Clinical significance of Aspergillus sensitisation in bronchial asthma.

Numerous studies have suggested a link between fungal sensitisation and severity of asthma. However, few studies have specifically evaluated the relationship between Aspergillus sensitisation and asthma severity. This study was aimed at investigating the clinical significance of Aspergillus sensitisation in asthma. In this prospective cross-sectional study, patients with asthma were subjected to pulmonary function test and an intradermal Aspergillus skin test (AST) apart from a detailed clinical history and physical examination. Assessment of asthma severity was carried according to the Global…

Impact of Aspergillus fumigatus in allergic airway diseases.

For decades, fungi have been recognized as associated with asthma and other reactive airway diseases. In contrast to type I-mediated allergies caused by pollen, fungi cause a large number of allergic diseases such as allergic bronchopulmonary mycoses, rhinitis, allergic sinusitis and hypersensitivity pneumonitis. Amongst the fungi, Aspergillus fumigatus is the most prevalent cause of severe pulmonary allergic disease, including allergic bronchopulmonary aspergillosis (ABPA), known to be associated with chronic lung injury and deterioration in pulmonary function in people with chronic asthma an…

A new species, Aspergillus persii, as an agent of onychomycosis.

Onychomycosis due to non-dermatophytic fungi is a well-known and increasingly common infection. For the first time ever, we report two cases of onychomycosis caused by Aspergillus persii, a recently described new Aspergillus species. After ten and three years respectively, both patients showed relapsing nail infections, and mycological tests were still positive for A. persii. In vitro antifungal susceptibility tests demonstrated that both strains were only susceptible to itraconazole, voriconazole, posaconazole and terbinafine of the agents tested.

Butenolide and furandione from an endophytic Aspergillus terreus.

A new butenolide, aspernolide D (1), and furandione, asperterone (2), together with four known butenolides, butyrolactones I-IV and aspernolide B, were obtained from cultures of the endophytic fungus Aspergillus terreus, isolated from the flowering plant Mammea siamensis. The structures of these compounds were elucidated by analysis of NMR spectroscopic and mass spectrometric data.

Pseudomembranous Aspergillus tracheobronchitis in a heart transplant recipient.

Aspergillus tracheobronchitis (AT) is an uncommon clinical presentation of pulmonary aspergillosis that frequently progresses to invasive pulmonary aspergillosis. Diagnosis of AT may be delayed because of its insidious onset, non-specific signs and symptoms, and scarcity of radiographic abnormalities. We report the case of a patient who received a heart transplant (HT) because of cardiac amyloidosis and who developed pseudomembranous AT. Possible risk factors concurrent in this case were splenectomy, lymphocytopenia, and previous cytomegalovirus infection. Chest computed tomography scan showed…

Contribution of peroxisomes to penicillin biosynthesis in Aspergillus nidulans.

Peroxisomal localization of the third enzyme of the penicillin biosynthesis pathway of Aspergillus nidulans, acyl-coenzyme A:IPN acyltransferase (IAT), is mediated by its atypical peroxisomal targeting signal 1 (PTS1). However, mislocalization of IAT by deletion of either its PTS1 or of genes encoding proteins involved in peroxisome formation or transport does not completely abolish penicillin biosynthesis. This is in contrast to the effects of IAT mislocalization in Penicillium chrysogenum.

Voriconazole for the treatment of refractory Aspergillus fumigatus keratitis.

We report a case of Aspergillus fumigatus keratitis in a 53-year-old, well-controlled diabetic female who did not respond to standard antifungal treatment. She was started on topical natamycin eye drops, but the infiltrate continued to progress. Topical amphotericin B and systemic ketoconazole was added, however, there was no response and the infiltrate increased further. She was then switched to topical and systemic voriconazole. Steady resolution of the infiltrate was noted within 2 weeks of therapy.

The characteristics of Aspergillus fumigatus mycetoma development: is this a biofilm?

Aspergillus fumigatus is an increasingly prevalent opportunistic fungal pathogen of various immuno-compromised individuals. It has the ability to filament within the lungs forming dense intertwined mycelial balls. These morphological characteristics resemble those of microbial biofilms, which are matrix enclosed microbial populations, adherent to each other and/or to surfaces or interfaces. The purpose of this paper is to review some recent experiments that indicate the potential biofilm forming capacity of A. fumigatus in vitro. Initially it was established that conidial seeding density is im…

Platelia Aspergillus assay for diagnosis of disseminated histoplasmosis.

The Platelia Aspergillus assay tested positive in 8 of 11 patients with disseminated histoplasmosis. While other available methods for diagnosis have several drawbacks, this cross-reactivity is particularly valuable in the perspective of practitioners outside the USA who cannot use the test detecting antigen of Histoplasma capsulatum.

Function analysis of steA homolog in Aspergillus oryzae.

The asexual ascomycete Aspergillus oryzae has an steA homolog in its genome. The steA homolog of A. oryzae was transcribed in the wild-type strain but steA disruption did not affect the phenotype of the disruptant strain. On the other hand, the steA-overexpressing strain showed the restriction of vegetative hyphal growth and conidiation, and the formation of balloon-shaped structures. The restriction of vegetative hyphal growth and conidiation were partially rescued by high osmolarity. In addition, the hyphal growth of the steA-overexpressing strain was inhibited by Calcofluor white, and the s…

Purification and characterization of xylanases from Aspergillus giganteus.

A strain of Aspergillus giganteus cultivated in a medium with xylan produced two xylanases (xylanase I and II) which were purified to homogeneity. Their molar mass, estimated by SDS-PAGE, were 21 and 24 kDa, respectively. Both enzymes are glycoproteins with 50 degrees C temperature optimum; optimum pH was 6.0-6.5 for xylanase I and 6.0 for xylanase II. At 50 degrees C xylanase I exhibited higher thermostability than xylanase II. Hg2+, Cu2+ and SDS were strong inhibitors, 1,4-dithiothreitol stimulated the reaction of both enzymes. Both xylanases are xylan-specific; kinetic parameters indicated …

Fatal Aspergillus fumigatus Myositis in an immunocompetent patient.

A 69-year old farmer developed Aspergillus myositis in the right psoas and paravertebral muscles extending to the retroperitoneum and the fifth lumbar vertebra. The infection appeared after two local instillations of steroid for back pain. Although the patient was not immunocompromised, surgical drainage and antifungal therapy failed to cure him; he died of a bacterial pulmonary superinfection while cultures of the abscess drainage fluid grew Aspergillus. The likely portal of entry in this patient was direct inoculation during infiltration of the steroid; the steroid probably caused a local im…

Aspergillus mastoiditis in a patient with acquired immunodeficiency syndrome.

We present the case history of a patient who was severely immunocompromised due to infection with the human immunodeficiency virus (HIV), and who subsequently developed acute mastoiditis due to Aspergillus fumigatus. Fungal otomastoiditis is a rarely reported complication of HIV infection. A high index of suspicion is required in these patients to facilitate early diagnosis and appropriate therapy.

[Localised aspergillosis of the lung caused by Aspergillus niger].

A 51-year old man with a history of pulmonary tuberculosis suffered an exacerbation without demonstration of tubercle bacilli. The mycological examination of sputum specimens discovered an infection with Aspergillus niger. Diagnosis was confirmed by the pure culture of this fungus in bronchial mucus and the cultural and histological identification in surgical material. As a histological feature strong double-refractile crystalline material was found in fungi and surrounding lung tissue (wall of cavern), probably oxalate crystals, developed from oxalic acid, produced by Aspergillus niger.
