Is leucocyte ascorbic acid an unreliable estimate of vitamin C deficiency?

Leucocyte ascorbic acid levels failed to identify six of seven elderly patients shown to be deficient on an oral vitamin C saturation test. Compared to those with a normal saturation test, patients judged deficient had lower levels for triceps skinfold thickness, mean arm muscle circumference and Quetelet's index; there was a significant association with the habit of eating alone, and with a dietary intake of less than 30 mg of ascorbic acid daily (the recommended daily allowance in the U.K.). No significant difference was found in the values for haemoglobin, serum albumin or potassium concent…

Rapid diagnostic tests for glucosuria are still influenced by ascorbic acid.

The influence of ascorbic acid on commercial rapid diagnostic tests for glucosuria is demonstrated in vitro. Although much work has been done to stabilize the tests against such interference, and although manufacturers of tests trips declare their products now to be fairly stable against ascorbic acid 'at higher glucose levels', all the investigated commercial rapid diagnostic tests for glucose at the lower pathological range of glucosuria were substantially influenced by ascorbic acid at potentially physiological concentrations.

Nicotine and ascorbic acid effects on cold-restraint ulcers in rats.

Rats were orally administered 1-ascorbic acid, nicotine 1-ascorbic acid and nicotine, or distilled water for 10 days. Following this treatment they were fasted for 24 h and then restrained in a cold environment for 2 h. Nicotine alone produced significantly more gastric ulcers than any other treatment. 1-Ascorbic acid increased ulceration relative to controls. The combined effects of 1-ascorbic acid and nicotine resulted in reduced ulcer incidence and severity. It appears that l-ascorbic acid and nicotine do not act synergistically to augment stress-induced gastric ulcer.

Plasma ascorbic acid levels in sheep experimentally infected with Fasciola hepatica.

Three groups of sheep, one control (A) and two infected with Fasciola hepatica metacercariae (B&C), were investigated. Each animal of group B was orally dosed with about 270 metacercariae and each sheep in group C received two such doses administered 14 days apart. In both infected groups, plasma ascorbic acid levels progressively decreased after exposure reaching a minimum by week 9, and then slightly increased during the subsequent five weeks. The decrease in ascorbic acid concentrations was mainly related to impaired synthesis as a result of the hepatic damage caused by the flukes.

Ascorbic acid deficiency in malignant diseases: a clinical and biochemical study.

In a study of the vitamin C status of 50 patients with malignant disease, 46 had leucocyte levels less than the lower limit of the normal range (18-50,μg/10(8) W.B.C.) and of these 30 had very low levels (< 12.5 μg/10(8) W.B.C.). Physical signs compatible with subclinical scurvy were frequently recorded and there was a significant decrease in capillary fragility in those with the lowest levels. Most patients had an inadequate dietary intake of ascorbic acid-containing foods and this was felt to be the major factor in producing the vitamin depletion.

Suppression of adrenocorticotrophic activity in the ascorbic acid deficient guinea-pig.

1. Adrenocortical hyperactivity caused by a marked increase in circulating corticotrophin occurred in guinea-pigs on a diet deficient in ascorbic acid.2. Betamethasone prevented the rise in the blood ACTH concentration in scorbutic animals and also the increased steroid production per gramme adrenal tissue in vitro. It diminished the adrenal hypertrophy and partially suppressed the rise in plasma cortisol.3. Ninety minutes after the injection of ascorbic acid corticotrophin could no longer be detected in the plasma of scorbutic animals.4. Neither the survival time nor the weight loss was affec…

Ascorbic acid deficiency and pituitary adrenocortical activity in the guinea-pig.

1. Guinea-pigs kept on a diet deficient in ascorbic acid lost weight and became moribund in about 24 days.2. The adrenal ascorbic acid concentration fell rapidly during the first 2 weeks, and the plasma corticosteroid concentration and 17-oxogenic steroid excretion rose sharply in the third week of ascorbic acid deficiency.3. Both histamine and corticotrophin increased the plasma corticosteroid concentration when injected during the second week but failed to change the pre-existing high concentration of the steroid in the third week of ascorbic acid deficiency.4. The observations confirm that …

Inhibition of ascorbic acid on Pseudomonas bubulae under super-chilled storage.

To explore the inhibitory effect of 1% ascorbic acid on spoilage bacteria under super-chilled storage, tilapia fillets either with natural microbiota or inoculated with Pseudomonas bubulae P1, a dominant spoiler derived from tilapia, were employed in this research. Results showed that ascorbic acid significantly inhibited bacterial growth, delayed sensory spoilage, and reduced the levels of total volatile basic nitrogen (TVB-N) in both types of fillets stored at -1.5 °C. Further analysis indicated that the reduction in TVB-N was attributed to the diminished ammonia production by P1. Transcript…

A copper-based ascorbic acid-activatable gel for postoperative glioblastoma therapy.

Glioblastoma (GBM) is prone to rapid postoperative recurrence, primarily due to its invasive growth pattern and immunosuppressive microenvironment. To combat these challenges, we developed an ascorbic acid (AA)-responsive implantable gel crosslinked by copper ions, denoted as CuTR-gel, for postoperative GBM therapy. CuTR-gel leveraged the copper-chelating properties of sodium alginate (SA) to co-deliver TPD-Cu (a novel ascorbic acid-activated copper complex prodrug) and R848 (a TLR7/8 agonist). Upon implantation, endogenous AA in GBM microenvironment triggered TPD-Cu cleavage to release strong…

Intra-articular injection of ascorbic acid enhances microfracture-mediated cartilage repair.

Previous studies have confirmed that ascorbic acid (AA) can promote cartilage repair and improve cartilage differentiation in bone marrow mesenchymal stem cells. However, the use of microfracture (MFX) combined with AA to repair cartilage damage has not been studied. This study established a rabbit animal model and treated cartilage injury with different concentrations of AA combined with MFX. Macroscopic observations, histological analysis, immunohistochemical analysis and reverse transcription quantitative polymerase chain reaction analysis of TGF-β, AKT/Nrf2, and VEGF mRNA expression were p…

In Situ Ratiometric Determination of Cerebral Ascorbic Acid after Ischemia Reperfusion.

Ascorbic acid (AA) is significant in protecting the brain from further damage and maintaining brain homeostasis after ischemia stroke (IS); however, the dynamic change of cerebral AA content after different degrees of ischemic stroke is still unclear. Herein, carboxylated single-walled carbon nanotube (CNT-COOH)- and polyethylenedioxythiophene (PEDOT)-modified carbon fiber microelectrodes (CFEs) were proposed to detect in situ cerebral AA with sensitivity, selectivity, and stability. Under differential pulse voltammetry scanning, the CFE/CNT-COOH/PEDOT gave a ratiometric, electrochemically res…

Ascorbic acid is associated with favourable hormonal profiles among infertile males.

INTRODUCTION: Infertility affects about 16% of North American couples, with the male factor contributing to ∼30% of cases. Reproductive hormones play an integral role in regulating the reproductive system and consequently, fertility. Oxidative stress reduces testosterone synthesis, and reduction in oxidative stress can improve hormone profiles. Ascorbic acid is a potent antioxidant that accounts for up to 65% of seminal antioxidant activity; however, its effects on reproductive hormones in humans are unknown. METHODS: The objective was to determine the association between serum ascorbic acid …
