection of Mucosal Tissue Superior to That of Nonneutralizing Antibodies.

UNLABELLED: Definition of the key parameters mediating effective antibody blocking of HIV-1 acquisition within mucosal tissue may prove critical to effective vaccine development and the prophylactic use of monoclonal antibodies. Although direct antibody-mediated neutralization is highly effective against cell-free virus, antibodies targeting different sites of envelope vulnerability may display differential activity against mucosal infection. Nonneutralizing antibodies (nnAbs) may also impact mucosal transmission events through Fc-gamma receptor (FcγR)-mediated inhibition. In this study, a pan

A broad antibody with enhanced HIV-1 neutralization via bispecific antibody-mediated prepositioning.

Antibodies targeting the highly conserved prehairpin intermediate (PHI) of class I viral membrane-fusion proteins are generally weakly neutralizing and are not considered viable therapeutic agents. We previously demonstrated that antibodies targeting the gp41 N-heptad repeat (NHR), which is transiently exposed in the HIV-1 PHI, exhibit enhanced broad neutralization in cells expressing the Fc receptor, FcγRI. To enhance neutralization in cells lacking FcγRI, we here develop a bispecific antibody (bsAb) by fusing an NHR-targeting antibody to an antibody against CD4, the HIV-1 receptor on T cells

Dengue virus neutralizing antibody: a review of targets, cross-reactivity, and antibody-dependent enhancement.

Dengue is the most common viral infection spread by mosquitoes, prevalent in tropical countries. The acute dengue virus (DENV) infection is a benign and primarily febrile illness. However, secondary infection with alternative serotypes can worsen the condition, leading to severe and potentially fatal dengue. The antibody raised by the vaccine or the primary infections are frequently cross-reactive; however, weakly neutralizing, and during subsequent infection, they may increase the odds of antibody-dependent enhancement (ADE). Despite that, many neutralizing antibodies have been identified aga

Semiquantitative Paper-Based Microfluidic Surrogate Virus Neutralization Test for SARS-CoV-2 Neutralizing Antibodies.

Neutralizing antibodies (nAbs) produced from infection or vaccination play an important role in acquired immunity. Determining virus-specific nAb titers is a useful tool for measuring aquired immunity in an individual. The standard methods to do so rely on titrating serum samples against live virus and monitoring viral infection in cultured cells which requires high biosafety level containment. The surrogate virus neutralization test (sVNT) reduces the biohazards and it is suitable for designing rapid test device in a lateral flow assay (LFA) format. Here, we introduce the fabrication and deve

High-throughput analysis of anti-poliovirus neutralization antibody titre in human serum by the pseudovirus neutralization test.

To monitor vulnerability of countries to poliovirus (PV) outbreaks, serosurveillance of anti-PV neutralization antibody is conducted by conventional PV neutralization test (cPNT), which uses live PV strains. We previously developed a pseudovirus PV neutralization test (pPNT) as an alternative to cPNT, which uses PV pseudovirus that expresses luciferase as a reporter in the infection without producing infectious PV. In the present study, we established a high-throughput pPNT (HTpPNT) for a large-scale serosurveillance. The HTpPNT system was evaluated with 600 human serum samples obtained from a

Modeling neutralization kinetics of HIV by broadly neutralizing monoclonal antibodies in genital secretions coating the cervicovaginal mucosa.

Eliciting broadly neutralizing antibodies (bnAb) in cervicovaginal mucus (CVM) represents a promising "first line of defense" strategy to reduce vaginal HIV transmission. However, it remains unclear what levels of bnAb must be present in CVM to effectively reduce infection. We approached this complex question by modeling the dynamic tally of bnAb coverage on HIV. This analysis introduces a critical, timescale-dependent competition: to protect, bnAb must accumulate at sufficient stoichiometry to neutralize HIV faster than virions penetrate CVM and reach target cells. We developed a model that i

Broadly neutralizing monoclonal antibodies derived from mRNA LNP immunization exhibit potent neutralizing ability against JN.1, KP.3.1.1 and XEC new Omicron variants.

Since the emergence of severe acute respiratory syndrome coronavirus 2 Omicron, dramatic changes in the receptor-binding domain have allowed for virus escape from many therapeutic antibodies. Development of antibodies effective against current viral strains is, therefore, necessary to provide useful clinical treatments and can also yield fundamental information about viral neutralization. Here, we utilized single B-cell antibody technology to isolate and characterize neutralizing antibodies from splenocytes of mice immunized with mRNA-LNPs. With this approach, we identified five BA.5 and four

Recombinant L1R protein induces cross-neutralizing antibodies against capripoxviruses despite a subdominant humoral response.

