Ultrastructure of the capsule of Bacillus megaterium ATCC 19213.

The ultrastructure of the firmly adherent capsule produced by Bacillus megaterium cultured on fructose mineral salts medium was examined using thin sectioning, freeze-etching, and critical point drying by transmission and scanning electron microscopy. The capsule material was shown to be fibrillar, with most fibrils containing bulbous protrusions. Two types of fibres were resolved. These were termed primary and cross-linking fibres. Primary fibres originated at the cell wall and had a diameter of 34-50 nm. They also contained bulbous protrusions and enlarged areas where branching occurred. Cro…

Recognition sites for chemotactic repellents of Bacillus subtilis.

Repellents of Bacillus subtilis include many membrane-active compounds, such as uncouplers of oxidative phosphorylation, local anesthetics, chlorpromazine (a central nervous system depressant), and tetraphenylboron (a lipophilic anion). Normally, bacteria swim smoothly, and occasionally tumble, but addition of repellent causes all bacteria to tumble, then later resume original frequency of swimming and tumbling (adaptation). Bacteria adapted to repellent can then be tested to determine the minimum concentration (threshold) of the same or different repellents that causes tumbling. The results i…

Biological characteristics of an enterotoxin produced by Bacillus cereus.

An enterotoxin synthesized during exponential growth by Bacillus cereus produces fluid accumulation in rabbit ileal loops, alters vascular permeability in the skin of rabbits, and kills mice when injected intravenously. All activities are eluted simultaneously from a Sephadex G-75 column and are distinct from the hemolysin and egg yolk turbidity factor of B. cereus. The enterotoxin is a true exotoxin. It interacts with intestinal receptor sites in a highly transient manner in the ileal loop system. Rabbit immune serum produced against the culture fluids from one strain of B. cereus neutralized…

Temperature response in animals infected with Bacillus anthracis.

Rats, rabbits, swine, guinea pigs, and monkeys were infected with anthrax spores, and their temperature responses were recorded. These were characteristic for a species and appeared independent of resistance or susceptibility of the species toward establishment of the disease. The rabbit appeared unique in that it not only failed to demonstrate a dose-response relationship over an 8-log dose range, but acted independently producing erratic body temperatures depending on spore dose. This limits the usefulness of the rabbit in studying anthrax pathogenesis, and poses questions regarding publishe

TRACE METAL REQUIREMENTS FOR SPORULATION OF BACILLUS MEGATERIUM.

Kolodziej, Bruno J. (Northwestern University, Evanston, Ill.), and Ralph A. Slepecky. Trace metal requirements for sporulation of Bacillus megaterium. J. Bacteriol. 88:821-830. 1964.-A sucrose-mineral salts medium was purified for the purpose of definitively re-examining the trace metal cations associated with the sporulation of Bacillus megaterium. Two heretofore unknown requirements for copper (0.013 mug/ml of cupric ion) and molybdenum (27.2 mug/ml of molybdate ion) were uncovered. In the purified copper-supplemented medium, sporulation levels of other metals were determined as follows: iro…