Capripoxviruses, including the goatpox virus (GTPV), sheeppox virus (SPPV), and lumpy skin disease virus (LSDV), cause substantial economic losses in domestic ruminants in Asia, Africa, the Middle East, and Europe. The identification of viral antigens that elicit protective immunity, along with the characterization of antigen-specific humoral responses, is crucial for the rational development of vaccines and serodiagnostic assays. L1R is an essential membrane protein of the intracellular mature virion (IMV) and a known neutralizing antibody target for poxviruses. In this study, a truncated L1R

Dynamics of neutralizing antibodies against enterovirus D68 in adults during the COVID-19 pandemic.

BACKGROUND: Enterovirus D68 (EV-D68) causes acute respiratory illnesses and has also implicated in acute flaccid myelitis. Neutralizing antibodies (NtAbs) represent key components of the adaptative immunity. However, the dynamics of EV-D68 NtAbs in adults remain poorly characterized. We evaluated EV-D68 NtAb levels in retail workers in Canada during the COVID-19 pandemic. METHODS: NtAbs were measured in paired serum samples (N = 194), from retail workers (≥18 years) enrolled in a prospective SARS-CoV-2 cohort at the Centre Hospitalier Universitaire de Québec-Université Laval, in Quebec City.

Delayed viral rebound post-ART interruption in infant macaques given SIV-specific neutralizing antibodies.

Interventions to prevent HIV rebound in the absence of antiretroviral therapy (ART) are needed. Here, we use a rhesus macaque model of pediatric HIV to assess the impact of simian immunodeficiency virus (SIV) Env-specific rhesus monoclonal antibodies (RhmAbs) on viral rebound. We find that RhmAb-treated infants experience delayed rebound compared to controls (median of 64 versus 7.5 days, p = 0.0007). Time to rebound very strongly correlates with both ITS102.01-LS concentrations (r = 0.9, p = 0.002) and anti-drug antibodies against ITS103.01-LS (r = -0.95, p = 0.008). SIVgag-specific CD8+ T ce

A facile method for purifying viral immunogens using magnetic beads to generate neutralizing antibodies.

Human parainfluenza virus type 3 (HPIV-3), an increasingly significant respiratory pathogen affecting both pediatric and elderly populations, currently lacks effective prophylactic or therapeutic interventions. The development of antibody-based therapeutics with the potential to prevent or treat HPIV-3 infection remains an active area of investigation. However, a major limitation of such antibody therapies is the emergence of escape mutant strains. Combination therapy employing multiple neutralizing antibodies targeting distinct epitopes has been explored as a strategy to circumvent viral esca

Development of a Norway rat hepacivirus reporter for high-throughput quantification of neutralizing antibodies.

Hepatitis C virus (HCV) remains a major global health challenge, with vaccine research hindered by a lack of robust immunocompetent animal models. Infection with the HCV-related Norway rat hepacivirus 1 (NrHV) in rats or mice shares HCV-defining characteristics, including chronicity, delayed immune responses, and liver disease, thereby providing immunocompetent small animal models for immune responses and pathology. Infectious NrHV cell culture systems allow studies of host factors and neutralizing antibodies; however, quantification relies on laborious manual counting of infected cells. To pe

Maturation of HIV-1 neutralizing antibodies in a germinal center conditional expression mouse model.

In germinal centers, activated B cells modify their antigen receptors through somatic hypermutation (SHM), followed by antigenic selection that favors expansion of high affinity B cells. The affinity maturation process is critical for development of broadly neutralizing antibodies (bnAbs) against the human immunodeficiency virus-1 (HIV-1). BnAbs have been isolated from some people living with HIV-1. Because these antibodies target conserved epitopes of the HIV-1 Envelope (Env) protein, they inhibit a broad spectrum of viruses. Eliciting bnAbs by vaccination is a top priority for HIV-1 preventi

Affinity maturation endows potent activity onto class 6 SARS-CoV-2 broadly neutralizing antibodies.

The emergence of SARS-CoV-2 variants of concern (VOCs) has greatly diminished the neutralizing activity of previously FDA-approved monoclonal antibodies (mAbs), including that of antibody cocktails and of first-generation broadly neutralizing antibodies such as S309 (Sotrovimab). In contrast, antibodies targeting cryptic conformational epitopes of the receptor binding domain (RBD) have demonstrated broad activity against emerging variants, but exert only moderate neutralizing activity, which has so far hindered clinical development. Here, we utilize in vitro display technology to identify and

Ultrapotent SARS coronavirus-neutralizing single-domain antibodies that clamp the spike at its base.

Therapeutic monoclonal antibodies can prevent severe disease in SARS-CoV-2 exposed individuals. However, currently circulating virus variants have evolved to gain significant resistance to nearly all neutralizing human immune system-derived therapeutic monoclonal antibodies that had previously been emergency-authorized for use in the clinic. Here, we describe the discovery of a panel of single-domain antibodies (VHHs) directed against the spike protein S2 subunit that broadly neutralize SARS-CoV-1 and -2 with unusually high potency. One of these VHHs tightly clamps the spike's monomers at a hi
